Seasonal testicular changes in Dendropsophus minutus Peters, 1872 (Anura, Hylidae) Biocell

The reproductive cycle in anurans may be either continuous or discontinuous. These differences may be connected to seasonal climate changes and/or to anthropic activity. Forty adult male individuals of the Dendropsophus minutus species were collected during one year, in the municipality of Chapada dos Guimarães (Mato Grosso, Brazil). The testicles were studied under light and transmission electron microscopy. No variations were observed when the diameter of the seminiferous tubules and the thickness of the interstitial tissue were studied. However, changes in spermatogenesis were conspicuous and indicated that the reproductive cycle of D. minutus in Chapada dos Guimarães is discontinuous and seems related to variations in air temperature and rainfall.

Structure of the stigma and style in sunflower (Helianthus annuus L.) Biocell

This is the first report of the ultrastructure of the stigma and style during and after anthesis in Helianthus annuus L. using light and transmission electron microscopy. The stigma is bifid with unicellular papillae. There is no secretion of lipids, carbohydrates or proteins at anthesis. The style is semisolid in the upper portion, closer to the stigma, and becomes solid below. Ultrastructural changes on cells of the stigma and the style are described. The transmitting tissue of the ovule is first evident 40 minutes after pollination and persists during the first stages of embryogenesis. Only one pollen tube per micropyle was observed growing through this tissue.

Ultrastructural analysis and identification of membrane proteins in the free-living amoeba Difflugia corona Biocell

Syntaxin-1 and 25-kDa Synaptosome-associated Protein (SNAP-25) are present in the plasma membrane of several different secretory cell types and are involved in the exocytosis process. In this work, the free-living amoeba Difflugia corona was studied in relation to ultrastructure, structural membrane proteins, and proteins such as Syntaxin-1 and SNAP-25. Our results obtained by scanning electron microscopy in the amoeba without its theca, showed many membrane projections and several pore-like structures. Using immunocytochemistry, we found structural proteins Syntaxin-1 and SNAP-25.

Structural and ultrastructural characterization of zebu (Bosindicus) spermatozoa Biocell

The ultrastructure of normal, ejaculated spermatozoa of Bos indicus was studied by means of electron microscopy, being evaluated in two principal parts, the head and the tail. The head is flat, oval or paddle-shaped with a square base, which provides a concave recess for the insertion of the tail. The acrosome tightly covers the anterior two thirds of the nucleus. A distinct unilateral acrosomal bulge was observed along the apical edge of the head. The equatorial region demarcates the acrosome from the post-equatorial region that covers the caudal one third of the nucleus. Cytochemical localization of acid phosphatase in Stigeoclonium tenue (Chaetophorales, Chlorophyceae) Biocell

Nonspecific acid phosphatases are a group of enzymes whose activity increases the availability of exogenous and endogenous orthophosphate either through extra- or intracellular hydrolysis of phosphate compounds. Our study demonstrates the activity of acid phosphatases in the filamentous freshwater alga Stigeoclonium tenue. These enzymes were detected following a cerium-based method in which cerium was used as an orthophosphate-capture reagent. In thalli from S. tenue from the natural environment, acid phosphatases were found in the longitudinal cell wall, plasmalemma, and vacuole. An ultrastructural study of spermiogenesis in two species of Sitophilus: Coleoptera: Curculionidae Biocell

The spermiogenesis of Sitophilus zeamais and Sitophilus oryzae, the maize and the rice weevil, respectively, was studied by light microscopy and scanning and transmission electron microscopy. Sitophilus spp. is the most widespread and destructive primary pest of stored cereals in the world. The spermiogenesis occurs within cysts. There are approximately 256 germ line cells per cyst. Inside each cysts, all the spermatids are in the same stage of maturation. The ultrastructure of the spermatozoa of S. zeamais and S. oryzae is similar to that described for other beetles. Protein detection in spermatids and spermatozoa of the butterfly Euptoieta hegesia (Lepidoptera) Biocell

This study was undertaken to detect protein components in both sperm types of the butterfly Euptoieta hegesia. These spermatozoa possess complex extracellular structures for which the composition and functional significance are still unclear. In the apyrene sperm head, the proteic cap presented an external ring and an internal dense content; basic proteins were detected only in external portions. In the tail, the paracrystalline core of mitochondrial derivatives and the axoneme are rich in proteins. The extratesticular spermatozoa are covered by a proteic coat, which presented two distinct layers. The effects of Photodynamic Therapy using 2nd generation photosensitizers have been widely investigated aiming clinical application treatment of solid neoplasms. In this work, ultrastructure changes caused by the action of two 2nd generation photosensitizers and laser irradiation on CHO-K1 and HeLa (neoplastic) cells were analyzed by transmission electron microscopy. Aluminum phthalocyanine chloride, aluminum phthalocyanine tetrasulfonate chloride and radiation from a semiconductor laser at a fluency of 0.5 J/cm² (Power=26mW; l=670nm) were used. Structure of the kidney of Bufo arenarum: Intermediate segment, distal tubule and collecting tubule Biocell

The ultrastructure of the intermediate segment (IS), distal tubule and collecting tubule (CT) of the south american toad Bufo arenarum, was studied by light and transmission electron microscopy. The IS is composed of cubical ciliated cells which propel the urine along the renal tubule. The distal tubule is divided into two portions: the early distal tubule (EDT) and the late distal tubule (LDT). The EDT is characterized by only one type of cells with well developed basolateral interdigitations and numerous elongated mitochondria, which are oriented normal to the basal surface. In order to reveal the differentiation characteristics of organelles of ciliates under different physiological status, the cellular ultrastructure of Urostyla grandis was studied by transmission electron microscopy. In the resting cells most ciliary shafts, kinetosomes and sub-pellicle microtubules were resorbed, and the endoplasmic reticulum (ER) disappeared with the autophagocytosis taking place within the cytoplasm. In sexually reproducing organisms, primordial germ cells (PGCs) give rise to the cells of the germ line, the gametes. In many animals, PGCs are set apart from somatic cells early during embryogenesis. This study explores the origin of primordial germ cells (PGCs) of the zebra fish and examines their morphology during early development (1st day-15th day). PGCs were selectively stained by the alkaline phosphatase histochemical reaction and viewed by light and electron microscopy from the time they are first detectable in the yolk sac endoderm. Abstract: Tryblionella is a large epipelic genus, widespread from fresh to marine waters. Most of the studies including the genus are based only in light microscopy. In this paper we present eleven species of Tryblionella found in epiphyton and microphytobenthos in Patos Lagoon salt marshes. Melosira (Diatomeae) taxa from the Iguaçu River in southern Brazil Biota Neotropica

Abstract The study of the genus Melosira in plankton samples from the lower Iguaçu River revealed the presence of four taxa: Melosira undulata (Ehrenberg) Kützing var. undulata, M. undulata var. normanni Arnott, M. varians C. Agardh and M. muscigena Iwahashi. We present illustrations of the frustules using light microscopy (LM), descriptions, and comments about the morphology of the four taxa. The analysis of Melosira muscigena under scanning electron microscopy revealed unprecedented details of the ultrastructure, such as the shape and distribution of the rimoportulae at the valve mantle. This is the first record of Melosira undulata var. undulata and the second of M. muscigena in Brazil.

Gomphonema Ehrenberg (Bacillariophyceae, Gomphonemataceae) of the São Francisco Falso River, Paraná, Brazil Biota Neotropica

Abstract: Gomphonema Ehrenberg is a genus well represented in periphytic algal flora of epicontinental environments. The substrate colonization is facilitated by the secretion of mucilage by the pore field, allowing cell adhesion. Samples were scraped off and analyzed from vegetative portions of the macrophyte Eichhornia azurea Kunth, collected in São Francisco Falso River, located in Santa Helena City, Paraná State. The sampling occurred in four different sites, in four different periods, totaling 16 samples. Morphology and ultrastructure of Schizaea fistulosa (Schizaeaceae) spores from Chile Boletín de la Sociedad

The spores of Schizaea fistulosa from Chile were studied using light microscopy (LM), scanning (SEM) and transmission electron microscopy (TEM). The spores are monolete and elliptic in polar view. The major equatorial diameter is 71-85 µm and the polar diameter is 54-61 µm. The laesurae are 50-60 µm long and, in some cases, bifurcated. The sporoderm ultrastructure is first mentioned and described here. The exospore is two-layered in section, verrucate-tuberculate with single or fused elements forming short ridges. Submicroscopical Features of Leaves of Xyris Species BABT

The leaf ultrastructure of five Xyris species were examined using scanning electron microscope (SEM), transmission electron microscope (TEM) and histochemical methods. All studied leaves show some features in epidermis and mesophyll, which were of considerable adaptative significance to drought stress. Such features included the occurrence of a pectic layer on the stomatal guard cells and the presence of a network of pectic compounds in the cuticle. Pectic compunds were also in abundance in lamellated walls of the mesophyll cells and on the inner surface of the sclerified cell walls of the vascular bundle sheaths. There were also specialized chlorenchymatous "peg cells" in the mesophyll and drops of phenolic compounds inside the epidermal cells.

Seminal analysis, cryogenic preservation, and fertility in matrinxã fish, Brycon cephalus (Günther, 1869) BABT

Aiming to improve fish reproduction techniques, the characterization and cryopreservation of semen of Brycon cephalus were performed. The seminal characteristics observed were: an almost transparent, milky semen with a mean volume of 4mL, and sperm concentration of 9.617±1.630 x 10(6) spermatozoa/mm³. Bulliform cells in Loudetiopsis chrysothrix (Nees) Conert and Tristachya leiostachya Nees (Poaceae): structure in relation to function BABT

This work reports anatomic and ultrastructural characteristics of bulliform cells in Loudetiopsis chrysothrix (Nees) Conert and Tristachya leiostachya Nees. Both the species presented leaf rolling under water stress. The main characteristics observed in these cells were: periclinal wall thinner than the adjacent epidermal wall; abundance of pectic substances in cuticular layer; sinuous anticlinal walls with ramified plasmodesmata; vacuome formed by a developed vacuole or innumerous small vacuoles; abundance of phenolic substances and oil drops. These characteristics suggested the involvement of bulliform cells in the mechanism of foliar involution in the studied species.
