Estuaries and coastal areas are often subjected to much pollution, including pesticides and polycyclic aromatic hydrocarbons (PAHs) produced by anthropic activities. Bay of Marennes-Oléron is an estuarine coastal zone in which there is a high level of aquacultural production and this has adverse effects in aquatic organisms. In fact, PAHs, which are widely spread throughout aquatic ecosystems, have a low polarity and so there is a clear tendency to bioaccumulate in marine organisms (1). The Gram-negative bacterium Vibrio harveyi is known to be highly pathogenic for the European abalone Haliotis tuberculata, which is a gastronomically important marine gastropod with a high commercial value. Since 1998, some particular bacterial strains are described as implicated in recurrent mortality outbreaks in French farm and field stocks of abalone. Cytogenetic abnormalities associated with viral infections, including from viruses of the Herpesvirales order, have been reported in vertebrate species. Ostreid herpesvirus 1 (OsHV-1) has been detected worldwide during mortality outbreaks of the Pacific oyster Crassostrea gigas. On the other hand, a high proportion of aneuploid cells in somatic tissues have been observed in C. gigas. In this study, we analysed the putative association between aneuploidy levels and the detection of OsHV-1 in gills of C. gigas, the Portuguese oyster C. angulata and their F1 hybrids cultured in Ria Formosa (Portugal). Défis de la surveillance des maladies chez les coquillages marins en France ArchiMer

The basic concepts and objectives of animal disease surveillance are common to all types of animal production. Yet, the procedures for implementation have been mostly developed for terrestrial animal productions. Disease surveillance in marine shellfish calls for specific measures in view of the peculiarities of marine biology and rearing techniques. The main objective of disease surveillance in marine shellfish is to detect as early as possible exotic and newly emerging diseases. With this objective in mind, a national surveillance network for surveillance of mollusc diseases called Repamo has been developed. It combines passive reporting and risk-based surveillance.

Detection of ostreid herpesvirus-1 (OsHV-1) by PCR using a rapid and simple method of DNA extraction from oyster larvae ArchiMer

A DNA extraction procedure was developed for the detection of ostreid herpesvirus-1 (OsHV-1) using the polymerase chain reaction (PCR) in oyster larvae. The DNA extraction procedure developed was tested on 8 larval samples. Abnormal nuclei with characteristic features associated with OsHV-1 infections were only observed in samples in which the viral DNA was detected by PCR. A previously described competitive PCR method was applied to detect inhibition during PCR reactions. The results show that the method can be used on small amounts of oyster larvae (3 mg) for the detection of OsHV-1 DNA by PCR.

Characterization of abalone Haliotis tuberculata-Vibrio harveyi interactions in gill primary cultures ArchiMer

The decline of European abalone Haliotis tuberculata populations has been associated with various pathogens including bacteria of the genus Vibrio. Following the summer mortality outbreaks reported in France between 1998 and 2000, Vibrio harveyi strains were isolated from moribund abalones, allowing in vivo and in vitro studies on the interactions between abalone H. tuberculata and V. harveyi.
