Effect of an energy-deficient diet on populations of ciliate protozoans in bovine rumen Arq. Bras. Med. Vet. Zootec.
Soares,P.C.; Martinele,I.; D'Agosto,M.; Maruta,C.A.; Sucupira,M.C.A.; Antonelli,A.C.; Mori,C.S.; Ortolani,E.L..
Ten young rumen-cannulated crossbred steers were randomly divided into two groups: a control group (C; n=4), which was fed a balanced diet for daily weight gain of 900g; and a pronounced energy-deprived group (PED; n=6), receiving 30% less of the required energy for maintenance. After 140 days of these alimentary regimes, rumen fluid and urine samples were collected for biochemical and functional tests, before feeding and at 1, 3, 6, and 9 hours after feeding. The energy-deprivation diet caused a significant reduction in the number of Entodinium, Eodinium, Isotricha, Dasytricha, Eremoplastron, Eudiplodinium, Metadinium, Charonina, Ostracodinium, and Epidinium protozoa. There was no effect of the time of sampling in both groups on the total number of...

Polymorphisms of the coding region of Slc11a1 (Nramp1) gene associated to natural resistance against bovine brucellosis Arq. Bras. Med. Vet. Zootec.
Paixão,T.A.; Martinez,R.; Santos,R.L..
No presente estudo, três novos polimorfismos do gene Slc11A1...

Impaired protamination and sperm DNA damage in a Nellore bull with high percentages of morphological sperm defects in comparison to normospermic bulls Arq. Bras. Med. Vet. Zootec.
Carreira,J.T.; Trevizan,J.T.; Kipper,B.H.; Perri,S.H.V.; Carvalho,I.R.; Rodrigues,L.H.; Silva,C.; Koivisto,M.B..
The routine semen evaluation assessing sperm concentration, motility and morphology, does not identify subtle defects in sperm chromatin architecture. Bulls appear to have stable chromatin, with low levels of DNA fragmentation. However, the nature of fragmentation and its impact on fertility remain unclear and there are no detailed reports characterizing the DNA organization and damage in this species. The intensive genetic selection, the use of artificial insemination and in vitro embryo production associated to the cryopreservation process can contribute to the chromatin damage and highlights the importance of sperm DNA integrity for the success of these technologies. Frozen-thawed semen samples from three ejaculates from a Nellore bull showed high...

Molecular characterization of Malassezia sympodialis and Malassezia furfur from cattle with and without otitis Arq. Bras. Med. Vet. Zootec.
Duarte,E.R.; Hamdan,J.S..
A molecular study of Malassezia strains isolated from cattle with or without otitis was carried out by random amplified polymorphic DNA analysis (RAPD). DNA was extracted and purified from nine strains of Malassezia sympodialis and fourteen of Malassezia furfur. These microorganisms were collected from eight different bovine herds in Minas Gerais state, Brazil. The RAPD analysis and phenograms did not show the formation of genetically distinct groups among the strain isolated from cattle with or without otitis raised in the same herds. Genetic heterogeneity was observed among Malassezia strains from different geographic origins. These data suggest that genetically similar M. sympodialis and M. furfur strains found as members of the normal ear microbiota...

Hajarian,H; Wahid,H; Rosnina,Y; Daliri,M; Dashtizad,M; Karamishabankareh,H; Abas Mazni,O.
The effectiveness of different cryodevices (open-pulled straw (OPS), electron microscopy grid (EMG), and Cryotop was evaluated for vitrification of immature bovine oocytes. Polar body, metaphase II stage (MII), survivability, and subsequent developmental rates were determined. Only oocytes with four or five layers of cumulus cells were used. Oocytes were equilibrated in two vitrification solutions - 1: 10% DMSO + 10% ethylene glycol (EG) for 30-45sec and 2: 20% DMSO + 20% EG +0.5M sucrose for 25sec -, mounted on one of the cryodevices and directly plunged into liquid nitrogen for 10 days. Immature vitrified oocytes using Cryotop showed the highest rates of polar body extrusion (PB) and nuclear maturity (MII); 41 and 58% respectively. Vitrified oocytes...

Effect of concentration and exposure period to butyrolactone I on meiosis progression in bovine oocytes Arq. Bras. Med. Vet. Zootec.
Adona,P.R.; Leal,C.L.V..
The effect of concentration and exposure period of bovine oocytes to butyrolactone I (BLI) on meiotic block and in vitro maturation (IVM) kinetics was studied. In experiment 1, all oocytes were at germinal vesicle stage (GV), after 6h in culture with 0, 50 and 100µM BLI. After 12h, all oocytes cultured with 50 and 100µM BLI remained in GV. After 24h, less oocytes were in GV with 50µM (82%) than with 100µM BLI (99%, P<0.05). In experiment 2, after 6h IVM, 93% of control oocytes (IVM only) were in GV, while treated oocytes (100µM BLI for 6, 12 or 24h prior to IVM) showed less oocytes in GV with increased exposure period to BLI prior to IVM (83 and 73%, for 6h and 12h, P<0.05). For a 24h inhibition, GV rates were similar to 12h (70%, P>0.05). After...

Bovine placentome preservation for light microscopy evaluation Arq. Bras. Med. Vet. Zootec.
Martins,E.; Martins,V.M.V.; Marques Júnior,A.P.; Vasconcelos,A.C.; Chiarini-Garcia,H.; Malard,P.F..
This work evaluates the preservation of bovine placentomes using paraformaldehyde and glutaraldehyde at 4% and solutions of Karnovsky, Bouin and Carnoy for 4, 12 and 24 hours fixation, at room temperature or at 4ºC, before inclusion in plastic resin and paraffin. The best preservation of the specimens was obtained after 12 hours of fixation with those solutions containing aldehydes. With Bouin solution the best time of fixation was 4 hours, while for Carnoy solution the best time of fixation was 24 hours. Inclusion in plastic resin gave better results than in paraffin, and the temperature did not influence the quality of preservation of the specimens of bovine placentomes for evaluation with light microscopy.

Reactive oxygen species in bovine embryo in vitro production Biocell
Dalvit,G.C.; Cetica,P.D.; Pintos,L.N.; Beconi,M.T..
Oxidative modifications of cell components due to the action of reactive oxygen species (ROS) is one of the most potentially damaging processes for proper cell function. However, in the last few years it has been observed that ROS participate in physiological processes. The aim of this work was to determine ROS generation during in vitro production of bovine embryos. Cumulus-oocyte complexes were recovered by aspiration of antral follicles from ovaries obtained from slaughtered cows and cultured in medium 199 for 22 h at 39ºC in 5% CO2: 95% humidified air. In vitro fertilization was carried out in IVF-mSOF with frozenthawed semen in the same culture conditions and embryo in vitro culture in IVC-mSOF at 90% N2: 5% CO2: 5% O2. ROS was determined in denuded...

Hematological and cerebrospinal fluid changes in cattle naturally and experimentally infected with the bovine herpesvirus 5 BABT
Lisbôa,Júlio Augusto Naylor; Isernhagen,Allan Jürgen; Borges,Alexandre Secorun; Amorim,Rogério Martins; Balarin,Mara Regina Stipp; Lunardi,Michele; Alfieri,Amauri Alcindo.
Bovine herpesvirus 5 (BoHV-5) meningoencephalitis is one of the main causes of mortality by encephalopathy in Brazilian cattle herds. However, the neurological signs observed are common to several encephalopathies and do not contribute decisively to a diagnosis. In order to verify hematological and cerebrospinal fluid (CSF) changes, blood and CSF samples from naturally and experimentally infected bovines were evaluated. In natural cases (n=17), the samples were collected only once, and in experimental cases (n=7), the samples were sequentially obtained throughout disease progression. While routine methods were used to examine the samples, BoHV-5 infection was confirmed by a PCR assay. Blood analyses did not reveal any consistent hematological alterations...

Validation of a PCR Assay for Chlamydophila abortus rRNA gene detection in a murine model BABT
Silva-Zacarias,Francielle Gibson da; Alfieri,Amauri Alcindo; Spohr,Kledir Anderson Hofstaetter; Lima,Bruna Azevedo de Carvalho; Negrão,Fábio Juliano; Lunardi,Michele; Freitas,Julio Cesar de.
Chlamydophila abortus (C. abortus) is associated with reproductive problems in cattle, sheep, and goats. Diagnosis of C. abortus using embryonated chicken eggs or immortalized cell lines has a very low sensitivity. Polymerase chain reaction (PCR) assays have been used to detect C. abortus infection in clinical specimens and organ fragments, such as placenta, fetal organs, vaginal secretions, and semen. The aim of this study was to develop a PCR assay for the amplification of an 856-bp fragment of the rRNA gene of the Chlamydiaceae family. The PCR assay was evaluated using organs from 15 mice experimentally infected with the S26/3 reference strain of C. abortus. The results of the rRNA PCR were compared to the results from another PCR system (Omp2 PCR) that...

Effect of charcoal-treated bovine follicular fluid on the secretion of FSH in ovariectomized and intact prepubertal heifers Braz. J. Vet. Res. Anim. Sci.
ALVAREZ,Rafael Herrera; ARCARO,Juliana Rodrigues Pozzi; RIBELA,Maria Teresa Carvalho Pinto; PERONE,Cibele Nunes.
The aim of the experiment was to determine whether charcoal-treated bovine follicular fluid (bFF) removed from visible (< 22 mm) follicles altered the secretion of follicle-stimulating hormone (FSH) in intact and ovariectomized prepubertal heifers. After a 10-ml injection of bFF given at three consecutive 8-h intervals, secretion of FSH was depressed in approximately 44% of the ovariectomized heifers but there was no effect in intact heifers. When the bFF treatment ceased, there were no rebound effects on FSH concentrations above that of controls pretreatment levels. These results suggest that proteins from bFF act at the pituitary level to inhibit FSH secretion and, differently of the intact, the ovariectomized heifer is an adequate model to put this...

Due to the high importance of the venereal transmission of bovine leptospirosis, this study aimed to test the ability of PCR to detect Leptospira interrogans serovar hardjo DNA in experimentally contaminated bovine semen. Employing primers directed to the 16S rRNA gene, 10 bacteria/ml of semen could be detected by PCR. Results achieved in this work show that PCR can have a great potential to detect Leptospira spp. in insemination centers.
