Early reproductive effort and growth were measured in 3 groups of the Pacific oyster Crassostrea gigas: 1 diploid group and 2 triploid groups resulting from either chemical induction (3nCB) or crosses between tetraploid and diploid parents (3nDT). Oysters were reared under intensive nursery conditions and sampled when 5 mo old. Reproductive effort was estimated by cross-sectional area measurements of the visceral mass (i.e. gonadic occupation) and maturation stage was assessed by qualitative histology. Ferritin has been shown as being the principal iron storage in the majority of living organisms. In marine species, ferritin is also involved in high-level accumulation of Po-210. As part of our work on the investigation of these radionuclides' concentration in natural environment, ferritin was searched at the gene and protein level. Ferritin was purified from the visceral mass of the oyster Crassostrea gigas by ion-exchange chromatography and HPLC. SDS-PAGE revealed one band of 20 kDa. The taxonomic status of Crassostrea angulata and Crassostrea gigas has long been a matter of controversy. Morphological and physiological similarities, as well as homogeneity in allelic frequencies on allozymes between the populations of the two taxa, lead most authors to suggest to regroup of the two within the same species. European and Asian populations of C. gigas and C. angulata have been studied using microsatellite and mitochondrial DNA markers. A semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) assay was developed to estimate tissue-specific expression of two amylase genes in five different tissues of the Pacific oyster Crassostrea gigas and the experimental effects of food level on the cellular, enzymatic and gene expression changes of the digestive gland. Lipid spray beads (SB) were prepared containing 13% w/w particulate riboflavin. Beads suspended in seawater lost 73% riboflavin after 24 h. Release of riboflavin from SE ingested by Pacific oyster (Crassostrea gigas) larvae was observed under epifluorescent light. Riboflavin concentrations in tissues of adult oysters fed on riboflavin-SB were significantly (SNK, P < 0.05) greater than those of oysters fed on seawater-filled SE. To investigate the control at the mRNA level of glycogen metabolism in the cupped oyster Crassostrea gigas, we report in the present paper the cloning and characterization of glycogen phosphorylase and synthase cDNAs (Cg-GPH and Cg-GYS, respectively, transcripts of main enzymes for glycogen use and storage), and their first expression profiles depending on oyster tissues and seasons. Oyster species suffer from numerous disease outbreaks, often causing high mortality. Because the environment cannot be controlled, genetic improvement for disease resistance to pathogens is an attractive option to reduce their impact on oyster production. We review the literature on selective breeding programs for disease resistance in oyster species, and the impact of triploidy on such resistance. The protozoan Bonamia ostreae (Ascetospora), a paasite of the flat oyster Ostrea edulis, was purified by differential and isopycnic centrifugations. Mice of the strain Balb/c were immunized with purified parasites and hybridomas were prepared by fusion of immunized mouse splenocytes with the mouse myeloma cell line P3-X63-Ag8-653.
