Assortative fertilization and selection at larval stage in the mussels Mytilus edulis and M-galloprovincialis ArchiMer
Bierne, Nicolas; David, Pierre; Boudry, Pierre; Bonhomme, F.
Assortative mating (prezygotic isolation) and reduced hybrid fitness (postzygotic isolation) are typically invoked to explain the stability of hybrid zones. In the tension zone model. these factors work in opposition to migration, which promotes genetic homogeneity. Many marine animals migrate over long distances through a planktonic larval stage. Therefore. strong reproductive isolation is needed to maintain stable marine hybrid zones. However, surprisingly little is known about mating preferences and hybrid fitness in marine organisms. Smooth-shelled mussels (Mytilus spp.) form a well-known species complex, with hybridization over extensive areas such as the contact zone of M. edulis and M. galloprovincialis around European Atlantic coasts. This paper...

Combining Two-Stage Testing and Interval Mapping Strategies to Detect QTL for Resistance to Bonamiosis in the European Flat Oyster Ostrea edulis ArchiMer
Lallias, Delphine; Gomez-raya, L.; Haley, C. S.; Arzul, Isabelle; Heurtebise, Serge; Beaumont, A. R.; Boudry, Pierre; Lapegue, Sylvie.
We have identified quantitative trait loci (QTL) in the flat oyster (Ostrea edulis) for resistance to Bonamia ostreae, a parasite responsible for the dramatic reduction in the aquaculture of this species. An F-2 family from a cross between a wild oyster and an individual from a family selected for resistance to bonamiosis was cultured with wild oysters injected with the parasite, leading to 20% cumulative mortality. Selective genotyping of 92 out of a total of 550 F-2 progeny (i.e., 46 heavily infected oysters that died and 46 parasite-free oysters that survived) was performed using 20 microsatellites and 34 amplification fragment length polymorphism primer pairs. Both a two-stage testing strategy and QTL interval mapping methods were used. The two-stage...

The Kinome of Pacific Oyster Crassostrea gigas, Its Expression during Development and in Response to Environmental Factors ArchiMer
Epelboin, Yanouk; Quintric, Laure; Guevelou, Eric; Boudry, Pierre; Pichereau, Vianney; Corporeau, Charlotte.
Oysters play an important role in estuarine and coastal marine habitats, where the majority of humans live. In these ecosystems, environmental degradation is substantial, and oysters must cope with highly dynamic and stressful environmental constraints during their lives in the intertidal zone. The availability of the genome sequence of the Pacific oyster Crassostrea gigas represents a unique opportunity for a comprehensive assessment of the signal transduction pathways that the species has developed to deal with this unique habitat. We performed an in silico analysis to identify, annotate and classify protein kinases in C. gigas, according to their kinase domain taxonomy classification, and compared with kinome already described in other animal species....

Temperature influence on pathogen transmission and subsequent mortalities in juvenile Pacific oysters Crassostrea gigas ArchiMer
Petton, Bruno; Pernet, Fabrice; Robert, Rene; Boudry, Pierre.
Since 2008, mass mortalities of 1-yr-old Crassostrea gigas associated with the ostreid herpesvirus OsHV-1 μVar have occurred along all the coasts of France when seawater temperature reaches 16 to 17°C. The present study aimed to characterize the effect of temperature on oyster survival in combination with OsHV-1 DNA quantification by standard real-time PCR and total vibrio population levels in oyster tissues. To examine the effect of seawater temperature on disease transmission and related mortality of oysters, cohabitation experiments were conducted between healthy naïve oysters and oysters previously exposed to field conditions in areas where mortalities were occurring. Oysters initially maintained in controlled conditions (free of mortality and negative...

Recent advances in EST sequencing in Crassostrea gigas: towards the sequencing of the Pacific oyster genome ArchiMer
Boudry, Pierre; Huvet, Arnaud; Lapegue, Sylvie; Sauvage, Christopher; Tanguy, A.; Moraga, D.; Goostrey, A.; Prunet, P.; Boulo, Viviane; Gueguen, Y.; Moal, Jeanne; Fleury, Elodie; Samain, Jean-francois; Mathieu, M.; Favrel, P..
Rationale for EST and genome sequencing of the Pacific oyster: • its membership of the Lophotrochozoa, an little studied branch of the Eukaryotes, • its worldwide aquacultural importance (4.2 million metric tons, worth 3.5 billion US dollars), • its high fecundity, with concomitantly high DNA polymorphism, • its use as a biosensor of coastal pollution. In 2004, an international community of biologists, the Oyster Genome Consortium (OGC) led by D. Hedgecock (University of Southern California) and uniting 70 participants from 10 countries, first presented the Pacific oyster Crassostrea gigas as a genome-sequencing candidate to Joint Genome Institute (JGI, USA) (Hedgecock et al., 2005). Besides this project (that remains to be accepted), several EST...

The taxonomic status and origin of the Portuguese oyster Crassostea angulata (Lamark, 1819) ArchiMer
Batista, Frederico; Leitao, Alexandra; Huvet, Arnaud; Lapegue, Sylvie; Heurtebise, Serge; Boudry, Pierre.
The taxonomic status of the Portuguese oyster Crassostrea angulata and the Pacific oyster Crassostrea gigas has often been a matter of controversy. Based on larval shell morphology, experimental hybridisation and electrophoretic studies of enzyme polymorphism several authors have considered these two species as being synonymous. During the recent years, several genetic studies based on mitochondrial DNA and microsatellites data have provided evidences that the two taxa are genetically distinct although closely related. Furthermore, karyotype analyses have also highlighted the close genetic similarity of these taxa in comparison with other cupped oyster species. However, a comparative analysis of restriction enzymes ideograms revealed differences between...

Characterization of GnRH-related peptides from the Pacific oyster Crassostrea gigas ArchiMer
Bigot, Laetitia; Zatylny-gaudin, Celine; Rodet, Franck; Bernay, Benoit; Boudry, Pierre; Favrel, Pascal.
Gonadotropin-releasing hormone (GnRH), a key neuropeptide regulating reproduction in vertebrates has now been characterized in a number of non-vertebrate species. Despite the demonstration of its ancestral origin, the structure and the function of this family of peptides remain poorly known in species as distant as lophotrochozoans. In this study, two GnRH-related peptides (Cg-GnRH-a and CgGnRH-G) were characterized by mass spectrometry from extracts of the visceral ganglia of the Pacific oyster Crassostrea gigas. These peptides showed a high degree of sequence identity with GnRHs of other mollusks and annelids and to a lesser extent with those of vertebrates or with AKH and corazonins of insects. Both the mature peptides and the transcript encoding the...

Apport d'un programme de génétique à une filière de production aquacole : l'exemple de l'ostréiculture ArchiMer
Lapegue, Sylvie; Bedier, Edouard; Goyard, Emmanuel; Degremont, Lionel; Baud, Jean-pierre; Gerard, Andre; Goulletquer, Philippe; Boudry, Pierre.
Two oyster species are currently present along the French coasts : the indigenous European flat oyster (Ostrea edulis), and the Pacific cupped oyster (Crassostrea gigas), that has been introduced from Japan since the beginning of the 70ies. The flat oyster successively suffered from two protozoan diseases during the 60ies and its production decreased from 20 000 tons/year by that time to 1 500 tons/year nowadays. Consequently, the oyster production is principally (99%) based upon the Pacific oyster species with approximately 150 000 tons/year among which 90% are grown from the natural spat. However, the hatchery production of this species is developing and was estimated to 400 to 800 millions spat in 2002. Moreover, strengthened relationships between...

Etude des ressources génétiques chez les huîtres creuses ArchiMer
Boudry, Pierre; Heurtebise, Serge; Lapegue, Sylvie; Lam, Katherine; Mccombie, Helen; Leitao, Alexandra; Gerard, Andre.

Characterisation of physiological and immunological differences between Pacific oysters (Crassostrea gigas) genetically selected for high or low survival to summer mortalities and fed different rations under controlled conditions ArchiMer
Delaporte, Maryse; Soudant, P; Lambert, C; Jegaden, M; Moal, Jeanne; Pouvreau, Stephane; Degremont, Lionel; Boudry, Pierre; Samain, Jean-francois.
Within the framework of a national scientific program named "MORtalites ESTivales de l'huitre creuse Crassostrea gigas" (MOREST), a family-based experiment was developed to study the genetic basis of resistance to summer mortality in the Pacific oyster, Crassostrea gigas. As part of the MOREST project, the second generation of three resistant families and two susceptible families were chosen and pooled into two respective groups: "R" and "S". These two groups of oysters were conditioned for 6 months on two food levels (4% and 12% of oyster soft-tissue dry weight in algal dry weight per day) with a temperature gradient that mimicked the Marennes-Oleron natural cycle during the oyster reproductive period. Oyster mortality remained low for the first two...

Variable microsatellites in the Pacific Oyster Crassostrea gigas and other cupped oyster species ArchiMer
Huvet, Arnaud; Boudry, Pierre; Ohresser, M; Delsert, Claude; Bonhomme, F.
Source/description : Genomic DNA was extracted from a whole Pacific oyster (C. gigas) after grinding in liquid nitrogen. Purified DNA was then digested by a mix of 3 restriction enzymes (AluI, RsaI, HaeIII). DNA fragments ranging from 250 to 500 bp were size-selected by agarose gel electrophoresis and ligated into the dephosphorylated blunt-ended SmaI-linearised pBKS2 plasmid (Stratagene Cloning System, CA). White colonies (11,500) were screened by hybridisation with double-stranded DNA probes containing dinucleotide repeats. Poly(dAdC), poly(dAdG), poly(dAdT) (Pharmacia Biotech, Sweden) were mixed at equimolar ratio and radiolabelled by [-32P] alpha dATP and dCTP using a random priming labelling kit (Life Technologies, Germany). Reproductive effort of Pacific oysters: A trait associated with susceptibility to summer mortality ArchiMer
Huvet, Arnaud; Normand, Julien; Fleury, Elodie; Quillien, Virgile; Fabioux, Caroline; Boudry, Pierre.
Summer mortality of the Pacific oyster Crassostrea gigas is the result of a complex interaction between oysters, their environment and their pathogens. The physiological status of an oyster, especially its reproductive status, is suspected to play a significant role in the outcome of this interaction. As genetic variability exists for susceptibility to summer mortality, resistant (R) and susceptible (S) oyster lines were produced using a divergent selection scheme. The present paper reports a histological study on gonad area, which is representative of reproductive effort, in randomly chosen five R and five S oyster lines. The R lines showed a significantly lower gonad area than the S lines (P<0.001). with an estimated mean difference of 12.5%, whereas,...

Impact of intensive larval rearing practices in oyster hatcheries : selective processes and loss of genetic variability (Poster) ArchiMer
Taris, Nicolas; Sauvage, Christopher; Boudry, Pierre.
Early-life history of oysters, like in most marine bivalves, is characterized by high fecundity and low survival rate. In hatcheries, which are ecoming more and more important for the production of juveniles for aquaculture production, rearing conditions are optimised (density, feeding, temperature...), and contrast with those observed in the wild. Additionally, slow growing larvae are culled by sieving.

We used a 2-DE proteomic approach to identify abundant proteins linked to oocyte quality in the Pacific oyster Crassostrea gigas, an economically important bivalve. Oocyte quality of 14 females was estimated by recording fertilisation and early developmental success until D-larval stage under controlled conditions. Proteins that were differentially expressed between females showing high or low oocyte quality were identified by nano-liquid chromatography tandem mass spectrometry. Twelve up-accumulated spots associated with low quality oocytes revealed 10 distinct proteins, including vitellogenin — breakdown products and metabolic enzymes. Eight up-accumulated spots from high quality oocytes revealed 6 distinct proteins, including chaperone molecules and...

Genetic variation and trade-offs for reproduction ans survival in the Pacific oyster Crassostrea gigas ArchiMer
Boudry, Pierre; Degremont, Lionel; Bedier, Edouard; Pouvreau, Stephane; Normand, Julien; Ernande, Bruno.
To date, the most significant method to genetically reduce reproductive effort in the Pacific oyster (Crassostrea gigas) has been through the production of triploids, especially since the development of tetraploid lines allowing the breeding of 'natural' triploids. Gametogenesis of triploid oysters is strongly reduced compared with diploids, although they are not fully sterile and can produce viable gametes and some progenies when crossed with diploids. Reduced reproductive allocation and higher heterozygocity are commonly proposed as the main reasons why triploids often present superior yield compared with diploids. Temperature and food availability are known to favour gametogenesis in triploids, but the existence of genetic variation for this trait...

QTL mapping for resistance to bonamiosis in the European flat oyster Ostrea edulis ArchiMer
Lallias, Delphine; Beaumont, Andy; Haley, Chris; Heurtebise, Serge; Boudry, Pierre; Lapegue, Sylvie.
A breeding programme has been undertaken by Ifremer to produce selected Ostrea edulis families which are resistant to Bonamia ostreae, a parasite which is the most significant constraint on aquaculture of this oyster. Using such families, we aimed to identify Quantitative Trait Loci (QTLs) of resistance/susceptibility to bonamiosis. We used an F2 family derived from a bi-parental cross between a wild oyster and an individual from one of the selected oyster families. A total of 550 F2 individuals from the family cohabited with wild oysters that were injected with the parasite. Mortality was checked daily over six months and heart smears were used on moribund or dying oysters to detect the presence and level of infection of the parasite. Ninety two F2...
