Overlapping of mononuclear cells derived from bone marrow in rats' intervertebral discs: an in vitro study Ciência Rural
Fontes,Erica Batista; Borges,Andréa Pacheco Batista; Pippi,Ney Luis; Rosa,Maurício; Sprada,Arícia; Togni,Monique.
In this study, bone marrow mononuclear cells (BM-MNC) derived from rats were used in order to promote intervertebral disc regeneration. These cells were isolated after centrifugation in a Ficoll-Paque™ PLUS density gradient and then placed in plastic dishes to proliferate during a period of 14 days. The BM-MNCs were previously labeled with the fluorescent membrane marker Chloromethyl-benzamidodialkylcarbocyanine (CM-DIL), and thereafter were implanted in rats' intervertebral discs explants as an in vitro experimental model. Daily analyses of the cells under a fluorescence microscope revealed morphological changes, which assumed a thin and elongated shape similar to cells that originally form the annulus fibroses. Histopathological analysis demonstrated the...

Exogenous ATP administration prevents ischemia/reperfusion-induced oxidative stress and tissue injury by modulation of hypoxanthine metabolic pathway in rat ovary Ciência Rural

In this study, xanthine oxidase (XO), malondialdehyde (MDA), myeloperoxidase (MPO) and glutathione (GSH) levels in the ovarian tissues of rats during the development of ischemia and postischemia-induced reperfusion were investigated, and the effect of ATP on ischemia-reperfusion (I/R) damage was biochemically and histopathologically examined. The results of the biochemical analyses demonstrated that ATP significantly reduced the level of XO and MDA and increased the amount of GSH in both ischemia and I/R-applied ovarian tissue at the doses administered. Furthermore, ATP significantly suppressed the increase in MPO activity that occurred following the application of post ischemia reperfusion in the ovarian tissue. The biochemical results obtained in the...

Coconut oil on biochemical and morphological parameters in rats submitted to normolipidic and hyperlipidic diets Ciência Rural
Schumacher,Bianca de Oliveira; Preuss,Edcarlos Maurino; Vargas,Carolina Galarza; Helbig,Elizabete.
ABSTRACT: This study aimed to evaluate the influence of replacing soybean oil with extra virgin coconut oil in normolipidic and hyperlipidic diets, on the lipid metabolism of Wistar rats. In the first stage of the experiment (30 days), 36 rats were divided into 2 groups and fed with a control or a hyperlipidic diet. Six animals from each group were then killed, and the remaining rats were redistributed into 4 new groups: 2 groups remained on the control and hyperlipidic diets, and in the diets of the other 2 groups, the soybean oil was replaced with coconut oil (30 days). At the end of the assay, the biological models were decapitated for blood collection and removal of organs and peritoneal fat. Although the diet intake differed among groups during both...
