Sucrose hydrolysis catalyzed by auto-immobilized invertase into intact cells of Cladosporium cladosporioides Electron. J. Biotechnol.

The enzyme known as invertase (E.C. 3.2.1.26 - β-D-fructofuranosidase) catalyzes the sucrose hydrolysis producing an equimolar mixture of glucose and fructose named inverted sugar. The fungus Cladosporium cladosporioides has invertase as its constituent. Hence, its use as a natural immobilized support for the invertase produces interesting results for the enzyme. The present work has the objective of determining the optimum operational conditions of auto-immobilized invertase, as well as its kinetic parameters (K M and Vmax). Gill tissues of the mussel Mytilus edulis chilensis: A new source for antimicrobial peptides Electron. J. Biotechnol.

Antimicrobial peptides are small-sized, cationic and amphipathic molecules able to neutralize pathogenic microorganisms. Their antimicrobial effects tie them to mechanisms of immune defense, which is why they have been normally purified from immune cells. We describe an apparently new group of antimicrobial peptides from gill tissues of the mussel Mytilus edulis chilensis. 20 specimens yielded 40 g of gills which produced 16 mg of an enriched fraction with antimicrobial activity as low as 0.045 µg/µl over reference strains. The genus Scoparia is native from Argentina. The diversity of colours and shape of their flowers bestows this genus a very interesting ornamental potential. The purpose of the present study is to explore the Scoparia species germplasm by means of in vitro polyploidization in order to improve their ornamental qualities. Accessions of S. montevidiensis var. montevidiensis, S. montevidiensis var. glandulifera, S. nudicaulis, S. hasleriana and S. dulcis were collected and maintained under greenhouse conditions. Effects of temperature on the sorption of Pb2+ and Cd2+ from aqueous solution by Caladium bicolor (Wild Cocoyam) biomass Electron. J. Biotechnol.

This report is based on the investigation of the effect of temperature on the removal of Pb2+ and Cd2+ in aqueous effluent using C. bicolor biomass in a batch sorption process. The result showed that the most suitable sorption temperature was 40ºC with maximum sorption capacities of 49.02 mg/g and 52.63 mg/g for Pb2+ and Cd2+ respectively. Various thermodynamic parameters, such as ΔGº, ΔHº, ΔSº and Ea have been calculated. The data showed that the sorption process is spontaneous and exothermic in nature and that lower solution temperatures favours metal ion removal by the biomass. Production of an extracellular keratinase from Chryseobacterium sp. growing on raw feathers Electron. J. Biotechnol.

The strain Chryseobacterium sp. kr6 shown to be useful for biotechnological purposes such as hydrolysis of poultry feathers and de-hairing of bovine pelts. The effect of temperature, initial pH and media composition on protease production by this keratinolytic strain was studied. The enzyme was produced between 25 and 37ºC, with maximum activity and yield at 30ºC. When protease production was tested in media with different initial pH, maximum activity was observed when cultivation was carried out at 30ºC and initial pH ranging from 6.0 to 8.0. Growth responses of Betula pendula ecotypes to red and far-red light Electron. J. Biotechnol.

The effect of Red light (R), Far-red light (FR) and R/FR combinations on shoot growth of latitudinal ecotypes of B. pendula was studied using special diodes that emit monochromatic lights. When a 12 hrs PAR (110 µmol m-2 s-1) was extended with R, FR or R/FR ratios, lower intensities of monochromatic lights could not prevent growth cessation. At 25 µmol m-2 s-1, FR compared to R enhanced stem elongation in all ecotypes. This was due to the inhibitive effect of R on internode elongation. When day-length was extended by R/FR at various ratios, there was continuous shoot elongation, but was found to be declining with increasing ratios. The genetically transformed roots of red beet have been shown, for the first time, to produce very high levels of peroxidase (POD; EC 1.11.1.7) accounting for 1.21 x 10(6) Units L-1. Of the ten clones established using different strains of Agrobacterium rhizogenes, one was that from the strain LMG-150, three each from A 2/83, A 20/83 and A4. All the clones showed true integration of T-DNA when tested by PCR and Southern hybridization methods. The potential use of anoxygenic phototrophic bacteria for treating latex rubber sheet wastewater Electron. J. Biotechnol.

A total of 92 isolates of the purple non sulphur photosynthetic bacteria (PNSB) were isolated from 23 samples of wastewater obtained from rubber sheet manufacturing processes from various places of southern, Thailand. The isolate DK6 had the best potential for use in wastewater treatment as it can out-compete indigenous strains of PNSB when grown with them under conditions of microaerobic-light conditions. The isolate DK6 was identified as being most closely allied to Rhodopseudomonas blastica. The optimal pH and temperature for cell growth were between 6.5-7.5 and 30ºC, respectively. Continuous citric acid secretion by a high specific pH dependent active transport system in yeast Candida oleophila ATCC 20177 Electron. J. Biotechnol.

The pH influence on continuous citric acid secretion was investigated in Candida oleophila ATCC 20177 (var.) under NH4+ limiting state steady conditions, using glucose. Highest citric acid concentration of 57.8 g/l, citrate/isocitrate ratio of 15.6, space-time yield of 0.96 g/(l x hr) and biomass specific productivity of 0.041 g/(g x hr) were obtained at pH 5 and 60 hrs residence time. Only 22.8 g/l (39.4%) and a ratio of 9.9 were achieved at pH 6 pH and 12.4 g/l (21.5%) and a ratio of 3.7 at pH 3. Under non producing conditions, in excess of nitrogen, biomass concentration increased at raising pH. Among the diseases affecting the commercial citrus production, the citrus black spot (CBS) is considered to cause substantial losses. The analyses of particles in suspension in the orchards and collected into a disc have been applied as a preventive action trying to identify the presence of fungus spores before symptom appearance. In this paper, we show the results of several shape analysis methods applied to the fungus, the first step to the aimed computer aided vision system, capable to assist the identification process. We report the growth promoting potential of Piriformospora indica, which is a newly discovered arbuscular mycorrhiza-like fungus. It is a facultative symbiont and unlike arbuscular mycorrhizal fungi, it can be cultured in vitro. Adhatoda vasica is a medicinal plant. Rapid proliferation of roots was recorded in A. vasica withan important root colonization estimated to 95% after 6 months. P. indica improved growth of A. vasica. O objetivo deste trabalho foi estudar o efeito do ambiente protegido cultivado com melão (Cucumis melo L.) sobre os elementos meteorológicos e sua relação com as condições externas. O experimento foi conduzido no período de 5-10-2001 a 7-1-2002, em estufa plástica de 420 m², construída com estrutura metálica galvanizada, com dois vãos de altura central de 4,6 m e lateral de 3,0 m, composto de quatro janelas frontais, cobertas com filme de polietileno transparente de alta densidade, com aditivo ultravioleta e espessura de 150 µm. Desempenho de um aquecedor de água a biogás REA

Estudos foram desenvolvidos para dimensionar e adaptar o injetor do queimador principal de um aquecedor de água tipo acumulação de 75 L. O diâmetro do injetor foi redimensionado em função da pressão de serviço de 100 mm H2O e poder calorífico inferior do biogás de 21.600 kJ m-3 n, garantindo a manutenção da potência calorífica do equipamento de 20.900 kJ h-1. Os resultados demonstraram que o queimador adaptado operou com biogás adequadamente, com chama estável. A eficiência média do aquecedor foi de 68%, para ganho térmico de 36,7 ºC, correspondendo à temperatura final da água igual a 62,7 ºC, sendo consumido 0,796 m³n de biogás, aquecendo 75 L de água em 72 minutos.

Uso da fertirrigação e teores de macronutrientes em planta de pimentão REA

Este estudo teve o objetivo de determinar os teores de macronutrientes (N, P, K, Ca, Mg e S) nos diferentes órgãos do híbrido de pimentão Elisa, sob fertirrigação e ambiente protegido, por meio de coletas de plantas durante o ciclo. Ao final do experimento, obtiveram-se os teores dos macronutrientes no fruto, folha, caule, raiz e na planta como um todo, em cada etapa de crescimento. A fertilização no ciclo estudado, 140 dias, foi feita via irrigação localizada (gotejamento).
