Biophysical and biochemical properties of an unusual birnavirus pathogenic for rotifers ArchiMer

A cytoplasmic dsRNA virus, rotifer birnavirus (RBV), has recently been isolated from the rotifer Brachionus plicatilis and is associated with a high mortality rate. Histologically, the viral lesions consist of characteristic inclusions, particularly amorphous dense bodies containing occluded particles. Purified virions are about 59 nm in diameter, single-shelled and display four capsomers per edge. The purified virions have a buoyant density of 1.290 (full particles) and 1.250 (empty particles) in CsCI gradients. Four major structural polypeptides of Mrs 60K, 52K, 33K and 27K were detected by SDS-PAGE. Ultrastructural study of a parasite first reported in 1978 in Korea, in the oocytes of the Pacific oyster Crassostrea gigas then reported again in 1984 in Chungmu Bay oysters. The observed stages are typical of those of the Paramyxea Levine 1980 group. The development consists of the production of secondary cells within a stem cell; these secondary cells then producing one tertiary cell. The specific evolution of the sporogenesis leading to the production of one single tricellular spore per sporont distinguishes the studied parasite from other Paramyxea

Evolution de la mortalité des huîtres portugaises C. angulata dans le bassin de Marennes ArchiMer

A year after the end of the Gill disease which heavily struck the Portuguese oysters Cassostrea angulata Lmk. (Marteil, 1968), a new disease whose causes are still unexplained caused a massive mortality in the same species in the summer of 1970 (Comps, 1972). It can be considered, in 1973, that the successive losses lead to the quasi-extinction of the species C. angulata in the farms of the Marennes basin, since, contrary to what has been done in the case of the Malpèque disease in Canada, the indigenous specious was replaced by another species, the Pacific oyster Crassostrea gigas Thunberg.

Following the mass mortality of Ostrea edulis, some experiments were conducted with O. chilensis broodstock and spat coming from Chiloe Island (Chile). Concurrently, spat of th same species produced in a hatchery was cultivated. After a small period of growth, these oysters died during the summer. Microscopic observations showed Marteilia refringens and Bonamia ostreae in the oysters' tissue. For these reasons, Ostrea chilensis has not been retained to temporarily replace O. edulis.

From 1988 to 1989, a study has been conducted pertaining to the growth of the flat oyster Ostrea edulis, as well as Marteilia refringens and Bonamia ostreae outbreaks. Commercial mono specific farming (flat oysters), with low density or mixed (flat and hollow oysters) along with higher density, has been taking place in the Arcachon basin oceanographic sectors, where main hydrobiological parameters were acquired simultaneously. The excellent growth of the flat oyster was witnessed in mono specific farming. However this growth was average in mixed farming, probably due to higher total oyster density and interspecies competition. In the autumn of 1966, visible lesions were reported on the gills of Portuguese oysters raised in the Marennes-Oléron basin. Subsequently, various observers noted the presence of identical lesions on the oysters of Arcachon and the coasts of Brittany. A new oyster disease then appeared and with such an intensity that during the year 1967 the losses affected a large proportion of farmers' production. The disease's symptoms are visible, when first observed, on the gills and palpi. The gills show V-shaped indentations, rather deep and rather numerous, that give them a serrated look. The sporogony of A. nelsoni is of the apansporoblastic type. Diplocaryons always occur during merogony, never occur during sporogony. Sporoblasts are produced by fission of a multinucleate and elongated plasmodium. Spore morphogenesis begins before the end of the end of the sporoblasts individualization. The polaroplast has a very dense outer area. The genus Ameson is very much like genus Perezia and both are badly classified respectively in the families Nosematidae and Unikaryonidae. The creation of the new family Pereziidae is proposed for both genera.

Abnormal mortalities of flat oysters, Ostrea edulis, have been monitored in June 1979 in some farms of l'Ile Tudy. The examination of these oysters with light and electron microscopy showed different forms of an intracellular parasite localized in the connective tissue of the gills and the digestive gland. It is also detectable in the blood vessels and the stomachal epithelium. Numerous parasited oysters showed distinctive branchial damages (perforations, indentations) visible through macroscopic examination. The epidemiological monitoring carried out on the Norman and Breton coast revealed the presence of this parasite in a large number of flat oyster farms. Summer mortalities in the Pacific oyster in the Bay of Arcachon were analysed during four years (1981 to 1984). Mortalities occur mostly among one year old oyster and affect the whole bay, with different prevalence. The phenomenon occurs at certain times during the summer spawning period and is associated with high temperatures. A thinning of the digestive tubule epithelium is observed, but there is no evidence of infectious disease. The failure to find a causative agent, pollution or pathogen, tends to strengthen a physiological disorder hypothesis, related to environmental conditions. Two viral infections associated to some epizooties have been observed in Crassostrea angulata Lmk. The first one concerns the Gill disease in which the viral damages go hand in hand with a cellular hypertrophy, while, in the second one, the contaminated cells are associated with a deterioration of the connective tissue. These branchial necrosis associated to a virus have been observed again recently in some Portuguese oysters from the natural deposits of the Cadix (Spain) area. Depuis plusieurs années on a constaté une baisse sensible de la productivité dans l'exploitation des parcs et claires à huîtres du bassin de Marennes-Oléron. En outre l'huître portugaise (Crassostrea angulata Lmk) élevée dans cette zone ostréicole devait subir en 1966 et 1967 les effets de la maladie des branchies qui entraîna des pertes appréciables (Franc & Arvy, 1970). Au cours de l'été 1970, parallèlement à des perturbations dans la reproduction, s'amorça une mortalité massive qui au printemps 1971 affectait environ 70% du cheptel.
