The pollack Pollachius pollachius is a commercially important fish, considered as a candidate for aquaculture in France. Rearing attempts are promising (Suquet et al., 1996), and more information is required about its reproduction, to contribute to a better management of breeders for this species. This study was designed to examine the first maturity in pollack and the effects of conditions such as origin and delayed reproduction season, on gonad maturation, reproductive and growth performances of adults. |

The Nme gene family, also known as Nm23 or NDPK, is a very ancient gene family that can be found in all kingdoms of life. In the late eighties, a gene of the Nme family, NME1, was identified as the first metastatic suppressor gene, resulting in a major interest for this family. Due to the complexity of the family, the need for a unified and evolutionary-supported gene nomenclature was recently stressed by the scientific community. Based on a complete evolutionary history study of the gene family in metazoans and vertebrates, a unified nomenclature was recently proposed and accepted by gene nomenclature consortia. In addition to its well-documented role in tumor metastasis, members of the Nme family are also involved in a wide variety of cellular and... |

P>The present study aimed to establish some basic characteristics of Atlantic bluefin tuna sperm from captive mature males, treated or untreated by gonadotropin releasing hormones agonist (GnRHa). Intratesticular milt was collected from treated and untreated fish (mean weight +/- SD: 122.9 +/- 29.2 kg, n = 21). There was no significant effect of GnRHa treatment on GSI (1.33 +/- 0.70%, n = 21) or on sperm concentration (3.8 +/- 1.3 x 1010 spermatozoa ml-1, n = 21) estimated by optical density at 260 nm. Similarly, the percentage of motile spermatozoa measured at 30 s post activation: activating medium (AM seawater containing 10 mg ml-1 BSA) was not significantly different between control and GnRHa implanted males. On the other hand, a significantly... |

Retinitis pigmentosa 2 (RP2) gene is responsible for up to 20% of X-linked retinitis pigmentosa, a severe heterogeneous genetic disorder resulting in progressive retinal degeneration in humans. In vertebrates, several bodies of evidence have clearly established the role of Rp2 protein in cilia genesis and/or function. Unexpectedly, some observations in zebrafish have suggested the oocyte-predominant expression of the rp2 gene, a typical feature of maternal-effect genes. In the present study, we investigate the maternal inheritance of rp2 gene products in zebrafish eggs in order to address whether rp2 could be a novel maternal-effect gene required for normal development. Although both rp2 mRNA and corresponding protein are expressed during oogenesis, rp2... |

The aims of the present study were (a) to compare sperm quality (percentage of motile spermatozoa, motility duration, density and fertility after cryopreservation) between precocious and normally maturing male European sea bass Dicentrarchus labrax, (b) to examine the potential of human chorionic gonadotropin (hCG) to increase spermiation in precocious males and (c) to examine the potential of hCG to induce spermatogenesis and spermiation in non-precocious 1-year-old males. One hundred precocious and 100 non-precocious fish were each randomly divided in two groups each: control (precocious saline-treated and non precocious saline-treated) and treated (precocious hCG-treated and non precocious hCG-treated). Treated groups were administered weekly with 1000... |

In production operations the application of daily examination of each female of broodstock should be a determinant progress in hatchery management in terms of fry output. The eggs expelled shortly after ovulation by means of a systematic fivefold a week stripping attempt of females, presented a high viability rate (72%) compared to that of eggs collected twice a week (46%) whereas the treatments did not have any effect on female fecundity. Increase of the stripping rhythm brought the hatching rates and broodstock fertilities respectively from 11% to 35% and from 20,000 to 65,000 larvae/kg of spawning female. The enhancement of egg quality and of the resulting larva production is discussed. |

The deliverable (D7.1) of WP7 “Establishment of a Computer Assisted Sperm Analysis (CASA) for the evaluation of grey mullet sperm” has been submitted. Preliminary analyses show that mullet sperm motility pattern is not similar to that of sea bass (Dicentrarchus labrax) or rainbow trout (Oncorhynchus mykiss) in terms of velocity as well as motility duration (ca 1 min; Fig. 1a). Our preliminary results, indicate that the major bio-technical settings, dilution of sperm, quality of activation and mastering of video recordings have been determined and can now be applied to experimental protocols. Since fish sperm quality is usually assessed through subjective observation, in terms of motility classes and duration, statistical analysis becomes less precise. The... |

This paper presents some features of turbot sperm and compares them with data reported in the literature on other freshwater and marine fish. In captivity, turbot spermiation period lasts 6 months. When compared to other fish species, turbot males are characterized by low values of maximum gonadosomatic index (0.6-0.8%) and are poor sperm producers in terms of volume (0.2-2.2 ml), concentration (0.7-11.0 x 10(9) spermatozoa/ml) and total sperm number released per stripping (0.2-12.0 x 10(9) spermatozoa). Compared to most freshwater species, the duration of cell movement is long (1-17 min). The spermatozoon of turbot can be considered to be of a primitive type. A very high protein content (8.8 mg/ml) is found in the seminal fluid of turbot. Increasing... |

P>The objective of this study was to complement published hake (Merlucius merlucius) sperm motility characteristics by obtention of data more specifically dependent on flagella: beat frequency, flagellar wave amplitude, wave length, number of waves along the length of the flagellum and curvature of the wave. The changes of these parameters are described in relation to the time period elapsed since activation are described using high resolution video images of flagella while quantifying the progression during the swimming period from the activity initiation to full cessation of movement several minutes later. The main characteristic of the swimming period of hake spermatozoa shows a progressive decrease of forward displacement resulting from a cumulative... |

The fishing of pollack which concerns essentially, France, United Kingdom and Norway, and at a lower level Ireland and Denmark, is situated between 10 and 15000 tons a year. In 1996, France was the first producer of Pollack. Since 1980, a decrease of production is observed in Europe. In France, landing reached 3000t. in 1998 (Suquet, 2001). However, the demand remaining strong, aquaculture could be a solution to satisfy the deficit between the French production and the domestic demand. |

Background: The Nme family, previously known as Nm23 or NDPK, is involved in various molecular processes including tumor metastasis and some members of the family, but not all, exhibit a Nucleoside Diphosphate Kinase (NDPK) activity. Ten genes are known in humans, in which some members have been extensively studied. In non-mammalian species, the Nme protein family has received, in contrast, far less attention. The picture of the vertebrate Nme family remains thus incomplete and orthology relationships with mammalian counterparts were only partially characterized. The present study therefore aimed at characterizing the Nme gene repertoire in vertebrates with special interest for teleosts, and providing a comprehensive overview of the Nme gene family... |

Bluefin tuna shows adaptative physiology features allowing life in a wide range of physical and climatic parameters like temperature, salinity and hydrostatic pressure, hence the species fulfil the main basic requirements for progressive domestication and rearing in captivity. The high mortalities recorded after handling and the losses observed after abrupt variations of environment in the cages, emphasised the extreme sensitivity to stress of tuna. As consequences, the progresses about the control of tuna biological cycle are very slow due to limited handling possibilities and tuna aquaculture is restricted to fattening of wild fish. At this time, bluefin tuna production progresses without domestication of the species. |

The movement characteristics of Pacific oyster trochophores have received very little coverage in the scientific literature. Described here are not only changes in the swimming characteristics of Crassostrea gigas trochophores (size: 53-77 mu m) in relation to time after fertilization and to inter-female variation but also the effects of salinity and pH on movement characteristics. The percentage of motile trochophores was measured on images obtained through a dissecting microscope and the Velocity Average Path (VAP) was assessed using a Computer Assisted Sperm Analysis (CASA) system. At 20 degrees C, the first movements of the trochophores were observed at 6.5 h post-fertilization. The mean (+/- SD) percentage of swimming trochophores and the VAP observed... |

Egg production data from individual hand stripped female turbot were collected and analysed over several consecutive spawning seasons, on natural or shifted conditions. Egg quantity, egg viability and number of spawns were examined from one spawning season to the other, and inside the spawning season. These parameters seemed to increase from a year to the next one, except for the oldest fishes. Spawns were spread over a three-month period and occurred at the same time in the period in successive seasons for most females. Egg production seemed related to the feeding level and the earliness of spawning season appeared depended on the growth rate. Without sand substrate in tanks, injuries could increase therefore no significant differences were observed in... |

Since the first work of Blaxter in 1953, fish sperm cryopreservation has been attempted on about 30 marine species. The present paper reviews the techniques used and the results published in these species. Particular attention is paid to the handling procedure of sperm before freezing, the problems of semen ageing and semen contamination with urine. The quality of frozen-thawed semen was evaluated using previously standardized biotests, such as a two-step motility activation technique adapted for the different species and fertilization assays using a discriminating insemination technique. Most extenders used in marine fish are saline or sugar solutions. From the investigated cryoprotectants, dimethyl sulphoxide (DMSO) generally leads to the best results.... |
