A preliminary genetic linkage map for the European flat oyster Ostrea edulis L. ArchiMer
Lallias, Delphine; Lapegue, Sylvie; Harley, Crhis; Boudry, Pierre; Heurtebise, Serge; Beaumont, Andy.
The flat oyster Ostrea edulis is the species endemic to European coasts, both Atlantic and Mediterranean. It has been exploited since Roman times, but its aquaculture production decreased from around 20000 tons in the 1950's to 1500 tons today because of two successive diseases due to the intracellular parasites Marteilia refringens and Bonamia ostreae. Since 1985, Ifremer has been undertaking a breeding program to produce families of oysters tolerant to Bonamia. In this context, a further step would be to identify QTLs for resistance to this parasite. Therefore, as a first step a genetic map is now being built. The establishment of a genetic linkage map will provide a foundation for the mapping of QTLs, with the ultimate objective to implement...

Fragmentation des populations naturelles d'Ostrea edulis: une adaptation locale de l'huître plate européenne ArchiMer
Harrang, Estelle; Bierne, Nicolas; Heurtebise, Serge; Morga, Benjamin; Lapegue, Sylvie.

Dardignac, Marie-jose; Bodoy, Alain; Garnier, Jacqueline; Heurtebise, Serge; Legrand, Jacqueline.

Can survival of European flat oysters following experimental infection with Bonamia ostreae be predicted using QTLs? ArchiMer
Harrang, Estelle; Heurtebise, Serge; Faury, Nicole; Robert, Maeva; Arzul, Isabelle; Lapegue, Sylvie.
The present study identifies quantitative trait loci (QTLs) in response to an experimental infection with the parasite responsible for bonamiosis, Bonamia ostreae, in two segregating families of the European flat oyster, Ostrea edulis. We first constructed a genetic-linkage map for each studied family and improved the existing genetic-linkage map for the European flat oyster with a set of SNP markers. This latter map now combines the best accuracy and the best estimate of the genome coverage available for an oyster species. Secondly, by comparing the QTLs detected in this study with those previously published for O. edulis in similar experimental conditions, we identified several potential QTLs that were identical between the different families, and also...

Individual growth variation and its relationship with survival in juvenile Pacific oysters, Crassostrea gigas (Thunberg) ArchiMer
Boudry, Pierre; Collet, B; Mccombie, Helen; Ernande, Bruno; Morand, B; Heurtebise, Serge; Gerard, Andre.
In order to study individual growth variability and its relationship with survival in juvenile Crassostrea gigas, parental oysters were sampled at four sites located along the French Atlantic coast and bred under controlled hatchery conditions. Four groups of larvae were obtained by crossing five males and five females from each of the four sites, and a fifth group by crossing these 20 males and 20 females together in a pool. Larvae were reared under conditions allowing the maintenance of a maximum variability of size and gave five experimental groups. Oysters were individually monitored for growth and survival from 3 to 10 months after fertilization. The individual growth performances were relatively stable over time and no noticeable compensation for...

Performance of triploid Pacific oysters Crassostrea gigas (Thunberg) reared in high carrying capacity ecosystem : survival, growth and proximate biochemical composition ArchiMer
Goulletquer, Philippe; Joly, Jean-pierre; Gerard, Andre; Le Gagneur, Eric; Moriceau, Jacques; Peignon, Jean-marie; Heurtebise, Serge; Phelipot, Pascal.
Triploid oysters Crassllstrea gigas were: produced in 1990 by treating fertilized eggs with cytochalasin B (CS). Triploids, treated diploids, and controls were deployed early 1991 in a high carrying capacity ecosystem on the Eastern Coast of Normandy (France). A monthly monitoring in 1992 showed that triploids yielded significantly higher growth rate and biochemical composition. However, growth was more heterogeneous. No evidence was found for a length or weight bimodal frequency distribution within triploid groups after a 26 months rearing cycle. Triploids showed retardation of gametogenesis. Carbohydrates content in triploids remained almost constant (40%) from June to September. Their survival rates were significantly lower than controls. Moreover...

Genetic basis of growth, survival and their plasticity in the Pacific oyster Crassostrea gigas ArchiMer
Ernande, Bruno; Boudry, Pierre; Heurtebise, Serge; Haure, Joel; Martin, Jean-louis.
Introduction : Growth and survival are the life-history traits of the most interest for oyster production since they determine biomass output (growth x survival). This study aimed to investigate whether there is genetic basis for growth, survival and their plasticity to know if a selection program is conceivable for these traits. Material and Methods : Nested half-sibs mating design was used to produce families of oysters: 5 males were each mated to three females producing 5 half-sibs families, each containing 3 full-sib families. At 15 months of age (May 1999), each full-sibs family was randomly split into five groups which were given different treatments. Two groups were placed in a constant environment: one with low food resources and another with high...

Is fertility of hybrids enough to conclude that the two oysters Crassostrea gigas and Crassostrea angulata are the same species? ArchiMer
Huvet, Arnaud; Gerard, Andre; Ledu, Christophe; Phelipot, Pascal; Heurtebise, Serge; Boudry, Pierre.
The distinction of the two cupped oysters Crassostrea gigas (Thunberg, 1793) and Crassostrea angulata (Lamark, 1819) into two species was chiefly due to their differing geographical distributions, C. gigas being present in Asia and C. angulata in Europe. Today it is commonly accepted that C. angulata and C. gigas are a single species according to morphological, genetic and F1 hybridization data. However, the demonstration of the fertility of their hybrids and the absence of any reproductive isolation remained to be investigated. Consequently, we studied the fertility of hybrids and sperm competition by performing three different experiments and producing G1 and G2 hybrid progenies between wild populations of C. angulata and C. gigas. Progenies showed very...

Mapping QTL for resistance to summer mortality in the Pacific oyster Crassostrea gigas ArchiMer
Sauvage, Christopher; Heurtebise, Serge; De Koning, D.; Haley, Chris; Boudry, Pierre; Lapegue, Sylvie.
Summer mortalities of the Pacific cupped oyster Crassostrea gigas have been described in the literature for decades. One solution to overcome this problem would be to select animals more tolerant to summer mortalities, as this trait was shown to be highly heritable (Degrémont, 2003). Such selective breeding would strongly benefit from the identification of markers associated to the resistance to allow the development of markers assisted selection. Recently, a set of a new kinds of markers (SNPs) have been developed in the Pacific oyster. These markers were identified and characterized in several genes involved in the immune response and energetic pathways (Sauvage, 2007). In this context, the mapping of SNPs, of 16 new EST-SSR based markers and of...

Bonamia ostreae-induced mortalities in one-year old European flat oysters Ostrea edulis: experimental infection by cohabitation challenge ArchiMer
Lallias, Delphine; Arzul, Isabelle; Heurtebise, Serge; Ferrand, Sylvie; Chollet, Bruno; Robert, Maeva; Beaumon, Andy; Boudry, Pierre; Morga, Benjamin; Lapegue, Sylvie.
Bonamiosis is a parasitic disease (causative agent: Bonamia ostreae) affecting the European flat oyster Ostrea edulis, responsible for a drastic decline in its aquaculture production. Selective breeding programs for resistance to bonamiosis have been undertaken to counter this disease. In the present study, a 6-month cohabitation challenge experiment was performed in order to transmit the disease from wild oysters injected with the parasite to two tested families of oysters (20 and 8-month old at the beginning of the experiment, with different genetic backgrounds) originating from a selective breeding program developed by IFREMER in France. Mortalities were checked daily and ventricular heart smears were performed on dying or moribund oysters to detect the...

Is the Portuguese oyster Crassostrea angulata in Southern Europe endangered by the expansion of the Pacific oyster C. gigas? ArchiMer
Batista, Frederico; Boudry, Pierre; Lapegue, Sylvie; Heurtebise, Serge; Monteiro, C. C..
The Portuguese oyster, Crassostrea angulata, was introduced from Portugal to the French Atlantic Coast in the 1860s. C. angulata quickly settled and expanded and leaded to the development of a new aquacultural industry in France. In the late 1960s, mortality associated with the detection of an iridovirus, led to the wipe out of C. angulata from French Atlantic waters and to the massive introduction of C. gigas to sustain production. In Southern Europe, similar symptoms were also observed in natural stocks of C. angulata from Sado River (Portugal) and from the area of Cadiz (Spain). Nowadays, only very few «pure» populations of C. angulata remain in southern Europe. These populations are potentially endangered by the current expansion of C. gigas...

Facteurs contrôlant la croissance de Ruditapes philippinarum dans 4 nurseries de production en Charente-Maritime (France) ArchiMer
Heral, Maurice; Deslous-paoli, Jean-marc; Garnier, Jacqueline; Prioul, Daniel; Heurtebise, Serge; Razet, Daniel.
Different types of pregrowth of the Japanese palourde are compared on the some site. The method by upwelling which is the more common used, to have good growth, requires a compromise between the running and the raised biomass. Two periods are critical, in the summer the growth of mollusks is stopped because of a decrease in the phytoplanktonic production ; in the winter, a fall in temperature lessens filtration and stops growth. The results concern clams consummatation rates of particular food (chlorophyll, protids, lipids, glucids) as weIl as the way to supply the decreasing growth : increase of phytoplankyonic production in the summer by recycling waters from nurseries that are loaded with ammoniac and raise winter temperatures by using under-ground...
