Field and mesocosm trials on passive sampling for the study of adsorption and desorption behaviour of lipophilic toxins with a focus on OA and DTX1 ArchiMer

It has been demonstrated that polymeric resins can be used as receiving phase in passive samplers designed for the detection of lipophilic marine toxins at sea and was referred to as solid phase adsorption toxin tracking (SPATT). The present study describes the uptake and desorption behaviour of the lipophilic marine toxins okadaic acid (OA) and dinophysistoxin-1 (DTX1) from Prorocentrum lima cultures by five styrene-divinylbenzene based polymeric resins Sepabeads (R) SP850, Sepabeads (R) SP825L, Amberlite (R) XAD4, Dowex (R) Optipore (R) L-493 and Diaion (R) HP-20. Le littoral français est touché tous les ans par des proliférations d'algues toxiques dont l'ampleur géographique et la durée sont très variables selon les genres/espèces et les années. Par exemple, à partir du mois de mai, voire fin avril certaines années, les côtes sud de la Bretagne et celles des Pays de la Loire sont touchées par des proliférations de Dinophysis spp, genre responsable des intoxications diarrhéiques et cause la plus fréquente des fermetures de zones conchylicoles. Etude de la décontamination de moules toxiques (toxines diarrhéiques) en laboratoire et en milieu naturel ArchiMer

Two studies of depuration, one in laboratory conditions and the other one in an oyster's pond were performed using 2 sets of contaminated mussels with diarrhetic toxins at a high and a low level (3 and 1 M.U.). Depuration kinetics were followed both by the adult mouse test and okadaïc acid analysis. Results are - in the pond, high toxic mussels need about 20 days to become eatable (toxicity level : 0.5 M.U.g-1 digestive glands), -this time decreases when the initial toxicity is low but it increases when mussels are underfed, - a good correlation is observed between the total toxicity and okadaïc acid concentration.

Depuis janvier 1993, des épisodes de forte toxicité des coquillages ont été observés lors des contrôles de routine en différents points des côtes françaises. Les souris ayant reçu des extraits de coquillages, par voie intrapéritonéale, présentaient des symptômes de type neurologique et mouraient au bout de 5 à 15 minutes. Cette toxicité n'a pu être attribuée à l'existence de phycotoxines connues. Blooms of Dinophysis in French coastal waters are implicated in most bans on marketing commercial bivalves. However, the relation between Dinophysis cell density and shellfish toxicity is not always consistent. Discrepancies may be due to the simple fact that it is nearly impossible to compare an integral over a few days (shellfish toxin content) and water samples. A bloom of the toxic dinoflagellate Alexandrium tamarense was observed for the first time in Thau iagoon between November and December 1998. Maximum cell concentrations detected in shellfish farming areas (85 000 Cells/1) were high enough to make mussels and clams unsuitable for human consumption (max. 850 ug. STX eq.100 g'1) while toxic levels in oysters did not exceed, or even reach, the international public health threshold established for PSP toxins. These toxins, produced by a few phytoplankton species which regularly occur in sea water, accumulate in shellfish, which become toxic. Mise au point d'un test microcrustacé marin ArchiMer

Because of the low number of currently available toxicity tests for marine environment, the authors describe an experimental procedure using a marine copepod : Tigriopus brevicornis. This method can be used to quantify the 50 % inhibition of larval production or growth. The variability in controls larval production reaches about 9 %. The toxicity of two contaminants : tributyltin (TBTF) and cadmium chloride (CdCl2) was determined with this method and the 50 % decreases in larval production are respectively 1,4 µg/l (TBTF) and 61 µg/l (Cd++).

The toxins of marine microalgae ArchiMer

Exotoxins exert direct deleterious effects on flora and fauna: Some of them damage the cellular membrane, others have inhibiting effects on phytoplanktonic populations susceptible to coming into competition. In terms of pollution, exotoxins are especially a significant menace for aquaculture and although some are also bothersome for Man, we will not treat them here. Endotoxins accumulate in the food chain and reach humans via consumed sea products. They constitute a potential risk for human health and a scourge for the shellfish farming economy.

Etude de la dégradation et de la toxicité de la roténone en milieu marin ArchiMer

The use of rotenone as a means of predators eradication is necessary for the success of shrimps culture. Because these compound is a fish toxicant, some precautions have to be tacken in order to avoid the contamination of marine environment by the residual waters ; so the dissipation and the toxicity have been studied together in laboratory and in field assays. It appears that a delay of 15 days should be respected : this time is required for a total disappearing of the residual toxicity.

ISSR as new markers for genetic characterization and evaluation of relationships among phytoplankton ArchiMer

In order to increase the molecular tools and markers needed for the identification of phytoplankton species, the inter simple sequence repeat ( ISSR) fingerprinting was adapted to micro-algae and its use in genetic analysis was demonstrated. Twelve strains, 6 Alexandrium, 4 Pseudo-nitzschia, 1 Skeletonema and 1 Tetraselmis were analysed for the first time with ISSR amplifications. The patterns were highly polymorphic and very reproducible. The 6 primers gave 223 polymorphic markers that clearly and easily distinguished all 12 strains ( mainly toxic ones) and gave 187 polymorphic markers among the Alexandrium and the Pseudo-nitzschia species. Les intoxications humaines résultant de la consommation de coquillages sont connues en France depuis plusieurs siècles. On peut supposer que la cause principale a d'abord été d'origine bactériologique, eu égard aux conditions de transport et à l'absence de contrôles de salubrité dans le passé. Quoi qu'il en soit, il faut compter aujourd'hui avec une autre origine, à savoir les proliférations de certaines algues unicellulaires planctoniques (phytoplancton), qui causent dans notre pays jusqu'à plusieurs milliers d'intoxications par an. Soulignons que la France a été jusqu'ici largement épargnée, puisque d'autres pays comptabilisent des dizaines ou centaines de décès depuis le début du siècle. Azadinium spinosum (Elbrächter and Tillmann), a small marine dinoflagellate, has been recently described as a de novo producer of azaspiracid-1 and -2 (AZA1 and -2) diarrhoeic toxins. A culture of A. spinosum was established in our laboratory and optimised for pilot-scale production of this organism, to evaluate and understand AZA1 and -2 accumulation and biotransformation in blue mussels (Mytilus edulis) fed with A. spinosum. Adult mussels were continuously exposed to A. spinosum over 1 week in 160 L cylindrical conical tanks. A cytodetection test of diarrhoeic phycotoxins was developped by both IFREMER and Nantes University. Its principle is based on cell morphological changes of KB strain, induced by inhibition of protein-phosphatases involved in cytoskeleton building. This test has been shown sensitive, fast and specific for protein phosphatase inhibitors like diarrhoeic toxins (specialy okadaic acid in France).
