Bacterial polysaccharides offer fascinating potential applications for the pharmaceutical industry. Although many known marine bacteria produce exopolysaccharides (EPS), continuation in looking for new polysaccharide-producing micro-organisms is promising. Marine bacteria, isolated from deep-sea hydrothermal vents, have demonstrated their ability to produce in aerobic conditions, unusual EPS. With the aim of discovering biological activities, EPS presenting different structural features were studied. Alteromonas infernus bacterium isolated from deep-sea hydrothermal vents can produce by fermentation a high molecular weight exopolysaccharide (EPS) called GY785. This EPS described as a new source of glycosaminoglycan-like molecule presents a great potential for pharmaceutical and biotechnological applications. However, this unusual EPS is secreted by a Gram-negative bacterium and can be therefore contaminated by endotoxins, in particular the lipopolysaccharides (LPS). An hydrobiological monitoring survey has been carried out in the Bay of Marennes-Oléron since 1977. Five stations, distributed over the Bay, were sam pied twice a month. Spatial and temporal trends of the physical characteristics (i.e., seawater, temperature, salinity, dissolved oxygen and suspended matter), nutrients (azote, nitrite, nitrate, silicate) and organic variables (chlorophyll a and pheopigments) were assessed by using a statistical time-series analysis mode\. Anti-Metastatic Properties of a Marine Bacterial Exopolysaccharide-Based Derivative Designed to Mimic Glycosaminoglycans ArchiMer

Osteosarcoma is the most frequent malignant primary bone tumor characterized by a high potency to form lung metastases. In this study, the effect of three oversulfated low molecular weight marine bacterial exopolysaccharides (OS-EPS) with different molecular weights (4, 8 and 15 kDa) were first evaluated in vitro on human and murine osteosarcoma cell lines. Different biological activities were studied: cell proliferation, cell adhesion and migration, matrix metalloproteinase expression. This in vitro study showed that only the OS-EPS 15 kDa derivative could inhibit the invasiveness of osteosarcoma cells with an inhibition rate close to 90%. Structural data on a bacterial exopolysaccharide produced by a deep-sea Alteromonas macleodii strain ArchiMer

Some marine bacteria collected around deep-sea hydrothermal vents are able to produce, in laboratory conditions, complex and innovative exopolysaccharides. In a previous study, the mesophilic strain Alteromonas macleodii subsp. fijiensis biovar deepsane was collected on the East Pacific Rise at 2600m depth. It was isolated from a polychaete annelid Alvinella pompejana and is able to synthesise and excrete the exopolysaccharide deepsane. La reproduction des coquillages a été soumise à des conditions climatiques exceptionnelles amplifiées par un hiver doux et sec. Concernant la reproduction des moules la formation des gonades s'est normalement effectuée vers le mois de février. Dans l'ensemble, le captage a été correct et la croissance des jeunes moules rapide malgré la sécheresse. La gamétogénèse des huîtres creuses a débuté très tôt. La reproduction s'est effectuée dans des conditions climatiques et de milieu peu habituels.

Biopolymers produced by marine organisms can offer useful tools for regenerative medicine. Particularly, HE800 exopolysaccharide (HE800 EPS) secreted by a deep-sea hydrothermal bacterium displays an interesting glycosaminoglycan-like feature resembling hyaluronan. Previous studies demonstrated its effectiveness to enhance in vivo bone regeneration and to support osteoblastic cell metabolism in culture. Thus, in order to assess the usefulness of this high-molecular weight polymer in tissue engineering and tissue repair, in vitro reconstructed connective tissues containing HE800 EPS were performed. Les conditions météorologiques semblaient cette année à priori favorables à la reproduction des moules. Cependant, le maturation des gonades a été très lente. Ainsi, les premières pontes, de faible importance, ont eu lieu début Avril. Plusieurs émissions tardives sont apparues fin Mai et se sont poursuivies jusqu'au début du mois de Juillet. Elles n'ont pas été importantes et ont conduit à une faible quantité de larves au stade fixation. Un captage "moyen" a été observé début Mai. Ensuite, la présence de quelques larves grosses n'a pas permis un recrutement suffisant. Les agriculteurs aménageant quelques centaines d'hectares du marais doux de Hiers Brouage (mise à plat, drainage) afin de le mettre en culture, il était important de pouvoir mesurer l'impact que peuvent avoir les rejets de drainage-pompage dans le milieu marin par l'intermédiaire des chenaux ostréicoles. Ceci afin d'éviter d'éventuels conflits entre groupes socio-professionnels (agriculteurs et conchyliculteurs) en proposant des mesures et de solutions. La décontamination bactériologique des huitres Crassostrea gigas a été étudiée expérimentalement dans des bassins dégorgeoirs de 13 m3 de conception proche de la configuration normale d'une station de purification française. Les huîtres contaminées artificiellement ont été placées dans des conditions dites "naturelles" d'épuration. Il n'a pas été utilisé de procédé physique ou chimique de purification de l'eau. Bilan de la surveillance microbiologique du bassin de Marennes-Oléron et de la rive droite de la Gironde (Avril 1989 - Décembre 1992) ArchiMer

La surveillance bactériologique de 1989 à 1992 du bassin de Marennes-Oléron et de la rive droite de la Gironde met en évidence la bonne qualité du milieu. Les dépassements de la norme (>300 coliformes fécaux/100 g de chair et liquide intervalvaire) sont peu fréquents. Quelques pollutions sporadiques peuvent apparaître, le plus souvent en été, et sont liées au dysfonctionnement passager des systèmes d'assainissement (postes de relèvement, stations d'épuration) ou aux infiltrations d'eaux usées dans les réseaux pluviaux. L'application de la directive européenne de 1991 aurait une incidence limitée sur le classement des zones de production conchylicoles du bassin de Marennes-Oléron.

The germs decreasing of artificially contaminated oysters Crassostrea gigas has been studied in experimental tanks similar to a shellfish depuration plant, without any chemical or physical water treatment. The oyster were treated by three different water management pattern, compared to a control tank and the germs decreasing studied by MPN method of fecal Coliforms and Streptococus. As a result: 1) it appaears somewhat difficult to conclude in this first attempt about the most efficient treatment. 2) the most important factor in shellfish depuration working is obviously the physiological activity (and stage) of the animals.

Free-radical depolymerization with metallic catalysts has been investigated to depolymerize an exopolysaccharide (EPS), with a high molecular weight (> 10(6) g/Mol), produced by the bacterium Alteromonas macleodii subsp. fijiensis biovar deepsane. Three metals (copper, zinc and manganese) were used either alone or mixed. These metals led to the depolymerization of the high molecular weight bacterial polysaccharide with different efficiency.
