P>The objective of this study was to complement published hake (Merlucius merlucius) sperm motility characteristics by obtention of data more specifically dependent on flagella: beat frequency, flagellar wave amplitude, wave length, number of waves along the length of the flagellum and curvature of the wave. The changes of these parameters are described in relation to the time period elapsed since activation are described using high resolution video images of flagella while quantifying the progression during the swimming period from the activity initiation to full cessation of movement several minutes later. The main characteristic of the swimming period of hake spermatozoa shows a progressive decrease of forward displacement resulting from a cumulative... |

High quality of sperm is essential to a high fertilization rate, especially post- cryopreservation. Assessment of sperm integrity, motility and energy reserves before cryopreservation is necessary for selection of milt with optimal fertilizing potential. We describe the effect of cryopreservation on the quality of black-lip pearl oyster, Pinctada margaritifera var. cumingii sperm. Evaluated quality indices of fresh and frozen/thawed P. margaritifera spermatozoa, included morphology, ultrastructure and motility characteristics relative to the energy content (ATP) and its capacity to be sustained by mitochondrial respiration. Morphology and ultrastructure were quantitatively evaluated using images obtained by optical microscopy assisted by the Image J... |

Plastics are persistent synthetic polymers that accumulate as waste in the marine environment. Microplastic (MP) particles are derived from the breakdown of larger debris or can enter the environment as microscopic fragments. Because filter-feeder organisms ingest MP while feeding, they are likely to be impacted by MP pollution. To assess the impact of polystyrene microspheres (micro-PS) on the physiology of the Pacific oyster, adult oysters were experimentally exposed to virgin micro-PS (2 and 6 µm in diameter; 0.023 mg·L−1) for 2 mo during a reproductive cycle. Effects were investigated on ecophysiological parameters; cellular, transcriptomic, and proteomic responses; fecundity; and offspring development. Oysters preferentially ingested the 6-µm micro-PS... |

The movement characteristics of Pacific oyster trochophores have received very little coverage in the scientific literature. Described here are not only changes in the swimming characteristics of Crassostrea gigas trochophores (size: 53-77 mu m) in relation to time after fertilization and to inter-female variation but also the effects of salinity and pH on movement characteristics. The percentage of motile trochophores was measured on images obtained through a dissecting microscope and the Velocity Average Path (VAP) was assessed using a Computer Assisted Sperm Analysis (CASA) system. At 20 degrees C, the first movements of the trochophores were observed at 6.5 h post-fertilization. The mean (+/- SD) percentage of swimming trochophores and the VAP observed... |

Harmful algal blooms are a threat to aquatic organisms and coastal ecosystems. Among harmful species, the widespread distributed genus Alexandrium is of global importance. This genus is well-known for the synthesis of paralytic shellfish toxins which are toxic for humans through the consumption of contaminated shellfish. While the effects of Alexandrium species upon the physiology of bivalves are now well documented, consequences on reproduction remain poorly studied. In France, Alexandrium minutum blooms have been recurrent for the last decades, generally appearing during the reproduction season of most bivalves including the oyster Crassostrea gigas. These blooms could not only affect gametogenesis but also spawning, larval development or juvenile... |

This study assessed the effects of an artificial bloom of the toxin-producing dinoflagellate, Alexandrium minutum, upon nutrition related processes and reproductive output of the Pacific oyster, Crassostrea gigas. Oysters were exposed to A. minutum, Paralytic Shellfish Toxins (PST) producer and compared to a control batch of oysters fed Isochrysis galbana clone Tahitian (T.Iso). The experiment was performed in June 2008, when oysters were found ripe. Several physiological variables of oysters, such as PSTs accumulation, digestive gland and histological observations as well as spermatozoa quality, were measured at the end of the exposure. Results indicate that the digestive gland was greatly impacted upon A. minutum exposure. Monoacylglycerol and... |

Because of its rapid growth, its good reputation and the promising aquaculture experiences conducted in Norway and Scotland, cod is considered as a good candidate for cold water aquaculture. The aim of this paper is to present the work carried out in France since 1999: the selection of cod as a promising candidate for aquaculture and the assessment of its rearing performances in this environment. |

While the detection and quantification of nano-sized plastic in the environment remains a challenge, the growing number of polymer applications mean that we can expect an increase in the release of nanoplastics into the environment by indirect outputs. Today, very little is known about the impact of nano-sized plastics on marine organisms. Thus, the objective of this study was to investigate the toxicity of polystyrene nanoplastics (NPs) on oyster (Crassostrea gigas) gametes. Spermatozoa and oocytes were exposed to four NPs concentrations ranging from 0.1 to 100 mg L−1 for 1, 3 and 5 h. NPs coated with carboxylic (PS-COOH) and amine groups (PS-NH2) were used to determine how surface properties influence the effects of nanoplastics. Results demonstrated the... |

Investigating the roles of chemical factors stimulating and inhibiting sperm motility is required to understand the mechanisms of spermatozoa movement. In this study, we described the composition of the seminal fluid (osmotic pressure, pH, and ions) and investigated the roles of these factors and salinity in initiating spermatozoa movement in the Pacific oyster. The acidic pH of the gonad (5.82 ± 0.22) maintained sperm in the quiescent stage and initiation of flagellar movement was triggered by a sudden increase of spermatozoa external pH (pHe) when released in seawater (SW). At pH 6.4, percentage of motile spermatozoa was three times higher when they were activated in SW containing 30 mM NH4Cl, which alkalinizes internal pH (pHi) of spermatozoa, compared... |

The king scallop Pecten maximus is a high valuable species of great interest in Europe for both fishery and aquaculture. Notably, there has been an increased investment to produce seed for enhancement programmes of wild scallop populations. However, hatchery production is a relatively new industry and it is still underdeveloped. Major hurdles are spawning control and gamete quality. In the present study, a total of 14 scallops were sampled in the bay of Brest (Brittany, France) to compare transcriptomic profiles of mature oocytes collected by spawning induction or by stripping. To reach such a goal, a microarray analysis was performed by using a custom 8x60K oligonucleotide microarray representing 45,488 unique scallop contigs. First we identified genes... |

Egg production data from individual hand stripped female turbot were collected and analysed over several consecutive spawning seasons, on natural or shifted conditions. Egg quantity, egg viability and number of spawns were examined from one spawning season to the other, and inside the spawning season. These parameters seemed to increase from a year to the next one, except for the oldest fishes. Spawns were spread over a three-month period and occurred at the same time in the period in successive seasons for most females. Egg production seemed related to the feeding level and the earliness of spawning season appeared depended on the growth rate. Without sand substrate in tanks, injuries could increase therefore no significant differences were observed in... |

The toxic dinoflagellate Alexandrium catenella recurrently blooms on the coasts of France and produces Paralytic Shellfish Toxins (PSTs) that accumulate in bivalves. These toxins can affect various physiological functions including reproduction. The present study aims to validate measurements of sperm viability, DNA content and mitochondrial membrane potential in Pacific oyster Crassostrea gigas using flow cytometry coupled with fluorescent markers, and to use these measurements to assess the cellular parameters of sperm from Pacific oysters exposed to A. catenella. These parameters may influence fertilization, embryogenesis and larval development in free-spawning shellfish. Sperm viability and DNA content estimation were assessed using SYBR-14, which only... |

Bluefin tuna shows adaptative physiology features allowing life in a wide range of physical and climatic parameters like temperature, salinity and hydrostatic pressure, hence the species fulfil the main basic requirements for progressive domestication and rearing in captivity. The high mortalities recorded after handling and the losses observed after abrupt variations of environment in the cages, emphasised the extreme sensitivity to stress of tuna. As consequences, the progresses about the control of tuna biological cycle are very slow due to limited handling possibilities and tuna aquaculture is restricted to fattening of wild fish. At this time, bluefin tuna production progresses without domestication of the species. |

Pacific oyster Crassostrea gigas is one major species for aquaculture, and the development of breeding programs and the need for preservation of wild stock genetic resources prompted the need for larvae cryopreservation. The objective of the present study was to choose the most reliable protocol from several existing publications, to test its biological and practical limitations, and to adapt it to hatchery conditions. The selected protocol was characterized by a very slow freezing rate without seeding, and by the use of ethylene glycol and sucrose as cryoprotectant. The best survivals after thawing and rearing up to 48 h post fertilization (hpf) were obtained with larvae that were frozen at late trochophore (20 hpf) and early-D (24 hpf) stages. Increasing... |

MR imaging is the most appropriate non-invasive technique for quantifying the growth of somatic and gonad tissues and to determine sex in the Pacific oyster, Crassostrea gigas. However, this technique is too costly for field studies where oysters are used as bioindicators of environmental quality or to be applied routinely in hatcheries. We have tested the ability of low Nuclear Magnetic Resonance relaxometry, a much less expensive technique, to obtain phenotype parameters that can be used to monitor the physiological state of oysters. NMR measurements were carried out at three different periods using a low field spectrometer equipped with a 50 mm diameter probe to investigate 60 oysters in their first year of maturity, which were then dissected to measure... |

The potential of wreckfish (Polyprion americanus) to breed in captivity was examined. Ovarian activity varied seasonally, with vitellogenesis beginning in November and ending in May. Slow-release implant with gonadotropin-releasing hormone agonist (GnRHa) prevented atresia, and both GnRHa injections or implants at the end of vitellogenesis triggered ovulation after 7 days. Spontaneous spawning was obtained only in one female, and the few eggs produced were of poor viability. Manual stripping and artificial fertilization produced viable gametes and embryos. Therefore, present captivity conditions inhibited final oocyte maturation (FOM) and spawning, but not vitellogenesis and spermatogenesis. GnRHa treatments stimulate FOM and ovulation of viable eggs. |

Fertilization of the oocyte is a critical process of sexual reproduction depending among other factors on spermatozoa functionality. Mitochondria participate in many crucial processes for spermatozoa motility and fertilizing ability. The Pacific oyster, Crassostrea gigas, is characterized by a high inter-individual variability of its reproductive success which has been shown to be partially due to the variability of gamete quality. The present study explored spermatozoa characteristics such as: i) relationships among mitochondrial functionality (ATP content, mitochondrial membrane potential (MMP), reactive oxygen species production); sperm viability (percentages of live, dying, and dead); and motility; and ii) their involvement in inter-oyster variability... |
