Background and purpose: Although the activities of polysaccharides such as beta-glucans are well known in Agaricus brasiliensis, there has been little research on their exopolysaccharides and its characterization or scale up studies. The aim of the study was to study the monosaccharide composition of the exopolysaccharide (EPS) produced by Agaricus brasiliensis LPB3 and its production in a stirred tank fermenter. Materials and methods: Agaricus brasiliensis LPB3 was cultured in submerged fermentation in a medium containing glucose, yeast extract, K2HPO4 and MgSO4. Extracellular polysaccharide was precipitated with four volumes of 95% ethanol. For monosaccharide composition, the EPSs were hydrolyzed, derivatized to alditol acetates, and analyzed by gas... |

There is an urgent need for improvements in animal production, particularly for ruminants, such that more sustainable and efficient processes are developed for obtaining more nutritious and efficient feeds. Filamentous fungi can add value to residual plant biomass, and may also have the potential to produce metabolites and enrich plant biomasses used in animal nutrition, converting them into nutraceutical sources. Thus, in this work, filamentous fungal fermentation of ruminant feed biomasses commonly used in Brazil was performed, and the enrichment for bioactive metabolites was tested. For this, Fistulina hepatica, Ganoderma lucidum, Pleurotus pulmonarius, Panus lecomtei, and Aspergillus terreus were grown for 28 days on different substrates: starchy... |

ABSTRACT Ustilago maydis is a basidiomycete known as the causative agent of 'common smut', worldwide disease of maize that is recognized by the galls it forms, which have considerable potential as a gourmet food. Results of infection are quite variable, even under optimal greenhouse conditions. In order to find pathogenic strains able to be used as a highly infective and stable inoculum for the successful production of galls either in greenhouses or in the field, ears with gall symptoms containing teliospores were recovered from maize plants. The teliospores were suspended in water and plated on nutrient-rich medium. Twenty-six colonies developed, containing three types of yeast-like colonies: saprotrophic, pathogenic, and solopathogenic. DAPI staining... |

Leaf-cutting ants of the genera Atta and Acromyrmex determine serious agricultural problems and live on symbiosis with Leucoagaricus gongylophorus. The aim of this study is to identify morphological and molecularly, as well as to verify the genotypic variability of the symbiotic fungus cultivated by A. heyeri and A. ambiguus from three different regions of the state of Rio Grande do Sul, Brazil. Fungus gardens were collected and fragments of mycelia were grown in selective medium. Total DNA was extracted and amplification of the ITS region was performed by PCR using universal primers. After DNA sequencing, the chromatograms were assembled and phylogenetic analyzes were performed by the Neighbor-Joining method. A total of six isolates of L. gongylophorus... |

The isolation and maintenance of symbiotic basidiomycete fungi living in association with ants of the tribe Attini has been hindered by the slow growth rate of these fungi and the presence of other microorganisms on the surface of the material which the ants maintain inside their nests to provide a growth substrate for their symbiont. In this paper we describe a method which increases the efficiency of isolation of these fungal symbionts by over seven fold as compared to traditional isolation procedures. Underground nests of attine ants of the genera Atta, Acromyrmex, Trachymyrmex and Mycetarotes were located, from which samples containing the fungal symbiont and ants were collected and transported to the laboratory where the ants were able to clean the... |
