Clinical samples from 71 bovine from different Brazilian states were processed for the analysis of anaerobe organisms with emphasis on the isolation and characterization of Clostridium spp. From these, eighty-nine Clostridium perfringens strains were recovered: 32 from liver, 19 from intestinal contents, 14 from kidney, 6 from rumen, 5 from nervous system, 4 from bone marrow, 2 from udder tract, blood, spleen and lung, and one from muscle. Eighty-nine of 144 isolates of Clostridium perfringens obtained from 187 samples of 71 bovine in several Brazilian states were submitted to esterase electrophoresis for typing. Mobilites electrophoresis, as parameter, were settled down by isolates from ATCC pattern of types A, B, C, and D. Of the 89 isolates, 43 (48.3%) were characterized as electrophoretic type A, 20 (22.5%) as D, 18 (20.2%) as C, and 3 (3.4%) as B. Five (5.6%) isolates did not identity with any type. Similarly, esterase electrophoresis enabled the typing of 94.4 % of the isolates, demonstrating to be an appropriate method for animal sample analyses. |

Of the 102 samples collected from mammals and birds, both domestic and captive wild, 48 were found to be positive for Clostridium perfringens. Most of the mammal isolates (84.38%) appeared to have been collected from clinically affected animals, while 33.33% of the bird samples were from clinically affected and 21.43% from apparently healthy birds infected with C. perfringens. Isolates revealed high sensitivity to ciprofloxacin, enrofloxacin and norfloxacin. Among the isolated C. perfringens, 30 (62.50%) showed DNase production. Ten out of eighty-nine strains biochemically identified as Clostridium perfringens, isolated from bovine organs, were selected by their different results showed in toxigenicity test on mice. Those and the standard strains, ATCC types A, B, C, and D, had their virulence exalted through serial intramuscular inoculation into guinea pigs. Results showed that, for toxigenic strains (6), one or two passages were enough to cause exaltation, while for the atoxigenic (4), five or six inoculations were needed. Toxigenic types of Clostridium perfringens are significant causative agents of enteric disease in domestic animals, although type E is presumably rare, appearing as an uncommon cause of enterotoxemia of lambs, calves and rabbits. We report herein the typing of 23 C. perfringens strains, by the polymerase chain reaction (PCR) technique, isolated from small intestine samples of bovines that have died suddenly, after manifesting or not enteric or neurological disorders. A prototoxina epsilon foi obtida a partir de cultivo líquido, rico em carboidratos, a partir de amostra de C. perfringens tipo D (INTA - Argentina), incubado em condições de anarobiose à 37ºC por oito horas. O sobrenadante da cultura foi concentrado utilizando-se o sistema Amicon com membrana de "cut off" de 10 kDa. O concentrado foi precipitado com sulfato de amônio (350 g/ml) e dissolvido em tampão fosfato 0,01 M (pH 7,2). A prototoxina purificada por cromatografia de troca iônica (DEAE-Sepharose CL 6B) e isolada no primeiro pico, mostrou uma banda única de aproximadamente 34kDa de peso molecular em SDS-PAGE, além de ser tóxica em camundongos. |

Avaliou-se a resposta de antitoxinas beta e épsilon de Clostridium perfringens em bovinos vacinados contra clostridioses com seis vacinas disponíveis no mercado. Quarenta e oito bezerros de seis a sete meses de idade foram divididos em oito grupos (T1 a T8) de seis animais cada. Os grupos de número 2 a 7 receberam as vacinas T2 a T7 nos dias 0 e 42 com a dose e via recomendadas pelos fabricantes. Empregaram-se os métodos cromatográficos de afinidade metálica e de imunoafinidade para purificação da toxina beta em sobrenadante de cultivo de Clostridium perfringens tipo C. Observaram-se, na eletroforese das primeiras frações eluidas nos dois métodos de purificação, uma banda de peso molecular aproximado de 38kDa, característica da forma monomérica de toxina beta de Clostridium perfringens tipo C, e bandas de peso moleculares superiores, referentes às suas formas oligoméricas. In this study, we screened differentially expressed genes in a multidrug-resistant isolate strain of Clostridium perfringens by RNA sequencing. We also separated and identified differentially expressed proteins (DEPs) in the isolate strain by two-dimensional electrophoresis (2-DE) and mass spectrometry (MS). The RNA sequencing results showed that, compared with the control strain, 1128 genes were differentially expressed in the isolate strain, and these included 227 up-regulated genes and 901 down-regulated genes. Clostridium perfringens is an important pathogen in human and veterinary medicine. In swine, the agent is responsible for necrotic enteritis and enterotoxemia characterized by diarrhea, weight loss, delayed development and, in some cases, death. In the present study amplified fragment length polymorphism analyses (AFLP) was used to characterize 54 C. perfringens strains isolated from swine presenting diarrhea. The objective of this paper was to assess bacteriological quality of drinking water in a peri-urban area located in the Metropolitan Region of São Paulo, Brazil. A total of 89 water samples were collected from community plastic tanks and 177 water samples from wells were collected bimonthly, from September 2007 to November 2008, for evaluating bacteriological parameters including: Escherichia coli, Enterococcus and heterotrophic plate count (HPC).
