Manganese phosphite in Coffee defence against Hemileia vastatrix, the coffee rust fungus: biochemical and molecular analyses. Repositório Alice
MONTEIRO, A. C. A.; RESENDE, M. L. V. de; VALENTE, T. C. T.; RIBEIRO JUNIOR, P. M.; PEREIRA, V. F.; COSTA, J. R. da; SILVA, J. A. G. da.
Phosphites (Phi) are inorganic salts of phosphorous acid that have been used as resistance elicitors to activate mechanisms underlying plant defence. This study assessed the effectiveness of manganese phosphite (MnPhi) for the management of rust (Hemileia vastatrix) on coffee seedlings. The effect of this compound on the induction of resistance was also measured by the expression of defence-related genes such as POX (peroxidase), CAT (catalase), GLU (b-1.3-glucanase) and PAL (phenylalanine ammonia-lyase) and by the activity of defence enzymes such as ascorbate peroxidase (APX), superoxide dismutase (SOD) and polyphenol oxidase (PPO). Foliar sprayings of MnPhi on coffee seedlings provided 63% control of rust severity. This compound induced defence responses...

Enzymatic activity as an early sensor in the evaluation of soil quality in silvopastoral systems. Repositório Alice
SANTOS, J. V. DOS; BENTO, L. R.; BRESOLIN, J. D.; OLIVEIRA, P. P. A.; BERNARDI, A. C. de C.; PEZZOPANE, J. R. M.; MENDES, I. de C.; MARTIN NETO, L..
The demand for food has intensified at the rate that the world population has grown, requiring production models capable of meeting world needs, through high productivity and conciliating with sustainable practices. In this scenario, the silvopastoral system (SPS) has been introduced as an alternative to current production models. Thus, this study aimed to evaluate the current soil condition in an SPS using chemical and microbiological parameters as indicators of soil quality, using a native forest and degraded pasture as a positive and negative reference, respectively. For this, soil samples were collected at a depth of 0-10 cm, and the total carbon and B-glycosidase activity (BGL) were determined. Both SPS sub-areas, in the tree rows (SPS-R) and between...

Soil attributes as indicators of the stabilization process of erosion in gullies at different formation stages in the southeast region of Brazil Repositório Alice
SILVA, C. F. da; PEREIRA, M. G.; RODRIGUES, L. A. da S.; TEODORO, S. A.; MARCELO ANTONIOL FONTES; GAIA-GOMES, J. H.; SILVA, E. M. R. da.
Water erosion is one of the main forms of soil degradation, causing economic, environmental, and social damage. This study evaluated the effects of different formation stages of gullies (initial - IG; juvenile- JG; mature - MG; and senile - SG) on the chemical, organic carbon and microbiological attributes of soil, using a secondary forest (SF) and two areas of pasture as references in the ?Mar de Morros? environment of the Brazilian Atlantic Forest biome. Soil samples (depth 0?0.05 m) were collected at the end of the rainy and dry seasons. Gullies in different stages of formation promote a decrease in soil fertility and modification of microbiological attributes, particularly when compared with secondary forest areas. Reductions of over 60% in total...

Bresolin,Iara Rocha Antunes Pereira; Bresolin,Igor Tadeu Lazzarotto; Silveira,Edgar; Tambourgi,Elias Basile; Mazzola,Priscila Gava.
The aim of this work was to isolate and purify bromelain extracted from the pineapple peel by ammonium sulfate precipitation (40-80%), followed by desalting and freeze-drying with a 75% activity recovery and 2.2 fold increased specific activity. Ion exchange chromatography on DEAE-Sepharose was able to separate the polysaccharides from the enzyme, which was recovered in the elution step, maintaining its enzymatic activity. The batch adsorption of bromelain was evaluated in terms of total protein and enzymatic activity using Langmuir and Langmuir-Freundlich models. Results showed that the process could be suitable for the recovery and purification of the enzyme, maintaining its specific activity.

Gioda,Carolina Rosa; Pretto,Alexandra; Freitas,Carine de Souza; Leitemperger,Jossiele; Loro,Vania Lucia; Lazzari,Rafael; Lissner,Leandro Ademar; Baldisserotto,Bernardo; Salbego,Joseânia.
ABSTRACT: The aim of this study was to verify the activity of some digestive enzymes in four fish species with different feeding habits. Knowledge of these enzymatic activities can help us to understand the species' digestive processes. The species chosen for this study were Ctenopharyngodon idella (herbivore), Rhamdia quelen (omnivore), Leporinus obtusidens (omnivore) and Hoplias malabaricus (carnivore). The digestive tract of these species was divided into four portions to estimate enzymatic activity: stomach, anterior, mid and posterior intestine. Ctenopharyngodon idella had the highest amylase and maltase activities in all portions of the gut, followed by L. obtusidens . The highest trypsin activity was observed in all gut portions of H. malabaricus,...

Bravo Rodríguez,Vicente; Jurado Alameda,Encarnación; Martínez Gallegos,Juan Francisco; Reyes Requena,Antonia; García López,Ana Isabel; Sampaio Cabral,Joaquim Manuel; Fernandes,Pedro; Pina da Fonseca,Luis Joaquim.
The influence of different commercial surfactants on the enzymatic activity of a commercial alpha-amylase from Bacillus licheniformis (Termamyl 300 L) has been studied. As non-ionic surfactants, alkyl polyglycosides (Glucopon® 215, Glucopon® 600 and Glucopon® 650) were studied, as were fatty alcohol ethoxylates (Findet 1214N/23 and Findet 10/15), and nonyl phenol ethoxylate (Findet 9Q/21.5NF). Also, an anionic surfactant, linear alkyl benzene sulfonate (LAS) was assayed. In general, none of the non-ionic surfactants studied, except Findet 10/15, vary substantially the enzymatic activity. Findet 10/15 has the strongest hydrophobic character and reduces the enzymatic activity more significantly the greater its concentration. Regarding LAS, this surfactant...

Cloning, expression and characterization of alcohol dehydrogenases in the silkworm Bombyx mori Genet. Mol. Biol.
Wang,Nan; Shi,Haifeng; Yao,Qin; Zhou,Yang; Kang,Lequn; Chen,Huiqin; Chen,Keping.
Alcohol dehydrogenases (ADH) are a class of enzymes that catalyze the reversible oxidation of alcohols to corresponding aldehydes or ketones, by using either nicotinamide adenine dinucleotide (NAD) or nicotinamide adenine dinucleotide phosphate (NADP), as coenzymes. In this study, a short-chain ADH gene was identified in Bombyx mori by 5'-RACE PCR. This is the first time the coding region of BmADH has been cloned, expressed, purified and then characterized. The cDNA fragment encoding the BmADH protein was amplified from a pool of silkworm cDNAs by PCR, and then cloned into E. coli expression vector pET-30a(+). The recombinant His-tagged BmADH protein was expressed in E. coli BL21 (DE3), and then purified by metal chelating affinity chromatography. The...

Physiological and biochemical changes in lettuce seeds during storage at different temperatures Horticultura Brasileira
Catão,Hugo Cesar RM; Gomes,Luiz Antonio Augusto; Guimarães,Renato M; Fonseca,Pedro Henrique F; Caixeta,Franciele; Galvão,Alexandre G.
ABSTRACT Dormancy in lettuce seeds is a process not yet fully understood. High storage temperatures can cause seed dormancy promoting physiological and enzymatic changes. The goal of this study was to investigate the influence of storage period and environments on the quality and dormancy of different lettuce cultivars. We also investigated the biochemical changes. A completely randomized experimental design was used in a 4x8x3 factorial arrangement to evaluate physiological quality on different storage periods (30, 60, 90 and 120 days) and environments (15, 25 and 35°C) of seeds from eight lettuce cultivars (Everglades, Babá de Verão, Elisa, Luiza, Grand Rapids, Hortência, Salinas 88 and Rubete). The biochemical activity was accessed by tetrazolium test...

Microbial extracellular enzyme detection on agar plates by means of fluorogenic methylum-belliferyl-substrates. ArchiMer
Kim, S; Hoppe, H.
A rapid and sensitive method to detect the extracellular enzymatic activity of bacteria colonies grown on agar plates is described. Selective agar media supplemented with protein, starch, chitin, Tween-80, etc. are conventionally used to detect biochemical properties of bacteria. It has been experimentally demonstrated with bacteria pure cultures that fluorogenic Methylumbelliferyl (MUF)-substrates are excellent substrate analogues for normally occurring polymers. Based on MUF-substrate hydrolysis the new method provides reliable qualitative estimates of extracellular enzymatic properties of bacteria within minutes using pure cultures as well as agar plates prepared for colony counts.

Adaptive features of a bacteriolytic enzyme from the style of the mussel Choromytilus meridionalis in response to environmental fluctuations. ArchiMer
Seiderer, L; Robb, F.
The extracellular digestive apparatus of bivalve molluscs consists of a crystalline style containing diverse degradative enzymes which are released during rotation and dissolution of the style. This paper reviews the occurrence of a temperature-regulated bacteriolytic enzyme in the black mussel Choromytilus meridionalis , which is an inhabitant of coastal reefs on the south and south west coast of South Africa. This agent is unable to lyse Micrococcus luteus or Escherichia coli , both of which are susceptible to lysis by a true lysozyme. It appears to have a restricted spectrum of activity which includes both free-living and kelp-associated gram-negative bacteria. In addition, the level of activity varies dramatically according to the environmental water...
