Although it is generally accepted that plant in vitro culture response is influenced by the donor genotype, the genetic and molecular bases of this phenomenon are barely known. As a consequence, the optimization of tissue culture protocols is mainly empirically done. Researchers of the IGEAF studied the genetic basis of the in vitro regeneration of various plant species, including the tissue culture response of artificially induced barley mutants. Background: The perennial medicinal herb Dioscorea zingiberensis is a very important plant used for steroid drug manufacturing for its high level of diosgenin in rhizome. Although the stimulation of diosgenin accumulation by ethylene has been reported in a few of plant species, its regulation is not yet characterized at the molecular level, the underlying molecular mechanism remains elusive. Color change and quality response of ‘lane late’ orange submitted to degreening process REA

This study aimed at evaluating the effects of ethylene on peel color and compositional changes in ‘Lane late’ orange stored under refrigerated and ambient conditions. Physiologically mature, but green-peeled, oranges were exposed to ethylene gas under room temperature and high relative humidity for 24 hours. Storage chamber was ventilated with fresh air after 12 hours to mitigate consequences derived from fruit respiration. Tomato (Lycopersicon esculentum Mill.) fruits, cv. Santa Clara, were harvested at the breaker stage from commercial fields in Brazlândia, Brazil, to investigate the ability of 1-methylcyclopropene (1-MCP) to retard tomato fruit ripening. Fruit without external blemishes were graded for size (diameter = 80±5 mm) and mass (m = 130±10 g), placed inside hermetically sealed boxes, and 1-MCP was applied for 12 hours (T = 22±1°C; RH = 80-85%) at four different concentrations: 0 (control), 250, 500 and 1000 mL.L-1. The experimental design was completely randomized in factorial scheme 4x4, with four storage temperatures (5, 10, 15 and 20ºC) and four evaluation periods (0, 3, 6 and 9 days after processing) with four replications. Differential esterase activity in aerial and root tissues from 6as DHR wheat lines after phytohormonal treatments JBAG

Activity of aerial and root esterases in wheat DHR lines of chromosome 6AS was tested. Different patterns were found after applying phytohormone treatments that mimic stress conditions. Ethylene treated plants showed the lowest aerial and root weights; also, the smallest foliar area, in conjunction with absence of esterase activity in more than half of the lines, was observed. Additional studies will contribute to clarify the key role of ET in the expression of defense related genes.

Tratamentos pré-germinativos em sementes de palmiteiro PFB - Pesquisa Florestal Brasileira

Este trabalho objetivou avaliar a eficiência de tratamentos pré-germinativos na promoção da germinação e crescimento inicial de plântulas de palmiteiro. Os tratamentos avaliados foram: controle, imersão em solução de GA3, exposição ao etileno, imersão em água, imersão em H2SO4, escarificação mecânica, estratificação por 30 dias a 10°C, e escarificação seguida de estratificação. A imersão de sementes em solução de ácido giberélico (GA3; 2000 µL L-1 durante 24 h) ou sua exposição ao etileno (1000 µL L-1 durante 24 h) são eficientes para a promoção da emergência e crescimento inicial de plântulas de palmiteiro.

Latex production with depolymerizing compounds of actin cytoskeleton in rubber trees PAB

The objective of this work was to assess stimulated latex flow from rubber trees (Hevea brasiliensis) with saturated macrolide (latrunculin A), 1, 5, and 10% potassium iodide in 2% methylcellulose compared with 0.3% ethylene in 2% methylcellulose (check) and 2% methylcellulose (blank). Latex output and contents of pure rubber, total solid, sucrose, inorganic phosphorus, thiol, and Mg2+ were measured. The treatments containing 1% KI or saturated macrolide increased latex yields compared to the blank with 2% methylcellulose alone. The 1% KI or saturated macrolide treatments were equal to that of 0.3% ethylene check treatment. However, 5 and 10% KI were harmful to bark of rubber trees, even caused prolonged tapping panel dryness.

Costly investments in storage and transport of onion bulbs makes it increasingly important to identify cultivars with the best chance of long-term storage. This paper discusses our own and other researchers' results and laboratory procedures on onion's pungency, soluble solids, dry matter, respiration rate, carbohydrates and catabolism-related enzymes under different storage conditions. The variability in parameters such as carbohydrate composition, pungency, soluble solids and soluble uronic acid concentration in water is growth dependent, and defines the quality of onion bulbs at harvest time. The aim of this work was to evaluate the germination response of Vochysia tucanorum Mart. seeds treated with GA3 and CEPA and germinated under white light or darkness. Newly collected seeds from a Cerrado area were stored for 14 days at two temperatures (25 °C ± 2 and 7 °C ± 1). After the storage period the seeds were pre-treated with distilled water (control), gibberellic acid (GA3), 2-chloroethylphosphonic acid (CEPA) and a mixture of GA3 + CEPA. Effect of aminoethoxyvinilglycine (AVG) on preharvest fruit drop and maturity of apples Rev. Bras. Frutic.

Apple trees cultivars Gala and Fuji were sprayed four weeks before commercial harvest with aminoethoxyvinilglycine (AVG), at doses of 0, 125, or 250 mg L-1, and assessed for preharvest fruit drop, fruit growth, and maturation on tree. In 'Gala', 64 days after AVG spraying, fruit drop for control treatment was 85%, and AVG (at 125 and 250 mg L-1) reduced it to 10%. In 'Fuji', 64 days after AVG spraying, fruit drop for control was 6%, while treatments with AVG (at 125 and 250 mg L-1) increased fruit drop to 10%. Abstract The objective of this study was to assess the effects of post-bloom (PB) or preharvest (PH) treatments of ‘Braeburn’ apple trees with prohexadione-calcium (ProCa; 300 mg L-1), an inhibitor of gibberellins biosynthesis, or GA4+7 (300 mg L-1) on fruit quality at harvest and after storage (four months at 0±0.5 oC, followed by five days of shelf life).
