Screening of basidiomycetes for the production of exopolysaccharide and biomass in submerged culture Rev. Microbiol.
Maziero,Rosana; Cavazzoni,Valeria; Bononi,Vera Lúcia Ramos.
Fifty-six strains of Basidiomycetes, including native Brazilian fungi isolated from different ecosystems and edible mushrooms, were screened for production of exopolysaccharides and biomass in submerged culture. Agaricus sp. (CCB 280) and Oudemansiella canarii (Jungh.) Hohn (CCB 179) were the highest exopolysaccharide producers (6.01 and 3.54 g dry w./l respectively) after 7 days of incubation. The best producer of biomass was Schizophyllum commune Fr.:Fr. (CCB 473) with 16.68 g dry w./l in 14 days of incubation. When the culture filtrate was submitted to freezing prior to polysaccharide precipitation, a gelatinous fraction was formed.

GERN, J. C.; SANTOS, L. F. dos; HABU, S.; NOSEDA, M. D.; LIMA, L. F. O. de; SILVA, A. L. L. da; SOCCOL, V. T.; BRANDAO, H. de M.; SOCCOL, C. R..
Background and purpose: Although the activities of polysaccharides such as beta-glucans are well known in Agaricus brasiliensis, there has been little research on their exopolysaccharides and its characterization or scale up studies. The aim of the study was to study the monosaccharide composition of the exopolysaccharide (EPS) produced by Agaricus brasiliensis LPB3 and its production in a stirred tank fermenter. Materials and methods: Agaricus brasiliensis LPB3 was cultured in submerged fermentation in a medium containing glucose, yeast extract, K2HPO4 and MgSO4. Extracellular polysaccharide was precipitated with four volumes of 95% ethanol. For monosaccharide composition, the EPSs were hydrolyzed, derivatized to alditol acetates, and analyzed by gas...

ABSTRACT The effect on different three carbon source (i.e. glucose, fructose and sucrose) on production, chemical characterization and antioxidant activity of exopolysaccharide (EPS) produced by Phellinus vaninii Ljup was investigated in this study. Amongst carbon sources examined, glucose and sucrose were favorable for the mycelia growth, while the maximum EPS yield was achieved when sucrose was employed. The predominant carbohydrate compositions in EPSs identified were gluconic acid, glucose, mannose and galactose acid. Then, FT-IR spectral analysis revealed prominent characteristic groups in EPSs. EPSs molecule exist as nearly globular shape form in aqueous solution. The variation also affects antioxidant activities by investigated by using hydroxyl and...

Influence of gastrointestinal system conditions on adhesion of exopolysaccharide-producing Lactobacillus delbrueckii subsp. bulgaricus strains to caco-2 cells BABT
Darilmaz,Derya Onal; Aslım,Belma; Suludere,Zekiye; Akca,Gülcin.
This study aimed to assess the transit tolerance of potential probiotic dairy Lactobacillus strains in human uppergastrointestinal tract in vitro, and to evaluate the effect of EPS production on the viability and adhesion of these strains. Survival and adhesion of two exopolysaccharide (EPS)-producing L. delbrueckii subsp. bulgaricus strains (B3 and B2) and E. coli ATCC11229 were assessed after the exposure of different pH (gastric juice) and gastric plus pancreatic juice challenges. In the artificial gastric juice (pH 2), both the viability of the strain B3 and B2 was decreased. Artificial juice treatments significantly reduced the adhesion to caco-2 cells (P< 0.05). High EPS-producing B3 survived better in the adverse gastrointestinal conditions and...

Ernandes,Fernanda Maria Pagane Guereschi; Garcia-Cruz,Crispin Humberto.
The aim of this work was to study the nutritional requirements of Zymomonas mobilis CCT 4494 for levan production in three chemically defined media. During the optimization of the fermentative process for the production of the exopolysaccharide, different concentrations of glucose, fructose and sucrose as carbon source and yeast extract as vitamin source were tested. Variations of incubation temperature and initial pH of the medium were observed. The results showed that medium containing 20.0% sucrose and 0.5% yeast extract, with initial pH of 7.0, incubated at 30°C gave a 43.0% yield of the biopolymer.

Bacterial exopolysaccharide and biofilm formation stimulate chickpea growth and soil aggregation under salt stress BJM
Qurashi,Aisha Waheed; Sabri,Anjum Nasim.
To compensate for stress imposed by salinity, biofilm formation and exopolysaccharide production are significant strategies of salt tolerant bacteria to assist metabolism. We hypothesized that two previously isolated salt-tolerant strains Halomonas variabilis (HT1) and Planococcus rifietoensis (RT4) have an ability to improve plant growth, These strains can form biofilm and accumulate exopolysacharides at increasing salt stress. These results showed that bacteria might be involved in developing microbial communities under salt stress and helpful in colonizing of bacterial strains to plant roots and soil particles. Eventually, it can add to the plant growth and soil structure. We investigated the comparative effect of exopolysacharide and biofilm formation...

Gordonia polyisoprenivorans from groundwater contaminated with landfill leachate in a subtropical area: characterization of the isolate and exopolysaccharide production BJM
Fusconi,Roberta; Godinho,Mirna Januária Leal; Hernández,Isara Lourdes Cruz; Bossolan,Nelma Regina Segnini.
A strain of Gordonia sp. (strain Lc), from landfill leachate-contaminated groundwater was characterized by polyphasic taxonomy and studied for exopolysaccharide (EPS) production. The cells were Gram-positive, catalase-positive, oxidase-negative and non-motile. The organism grew both aerobically and, in anoxic environment, in the presence of NaNO3. Rods occured singly, in pairs or in a typical coryneform V-shaped. The organism had morphological, physiological and chemical properties consistent with its assignment to the genus Gordonia and mycolic and fatty acid pattern that corresponded to those of G. polyisoprenivorans DSM 44302T. The comparison of the sequence of the first 500 bases of the 16S rDNA of strain Lc gave 100% similarity with the type strain of...

Growth and exopolysaccharide production by Streptococcus thermophilus ST1 in skim milk BJM
Zhang,Tiehua; Zhang,Chunhong; Li,Shengyu; Zhang,Yanchun; Yang,Zhennai.
To analyze the exopolysaccharide (EPS) production by Streptococcus thermophilus ST1, cultures were cultivated in 10% (w/v) reconstituted skim milk under different growth conditions including various temperatures and pHs of growth medium, supplementation of the medium with various carbon sources (glucose, lactose, sucrose, galactose and fructose) and nitrogen source (whey protein concentrate, or WPC). The results showed that most EPS production by strain ST1 was obtained at a temperature (42°C) and pH (6.5) optimal for its growth. Supplementation of the skim milk medium with either carbohydrates or WPC increased both growth and polymer formation by different extents, with sucrose being most effective among the carbon sources tested. Under the optimal...

Industrial wastewater as raw material for exopolysaccharide production by Rhizobium leguminosarum BJM
Sellami,Mohamed; Oszako,Tomasz; Miled,Nabil; Ben Rebah,Faouzi.
The objective of this study was to evaluate the exopolysaccharide (EPS) production by Rhizobium leguminosarum cultivated in wastewater generated by oil companies (WWOC1 and WWOC2) and fish processing industry (WWFP). The results obtained in Erlenmeyer flasks indicated that the rhizobial strain grew well in industrial wastewater. Generally, wastewater composition affected the growth and the EPS production. WWFP allowed good bacterial growth similar to that obtained with the standard medium (YMB). During growth, various quantities of EPS were produced and yields varied depending on the media. Growing in YMB, EPS production did not exceed 9.7 g/L obtained after 72 h of growth. In wastewater, the maximum EPS value reached 11.1 g/L obtained with the fish...

Rheology of spreadable goat cheese made with autochthonous lactic cultures differing in their ability to produce exopolysaccharides Ciênc. Tecnol. Aliment.
Florencia,Frau Silvia.
The aim of this study was to compare the rheology of spreadable cheeses elaborated with autochthonous lactic starter cultures without the addition of exopolysaccharide-producing strain in the same starter with exopolysaccharide-producing strain. From a rheological standpoint, both samples were characterized as weak viscoelastic gels and pseudoplastic products. It was concluded that cheese made with exopolysaccharide-producing strain showed smaller G', G", and η* values over the range of frequencies studied and smaller critic stress values than the cheese without exopolysaccharide-producing strain. The results obtained indicate that cheeses without exopolysaccharide-producing strain need to be added with any texture enhancer product.

Computational analysis suggests that virulence of Chromobacterium violaceum might be linked to biofilm formation and poly-NAG biosynthesis Genet. Mol. Biol.
Becker,Sidnei; Soares,Cíntia; Porto,Luismar Marques.
Groups of genes that produce exopolysaccharide with a N-acetyl-D-glucosamine monomer are in the genome of several pathogenic bacteria. Chromobacterium violaceum, an opportunistic pathogen, has the operon hmsHFR-CV2940, whose proteins can synthesize such polysaccharide. In this work, multiple alignments among proteins from bacteria that synthesize such polysaccharide were used to verify the existence of amino acids that might be critical for pathogen activity. Three-dimensional models were generated for spatial visualization of these amino acid residues. The analysis carried out showed that the protein HmsR preserves the amino acids D135, D228, Q264 and R267, considered critical for the formation of biofilms and, furthermore, that these amino acids are...

Exopolysaccharides produced by bacteria isolated from deep-sea hydrothermal vents: new agents with therapeutic potential ArchiMer
Colliec-jouault, Sylvia; Zanchetta, P; Helley, D; Ratiskol, Jacqueline; Sinquin, Corinne; Fischer, A. M.; Guezennec, Jean.
Bacterial polysaccharides offer fascinating potential applications for the pharmaceutical industry. Although many known marine bacteria produce exopolysaccharides (EPS), continuation in looking for new polysaccharide-producing micro-organisms is promising. Marine bacteria, isolated from deep-sea hydrothermal vents, have demonstrated their ability to produce in aerobic conditions, unusual EPS. With the aim of discovering biological activities, EPS presenting different structural features were studied. An EPS secreted by Vibrio, diabolicus was evaluated on the restoration of bone integrity in experimental model and was demonstrated to be a strong bone-healing material. Another EPS produced by Alteromonas infernus was modified in order to obtain new...

Transparent Exopolymeric Particles (TEP) Selectively Increase Biogenic Silica Dissolution From Fossil Diatoms as Compared to Fresh Diatoms ArchiMer
Toullec, Jordan; Moriceau, Brivaela.
Diatom production is mainly supported by the dissolution of biogenic silica (bSiO(2)) within the first 200 m of the water column. The upper oceanic layer is enriched in dissolved and/or colloidal organic matter, such as exopolymeric polysaccharides (EPS) and transparent exopolymeric particles (TEP) excreted by phytoplankton in large amounts, especially at the end of a bloom. In this study we explored for the first time the direct influence of TEP-enriched diatom excretions on bSiO(2) dissolution. Twelve dissolution experiments on fresh and fossil diatom frustules were carried out on seawater containing different concentrations of TEP extracted from diatom cultures. Fresh diatom frustules were cleaned from the organic matter by low ash temperature, and...

Articular cartilage is an avascular, non-innervated connective tissue with limited ability to regenerate. Articular degenerative processes arising from trauma, inflammation or due to aging are thus irreversible and may induce the loss of the joint function. To repair cartilaginous defects, tissue engineering approaches are under intense development. Association of cells and signalling proteins, such as growth factors, with biocompatible hydrogel matrix may lead to the regeneration of the healthy tissue. One current strategy to enhance both growth factor bioactivity and bioavailability is based on the delivery of these signalling proteins in microcarriers. In this context, the aim of the present study was to develop microcarriers by encapsulating...

Effects of a sulfated exopolysaccharide produced by Altermonas infernus on bone biology ArchiMer
Velasco, C. Ruiz; Baud'Huin, M.; Sinquin, Corinne; Maillasson, M.; Heymann, D.; Colliec-jouault, Sylvia; Padrines, M..
The growth and differentiation of bone cells is controlled by various factors which can be modulated by heparan sulphates. Here, we investigated the effects of an oversulphated exopolysaccharide (OS-EPS) on bone. We compared the effect of this compound with that of a native exopolysaccharide (EPS). Long-term administration of OS-EPS causes cancellous bone loss in mice due, in part, to an increase in the number of osteoclasts lining the trabecular bone surface. No significant difference in cancellous bone volume was found between EPS-treated mice and age-matched control mice, underlying the importance of sulphation in trabecular bone loss. However, the mechanism sustaining this osteoporosis was unclear. To clarify OS-EPS activities, we investigated the...
