Description of Amazonian Theobroma L. collections, species identification, and characterization of interspecific hybrids Acta Botanica

There are two major in vivo collections of species of the genus Theobroma L. in Belém and Marituba, state of Pará, Brazil, and in both there are natural species, as well as natural and artificial interspecific hybrids. In order to organize a database of Brazilian Theobroma species, a description of the existing collections and detailed information about the interspecific hybrids, including an artificial key for their identification, are presented in this article.

Avaliou-se o complemento cromossômico de 16 acessos de Pennisetum disponíveis na Coleção de Germoplasma da Embrapa Gado de Leite – Juiz de Fora, Estado de Minas Gerais. Para a avaliação citogenética, utilizou-se a técnica de esmagamento e coloração com Feulgen. Para quatro acessos foram obtidas metáfases com 2n = 28 cromossomos, confirmando sua classificação como P. purpureum. A origem híbrida P. purpureum x P. glaucum foi constatada em oito acessos, os quais apresentaram 2n = 21. Em um acesso identificado como híbrido entre P. purpureum x P. glaucum, foram observadas metáfases com 2n = 54, permitindo inferir que não se trata de um híbrido interespecífico triplóide. Protoplast production from napier grass and pearl millet triploid hybrids Ciência e Agrotecnologia

The objective of this work was to obtain protoplasts from napier grass and pearl millet triploid hybrids as a basis for future studies on chromosomal duplication. Explants were taken from mesophyll of in vitro- and in vivo-cultured plants or from calli of two triploid hybrids (H1 and H2), which were treated with enzymatic solutions containing different concentrations of cellulase R-10 (0.5, 1.0, 1.5 and 2.0%) with an additional 0.2% macerozyme and 0.1% driselase or 1.0% pectolyase Y-23 and 0.5% hemicellulase. Enzymatic digestion was monitored once every hour for five hours. ABSTRACTNapier grass and pearl millet are tropical forages from the genus Pennisetum. The variability in those species is explored in breeding programs of forages, as well as in the production of interspecific hybrids. Hybridization is a phenomenon that leads to intergenomic conflicts following the elimination of genomic sequences. In this sense, the present work aimed to study the genomic alterations occurring after interspecific hybridization of pearl millet and Napier grass with the use of cytogenetics and flow cytometry tools. Cytogenetics of Pennisetum purpureum Schumack x Pennisetum glaucum L. hybrids and their parents Ciência e Agrotecnologia

Cytogenetic studies were carried out on five accesses of interspecific hybrids of Pennisetum purpureum and Pennisetum glaucum and their parentals, which are part of the Forage Germplasm Bank of the "Centro Nacional de Pesquisa de Gado de Leite" of EMBRAPA, Brazil. The mitotic cytogenetic analysis, performed using stain by Schiff reactive, confirmed the chromosome number of 2n = 14, 28 and 21 for the accesses of P. glaucum, P. purpureum and their hybrids, respectively. The fruit flies Drosophila mulleri and Drosophila navojoa are included in the mulleri complex of the mulleri subgroup and Repleta group. Although there is no demonstration that interspecific crosses between them occur in nature, they intercross in the laboratory in both cross directions. Previous data have shown the occurrence of nucleolar dominance in interspecific hybrids of some species in the mulleri complex. The cultivated and sexually compatible species Pennisetum purpureum (elephant grass, 2n = 4x = 28) and Pennisetum glaucum (pearl millet, 2n = 2x = 14) can undergo hybridization which favors the amplification of their genetic background and the introgression of favorable alleles into breeding programs. The main problem with interspecific hybrids of these species is infertility due to triploidy (2n = 3x = 21). The first investigated the horizontal transmission of a putative causal agent of the mortality among two stocks of wild Mytilus edulis and the potential genetic resistance of adults and their progenies. The objective of this work was to evaluate the genomic behavior of hybrid combinations between elephant grass (Pennisetum purpureum) and pearl millet (P. glaucum). Tetraploid (AAA'B) and pentaploid (AA'A'BB) chromosome races resulting from the backcross of the hexaploid hybrid to its parents elephant grass (A'A'BB) and pearl millet (AA) were analyzed as to chromosome number and DNA content. Genotypes of elephant grass, millet, and triploid and hexaploid induced hybrids were compared. Pentaploid and tetraploid genomic combinations showed high level of mixoploidy, in discordance with the expected somatic chromosome set.
