OCCURRENCE OF MELOIDOGYNE JAVANICA PARASITING ROOTS AND NODULES OF PEANUTS IN PARAGUAY Bragantia

The root-knot nematode Meloidogyne javanica (Treub, 1885) Chitwood, 1949, was identified in roots, nodules and shells of peanut plants (Arachis hypogaea L.), cv. Florman-INTA, collected from a field in Menno colony, Chaco Central region of Paraguay. Infected plants were observed in patches, showing wilting and drying of vegetative parts. Peanut had been grown ininterruptly for many years in the same area and patches had already been observed in the preceding year in cv. Starr. Nematodes collected from roots in this field reproduced on tomato cultivar rutgers and peanut cultivar florunner and produced egg-masses on roots, nodules and shells of cultivar Florman. This is the first report on M. javanica parasiting peanut in Paraguay.

ABSTRACT This study aimed to explore the effects of two siderophore-producing bacterial strains on iron absorption and plant growth of peanut in calcareous soil. Two siderophore-producing bacterial strains, namely, YZ29 and DZ13, isolated from the rhizosphere soil of peanut, were identified as Paenibacillus illinoisensis and Bacillus sp., respectively. Effect of gamma irradiation on the inactivation of aflatoxin B1 and fungal flora in peanut BJM

The effect of gamma irradiation on aflatoxin B1 levels and fungal infection were investigated in peanut samples, Tatu Vermelho cultivar. At a radiation dose of 10 KGy, growth of molds was completely inhibited. Doses of 15, 20, 25 and 30 KGy were sufficient for destruction of aflatoxin B1 by 55-74%. The results suggested that the decontamination of molds by irradiation, before production of aflatoxin B1, is the most acceptable method in the preservation of peanut.

Risk assessment of the occurrence of aflatoxin and fungi in peanuts and cashew nuts BJPS

In the present study, the occurrence of fungi and aflatoxins (AFs) in peanut and cashew nut samples was investigated. Mycological analysis revealed the presence of fungi in 58.8% of samples, and assessment of AFs by chromatographic methods revealed that 52.9% were contaminated by AFs. AFB1 was the principal component in all AF-contaminated samples, with a mean level of 14.0, and 1.08 µg/kg in peanut and cashew nut, respectively. Eleven samples (32.4%) exceeded the total AF maximum level (4 μg/kg) and 8 samples (23.5%) exceeded the AFB1 (2 μg/kg) established by the European Commission. The objectives of this work were to formulate two gluten-free cookies added with iron, peanut or almond, and analyze their physicochemical, chromatic and sensory properties. A novel hybrid identification protocol was developed for F0:1 peanut seeds resulting from crosses between normal oleate cultivars with wild type FAD2B gene and high oleate genotypes with an A insertion in FAD2B gene. Presence of a series of overlapped peaks in trace file of the PCR product amplified with bF19/R1 primers was an indication of hybridity. This protocol may facilitate high oleate breeding and genetic studies in peanut.

Simple method to prepare DNA templates from a slice of peanut cotyledonary tissue for Polymerase Chain Reaction Electron. J. Biotechnol.

An efficient DNA extraction method was developed for peanut seed, where only 3-5 mg cotyledonary tissue was enough for more than 50 PCR reactions with a reaction volume of 15 &#956;l. Both low copy number and high copy number DNA sequences were successfully amplified. Processing one seed sample only took about half an hour. Sampling had no significant effects on germination and development. The DNA extraction method makes it possible to identify transformants and conduct molecular marker studies prior to sowing, and thus may greatly hasten research progress.

Cloning and analysis of a NBS-LRR disease resistance gene candidate PnAG1 from peanut (Arachis hypogaea L.) Electron. J. Biotechnol.

Background: Based on the conserved sequences of a known NBS resistance gene, a pair of degenerate primers was designed to amplify the NBS-LRR resistance gene from peanut using PCR and RACE methods. Results: Analyzing the amino acid sequence by BLAST on NCBI, which was deduced from the 1088bp-long gene named PnAG1-2, showed that it had a certain homology with some resistance proteins, among which Arachis cardenasii resistance protein gene had the highest homology (66%). Peanut is an important legume crop across the world. However, in contrast to most legume crops, groundnut lacks taxonomic proximity to any major model genome. A relatively large number of genomic sequences were generated from groundnut as part of a microsatellite marker development project. In the current study, a total of 1312 sequences were analyzed of which 448 contained microsatellite motifs. Identification of chilling-responsive transcripts in peanut (Arachis hypogaea L.) Electron. J. Biotechnol.

To isolate differentially expressed peanut genes responsive to chilling, a suppression subtractive hybridization (SSH) cDNA library was constructed for a chilling tolerant peanut cultivar A4 with mRNAs extracted from the seeds imbibed at 2ºC and 15ºC, respectively, for 24 hrs. A total of 466 cDNA clones were sequenced, from which 193 unique transcripts (73 contigs and 120 singlets) were assembled. Screening of peanut seeds resulting from 0.39% sodium azide treatment with NIRS calibration equation for bulk seed samples identified a plant with more than 60% oleate. Oleate content in individual seeds of the plant, as predicted by NIRS calibration equation for intact single peanut seeds, ranged from 50.05% ~ 68.69%. Three seeds with >60% oleate thus identified were further confirmed by gas chromatography. Multiple sequence alignments of the FAD2B gene from Huayu 22 (wild type) and peanut seeds with elevated oleate (mutant type) revealed a C281T transition in the coding region causing an I94T substitution in the oleoyl-PC desaturase, which may be responsible for reduction in the enzyme activity.

Amplified Fragment Length Polymorphism (AFLP) was used to establish the genetic relationships among 20 species from seven of the nine sections of genus Arachis. The level of polymorphism among nine accessions of the cultivated peanut, A. hypogaea L., was also evaluated. Three combinations of primers were used to amplify the AFLPs. The fragments were separated in 6% denaturing acrylamide gels. A total of 408 fragments were analyzed.
