The effects of storage temperature on the quality and phenolic metabolism of whole and minimally processed kale leaves Agronomy

We studied the effects of storage temperature on the activities of phenylalanine ammonia-lyase (PAL), peroxidase (POD) and polyphenol oxidase (PPO) in minimally processed kale (Brassica oleracea var. acephala) that was stored for 15 and 9 days at 5 ± 1ºC and 10 ± 1ºC, respectively. The main visual evidence for quality loss in whole leaves was yellowing and loss of turgescence. Minimally processed leaves presented significant browning, indicating increased POD and PPO activities. The PAL activity in minimally processed leaves stored at 5ºC was fourfold higher than that of whole leaves after two days of storage. Efeito comparativo do calor, S0(2) e ácido ascórbico na atividade da polifenol oxidase e peroxidase de algumas frutas e hortaliças Anais da ESALQ

O objetivo deste trabalho foi o de comparar o efeito do calor, do SO2 e do ácido ascorbico, visando determinar qual o método mais eficiente para controlar o escurecimento enzímico de cada uma das frutas e hortaliças estudadas. Os resultados mostraram que para a banana, pêssego, maça, cenoura, couve-flor e palmito, o calor foi o melhor agente inativador do sistema enzímico responsável pelo escurecimento. O SO2 foi mais eficiente para a pera, e para o figo e batata, o melhor agente inibidor foi o ácido ascórbico.

Caracterização preliminar de Amaranthus hybridus L. e Amaranthus viridis L. através de isoenzimas Anais da ESALQ

Através da técnica de eletroforese de gel de amido com migração horizontal, pode-se observar que para esterase, apenas o Amaranthus hybridus tipo verde apresentou variação, na forma de presença ou ausência da banda na posição 0.7. Por outro lado, para peroxidase, para todas as espécies e biótipos (A.viridis e A.hybridus tipo verde e tipo roxo) houve variação no padrão de bandas. Através da existência de bandas comuns às duas espécies, pode-se detectar evidências de hibridação e introgressãc entre elas.

The influence of the allelochemicals ferulic (FA) and vanillic (VA) acids on peroxidase (POD, EC 1.11.1.7) and phenylalanine ammonia-lyase (PAL, EC 4.3.1.5) activities and their relationships with phenolic acid (PhAs) contents and root growth of soybean (Glycine max (L.) Merr.) were examined. Three-day-old seedlings were cultivated in nutrient solution containing FA or VA (0.1 to 1 mM) for 48 h. Both compounds (at 0.5 and 1 mM) decreased root length (RL), fresh weight (FW) and dry weight (DW) and increased PhAs contents. Effect of Blanching on Enzyme Activity and Bioactive Compounds of Blackberry BABT

ABSTRACT The inactivation of peroxidase and polyphenol oxidase and the degradation of phenolic compounds, anthocyanins, and antioxidant activity in blackberry were investigated during blanching in water at 80-90 °C, and in steam, from 0 to 10 minutes. The enzyme kinetics indicated the presence of two isoenzymes with different thermal stability, following two-phase model, in which the reaction rate constant increased with increasing temperature for both the heat labile and heat resistant component. The phenolic compounds also followed a biphasic pattern, indicating a more heat resistant fraction, and the rate constants increased with increasing temperature. Interference of a commercial catalase preparation in laccase and peroxidase activities BABT

The influence of commercial catalase preparations (fungal and bovine origin) on laccase and peroxidase activity assays was evaluated using enzymatic extracts obtained from several basidiomycetes grown under different culture conditions. No hydrogen peroxide was detected in the extracts. Inhibition of laccase activity by 40 to 80% was related to the catalase source. In addition, oxidation of the substrate (ABTS) by fungal catalase in the absence of the enzymatic extract from basidiomycetes was observed. The results demonstrated the need for the evaluation of interference of the commercial catalase preparation when its use was required in the reaction mixture.

Cloning of a peroxidase gene from cassava with potential as a molecular marker for resistance to bacterial blight BABT

Cassava bacterial blight (CBB), caused by Xanthomonas axonopodis pv. manihotis, is considered one of the most important bacterial diseases of cassava (Manihot esculenta Crantz). In order to characterize the cassava genes involved in resistance to this disease, a genomic clone of a cationic peroxidase gene, MEPX1, was isolated by PCR from cassava cultivar MCOL 22. The DNA sequence of MEPX1 showed high homology with other plant peroxidase genes and contained a large intron typical of peroxidase genes. The predicted translation product showed a heme-ligand motif, also a characteristic of peroxidases, as well as phosphorylation, myristoylation and glycosylation sites. Biochemical and morphological responses to abiotc elicitor chitin in suspension-cultured sugarcane cells BABT

Cells of Saccharum officinarum submitted to hydrolyzated chitin for 1 to 8h produced phenolic compounds. These alterations were observed through cytochemical methods using Toluidine Blue and Phloroglucinol/HCl. After 4 h, besides cell wall change, there was a change in nuclear pattern of chitin treated cells. There was a 96% increase in nuclear area in 6 h chitin treated material, as observed by Feulgen reaction. The treated cells showed chromatin compacted regions and a degeneration process of nucleoli. Activity of enzymes associated with the enzymatic browning of minimally processed potatoes BABT

The purpose of the present study was to evaluate the effect of different potato cultivars and storage temperatures on the specific activity of phenylalanine ammonia-lyase (PAL), polyphenol oxidase (PPO) and peroxidase (POD) in minimally processed potatoes. Potato cultivars Agata, Asterix and Monalisa were selected, washed, peeled, diced, sanitized, centrifuged, vacuum- packed and stored at 5 and 15°C for 9 and 5 days, respectively. Naringenin inhibits the growth and stimulates the lignification of soybean root BABT

The flavanone naringenin, an intermediate in flavonoid biosynthesis, was tested for its effect on root growth, phenylalanine ammonia-lyase (PAL) and peroxidase (POD) activities, as well as phenolic compounds and lignin contents in soybean (Glycine max L. Merrill) seedlings. Three-day-old seedlings were cultivated in half-strength Hoagland nutrient solution (pH 6.0), with or without 0.1 to 0.4 mM naringenin in a growth chamber (25°C, 12-h photoperiod, irradiance of 280 µmol m-2 s-1) for 24 h. Cytokinin Dehydrogenase activity in primary roots and characterization of primary metabolites from leaves and rootlets of Ricinus Communis BABT

The aim of this study was to determine the activity of cytokinin dehydrogenase (CKX) and to measure other biochemical components in the primary leaves and radicles of castor seedlings (BRS Energia) in the initial phase of growth. The crude protein extract obtained after a 1-h extraction from the root tissues of seedlings showed no detectable CKX enzymatic activity when incubated with the substrate isopentenyl adenine for 1 h. However, after precipitation with ammonium sulfate at 70% saturation, the pellet showed CKX activity. The peroxidase enzyme activity was higher in the leaves than in the radicles. The shiitake mushroom (Lentinula edodes) has been used in research involving the prevention and the control of human and plant diseases. In cucumber plants, treated with aqueous extracts from mushroom basidiocarps, there was a reduction in anthracnose severity caused by Colletotrichum lagenarium, and an increase in peroxidase activity in the leaves. Lignin degradation, ligninolytic enzymes activities and exopolysaccharide production by Grifola frondosa strains cultivated on oak sawdust BJM

Fourteen strains of Grifola frondosa (Dicks.) S. F. Gray, originating from different regions (Asia, Europe and North America) were tested for lignin degradation, ligninolytic enzyme activities, protein accumulation and exopolysaccharide production during 55 days of cultivation on oak sawdust. Lignin degradation varied from 2.6 to7.1 % of dry weight of the oak sawdust substrate among tested strains. The loss of dry matter in all screened fungi varied between 11.7 and 33.0%, and the amount of crude protein in the dry substrate varied between 0.94 to 2.55%. In vitro effects of triacontanol (TRIA) on antioxidant enzymes and photosynthetic characteristics were studied in Arachis hypogaea L. cultivars (M-13 and PBS24030). The in vitro impact of TRIA on multiplication potential was also evaluated, which was found to be best at 2.0 ml L-1 TRIA in combination with 6- Benzyl adenine (BA, 3 mg L-1) in both the groundnut cultivars. The role of arbuscular mycorrhizal fungi (AMF) on cadmium (Cd) accumulation and on the possible attenuation of Cd stress was studied in maize plants (Zea mays L. var. Exceller). Plants inoculated or not with Glomus macrocarpum were exposed to Cd (0-20 µmol L-1), at two P levels (5 and 10 mg L-1) in the nutrient solution. The experiment was conducted in a hydroponic system, using a randomized 2 x 2 x 2 factorial design. The mycorrhiza-Cd interaction on plant growth, nutrients and Cd accumulation, AMF root colonization and on extra-radical mycelium was investigated. Jasmonates are a class of plant hormones that mediate various aspects in gene and metabolic regulation, defense, stress responses, reproduction and, possibly, communication. Changes in the antioxidative enzyme activities and lipid peroxidation in wheat seedlings exposed to cadmium and lead stress Braz. J. Plant Physiol.

Wheat seedlings were grown in presence of CdCl2 (0-200 µM) and Pb(NO3)2 (0-2000 µM) separately. The growth of metal-treated seedlings was significantly depressed. The activities of antioxidative enzymes namely superoxide dismutase (SOD), catalase (CAT) and guaiacol peroxidase (POX) were altered both in root and shoot tissues of 7-d-old seedlings. Response to chitin in suspension-cultured Citrus aurantium cells Braz. J. Plant Physiol.

Morphological changes and enzyme activity in suspension-cultured cells of Citrus aurantium were followed after chitin elicitation. Chitin oligomers presented a rapid effect with a maximum activity after 3 h followed by a gradual decrease to the original levels at 8 h of incubation. Cell walls presented a lignification process and the cytoplasmatic protein became less reactive to anionic stains. In this study, proteins extracted from laticifer cells of three plants were examined by electrophoresis, mass spectrometry (MALDI-TOF) and characterized in respect of proteolytic, chitinolytic and anti-oxidative activities by means of zymography and colorimetric assays. Acidic proteins with molecular masses between 12.5 and 74.5 kDa predominated in laticifers of P. rubra. This profile was not found in laticifers of C. grandiflora and E. tirucalli. The later was poor in respect of proteins. Strong anti-oxidative activity of superoxide dismutase (E.C. 1.15.1.1) was detected in P. rubra and C. grandiflora latices, and to a lesser extent ascorbate peroxidase (E.C. 1.11.1.1) and isoforms of peroxidase were seen.
