Abstract Abstract: Isolation and screening of new fungal strains from extreme and understudied environments, such as caves, is a promising approach to find higher yields enzyme producers. Cellulolytic fungal strains isolated from a Brazilian cave were evaluated for their enzymatic production after submerged (SmF) and solid-state fermentation (SSF). After SmF, three strains were selected for their high enzymatic activities: Aspergillus ustus for endoglucanase (4.76 U/mg), Talaromyces bruneus for β-glucosidase (11.71 U/mg) and Aspergillus sp. Optimization of extraction parameters for recovery of alpha-amylase from the fermented bran of Bacillus circulans GRS313 BABT

The extraction of alpha-amylase in the solid state fermentation of wheat bran by Bacillus circulans GRS313 was optimized. Among various solvents tested, maximum extraction was achieved when 2.5% (v/v) glycerol was added. The optimum conditions for extraction were 2.5 hrs soaking time at 30° C under recirculation and agitated condition, which yielded 590 U/g bran of enzyme activity. Whereas under stationary soaking condition the activity of the enzyme was 439.58U/g bran. With repeated wash under the above optimum conditions showed that 600 U/g and 184.2 U/g of enzyme extracted in the 1st and 2nd washes respectively and only 36.84 U/g was recovered in the 3rd wash

Evaluation of apple pomace based reconstituted feed in rats after solid state fermentation and ethanol recovery BABT

To utilize apple pomace in an economical and effective way, a feed was developed by solid state fermentation (SSF) using sequential interactive co-culture of Candida utilis and Kloeckera. Removal of ethanol and drying of the left - over residue. Feeding trial was conducted in white albino rats before and after reconstitution of apple pomace feed in the choice and no choice study. Feeding of apple pomace feed in the rats before reconstitution indicated that neither in fermented nor in unfermented form it was acceptable. Production of spores of Trichoderma harzianum on sugar cane molasses and bagasse pith in solid state fermentation for biocontrol BABT

Solid state fermentation was carried out for the production of spores from Trichoderma harzianum No 53 using sugar cane bagasse pith as solid matrix and sugar cane molasses as carbon and energy source. Different nitrogen sources such as urea, (NH4)2SO4 , NH4H2PO4 and (NH4)2HPO4 were added in the media to test their effect on spores production. Among these, urea was found most useful that resulted high no of spores (1x10(9)/gDM). Solid-state fermentation of Acanthogobius hastaprocessing by-products for the production of antioxidant protein hydrolysates with Aspergillus oryzae BABT

Functional properties and antioxidative activity of a protein hydrolysate prepared from Acanthogobius hasta processing by-product protein during solid-state fermentation with Aspergillus oryzae were investigated. Overall, protease activity increased with the degree of hydrolysis (DH) decreased during solid-state fermentation. All the protein hydrolysate had excellent solubility, possessed interfacial properties, and varying degrees of antioxidant activity which were governed by their concentrations and DH, molecular weight distribution and amino acid composition. Microbial production of citric acid BABT

Citric acid is the most important organic acid produced in tonnage and is extensively used in food and pharmaceutical industries. It is produced mainly by submerged fermentation using Aspergillus niger or Candida sp. from different sources of carbohydrates, such as molasses and starch based media. However, other fermentation techniques, e.g. solid state fermentation and surface fermentation, and alternative sources of carbon such as agro-industrial residues have been intensively studied showing great perspective to its production. This paper reviews recent developments on citric acid production by presenting a brief summary of the subject, describing micro-organisms, production techniques, and substrates, etc.

Purification and characterization of an anti-cancer enzyme produced by marine Vibrio Costicola under a novel solid state fermentation process BABT

L - Glutaminase, a therapeutically and industrially important enzyme, was produced from marine Vibrio costicola by a novel solid state fermentation process using polystyrene beads as inert support. The new fermentation system offered several advantages over the conventional systems, such as the yield of leachate with minimum viscosity and high specific activity for the target product besides facilitating the easy estimation of biomass. The enzyme thus produced was purified and characterised. It was active at physiological pH, showed high substrate specificity towards L - glutamine and had a Km value of 7.4 x 10-2 M. It also exhibited high salt and temperature tolerance indicating good scope for its industrial and therapeutic applications.

Optimization of thermostable α- amylase production by Streptomyces erumpens MTCC 7317 in solid-state fermentation using cassava fibrous residue BABT

Production of α- amylase under solid state fermentation by Streptomyces erumpens MTCC 7317 was investigated using cassava fibrous residue, one of the major solid waste released during extraction of starch from cassava (Manihot esculenta Crantz). Response surface methodology (RSM) was used to evaluate the effect of the main variables, i.e., incubation period (60 h), moisture holding capacity (60%) and temperature (50(0)C) on enzyme production by applying a full factorial Central Composite Design. Enhanced lipase recovery through RSM integrated differential evolutionary approach from the fermented biomass BABT

The aim of this work was to apply a modeling integrated optimisation approach for a complex, highly nonlinear system for an extracellular lipase extraction process. The model was developed using mutation, crossover and selection variables of Differential Evolution (DE) based on central composite design of Response Surface Methodology. The experimentally validated model was optimized by DE, a robust evolutionary optimization tool. Copper improves the production of laccase by the white-rot fungus Pleurotus pulmonarius in solid state fermentation BABT

Pleurotus pulmonarius (Fr) Quélet, produced laccase as the main ligninolytic enzyme when cultivated on solid-state cultures using corn cob as substrate. The addition of copper greatly increased the production of enzyme. The addition of 25.0 mM CuSO4 increased the level of laccase from 270 to 1,420 U.L-1 and the fungus showed high resistance to copper under the conditions used in this work.

Solid State production of manganese peroxidases using arecanut husk as substrate BABT

The lignocellulosic biomass from arecanut husk (Areca catechu Linnaeus) was evaluated as a new substrate for cultivation of Phanerochaete chrysosporium and Phanerochaete sp for solid state fermentation of manganese peroxidase (MnP). Arecanut had a moisture content of 79.84 % for ripe nut husk whereas green nut husk had 68.39 % moisture and a pH of 5.0, 3.0 and 7.0 for raw, ripe and dry husk. Reducing sugar content was 14.31, 19.21 and 1.77(mg/g of husk) for raw, ripe and dry nut husk, respectively. Non reducing sugar was 1.04(mg/g of husk) for raw and 0.68 (mg/g of husk) for dry husk. Bioethanol production from sweet potato (Ipomoea batatas L.) flour using co-culture of Trichoderma sp. and Saccharomyces cerevisiae in solid-state fermentation BABT

The aim of this work was to study the optimiation of co-culturing of Trichoderma sp. and Saccharomyces cerevisiae (1:4 ratio) on sweet potato (Ipomoea batatas L.) flour (SPF) for the production of bio-ethanol in solid-state fermentation (SSF). A comparison of methods to determine tannin acyl hydrolase activity BABT

Six methods to determine the activity of tannase produced by Aspergillus niger Aa-20 on polyurethane foam by solid state fermentation, which included two titrimetric techniques, three spectrophotometric methods and one HPLC assay were tested and compared. All methods assayed enabled the measurement of extracellular tannase activity. However, only five were useful to evaluate intracellular tannase activity. Studies on the effect of pH on tannase extraction demonstrated that tannase activity was considerably under-estimated when its extraction was carried out at pH values below 5.5 and above 6.0. Results showed that the HPLC technique and the modified Bajpai and Patil methods presented several advantages in comparison to the other methods tested.

Recovery of phytase produced by solid-state fermentation on citrus peel BABT

The extraction of phytase produced by solid-state fermentation of citrus peel was studied employing a multistage leaching process. It was observed that the extracts containing EDTA retained over 90% of phytase activity at room temperature after 24 h after the leaching. A fractional design 2² (with 4 replicates at the central point) was carried out for testing the pH and agitation as process independent factors. Only the interaction between the pH and agitation showed a significant influence. These factors were optimized with a central composite design. Investigations were carried out to optimize the culture conditions for the production of b-D-fructan-fructanohydrolase by an indigenously isolated bacterial culture of Staphylococcus sp. RRL8. Comparative study of a new alkaline L-methioninase production by Aspergillus ustus AUMC 10151 in submerged and solid-state fermentation BABT

Twenty four fungal species were screened for their ability to produce alkaline L-methioninase on methionine-glucose liquid medium. Aspergillus ustus AUMC 10151 displayed the highest yield of enzyme (10.8 U/mg protein), followed by A. ochraceus and Fusarium proliferatum. Upon optimization of the submerged fermentation (SmF)conditions, the maximum enzyme yield (18.23 U/mg protein) was obtained on a medium containing L-methionine (0.5%), sucrose (0.95%), KH2PO4 (0.1%) and 175 rpm. Bioconversion of biomass: a case study of ligno-cellulosics bioconversions in solid state fermentation BABT

Lignocellulosic residues obtained from crops cultivation form useful sources to be used as substrate for bioconversion processes. Sugarcane bagasse, which is a complex substrate obtained from the processing of sugar cane, is an important biomass among such sources. Due to its abundant availability, it can serve as an ideal substrate for microbial processes for the production of value added products. This paper reviews recent developments on biological processes developed on production of various products in solid state fermentation using sugarcane bagasse as the substrate and describes production of protein enriched feed, enzymes, amino acid, organic acids and compounds of pharmaceutical importance, etc. through microbial means.

ABSTRACT Nidularium procerum and Nidularium innocentii (Bromeliaceae) were cultivated in vitro on media supplemented with different sources and levels of GA3 (gibberellic acid). These sources were the commercial powder (analytical degree) and fermented extract obtained by Fusarium moniliforme via solid state fermentation. The in vitro elongation and rooting of these plants were evaluated after 50 days of cultivation. The GA3 present in the fermented extract possess the same effect of purified GA3 (analytical degree) for the increase of the height of aerial part of shoots of N. innocentii, but not for the N. procerum being the GA3 fermented extract in a lesser degree.
