Effect of alcohol and formaldehyde on the δ13C and δ15N isotopic composition of Plagioscion squamosissimus and Hypophthalmus edentates (Pisces, Osteichthyes) - doi: 10.4025/actascibiolsci.v33i4.70 Biological Sciences
Manetta, Gislaine Iachstel; UEM; Benedito, Evanilde; UEM, Maringá; Ducatti, Carlos; UNESP.
The present study investigates the δ13C and δ15N isotopic composition in frozen samples (control), samples in alcohol and in formaldehyde of Plagioscion squamosissimus and Hypophthalmus edentates. From each individual we extracted a strip of muscle from the region above the lateral line, in the dorsal fin base, that was divided into three equal parts, each one was submitted to one type of treatment: freeze – control group (-15oC), conservation in alcohol 70% and fixation in formaldehyde 4%. Samples were kept under those treatments for 30 days, washed and submerged in distilled water for 4 hours. Afterwards, they were dried up in air oven at 60oC for 48 hours and macerated until the obtaining of a fine powder. A significant difference...

15N-labed glycine synthesis Anais da ABC (AABC)
Tavares,Claudinéia R.O.; Bendassolli,José A.; Coelho,Fernando; Sant'ana Filho,Carlos R.; Prestes,Clelber V..
This work describes a method for 15N-isotope-labeled glycine synthesis, as well as details about a recovery line for nitrogen residues. To that effect, amination of alpha-haloacids was performed, using carboxylic chloroacetic acid and labeled aqueous ammonia (15NH3). Special care was taken to avoid possible 15NH3 losses, since its production cost is high. In that respect, although the purchase cost of the 13N-labeled compound (radioactive) is lower, the stable tracer produced constitutes an important tool for N cycling studies in living organisms, also minimizing labor and environmental hazards, as well as time limitation problems in field studies. The tests were carried out with three replications, and variable 15NH3(aq) volumes in the reaction were used...

13CO2 recovery fraction in expired air of septic patients under mechanical ventilation BJMBR
Martins,M.A.; Coletto,F.A.; Martins-Filho,O.A.; Marchini,J.S.; Basile-Filho,A..
The continuous intravenous administration of isotopic bicarbonate (NaH13CO2) has been used for the determination of the retention of the 13CO2 fraction or the 13CO2 recovered in expired air. This determination is important for the calculation of substrate oxidation. The aim of the present study was to evaluate, in critically ill patients with sepsis under mechanical ventilation, the 13CO2 recovery fraction in expired air after continuous intravenous infusion of NaH13CO2 (3.8 µmol/kg diluted in 0.9% saline in ddH2O). A prospective study was conducted on 10 patients with septic shock between the second and fifth day of sepsis evolution (APACHE II, 25.9 ± 7.4). Initially, baseline CO2 was collected and indirect calorimetry was also performed. A primer of 5 mL...

Obese women on a low energy rice and bean diet: effects of leucine, arginine or glycine supplementation on protein turnover BJMBR
Marchini,J.S.; Lambertini,C.R.; Ferriolli,E.; Dutra de Oliveira,J.E..
This study examined if leucine, arginine or glycine supplementation in adult obese patients (body mass index of 33 ± 4 kg/m²) consuming a Brazilian low energy and protein diet (4.2 MJ/day and 0.6 g protein/kg) affects protein and amino acid metabolism. After four weeks adaptation to this diet, each subject received supplements of these amino acids (equivalent to 0.2 g protein kg-1 day-1) in random order. On the seventh day of each amino acid supplementation, a single-dose 15N-glycine study was carried out. There were no significant differences in protein flux, synthesis or breakdown. The protein flux (grams of nitrogen, gN/9 h) was 55 ± 24 during the nonsupplemented diet intake and 39 ± 10, 44 ± 22 and 58 ± 35 during the leucine-, glycine- and...

Phloematic mobility of 10Boron in kiwifruit (Actinidia deliciosa) mixedshoots Ciencia e Investigación Agraria
Sotomayor,Carlos; Ruiz,Rafael; Muñoz,Luis.
Boron (B) is an essential micronutrient showing restricted mobility in plants, except in species that synthesize polyols. The ability of kiwifruit to mobilize B through phloem transport has not yet been confirmed, despite its ability to synthesize the polyol myo-inositol. This study examined kiwifruit plants in which boric acid enriched with the stable 10B isotope was applied to the distal leaves of mixed shoots with flowers. At 24, 72 and 144 h, both 10B-treated and un-treated leaves and flowers from treated shoots were sampled and subsequently analyzed via mass spectrometry to determine the resulting 11B/10B ratios. Control leaves and flowers showed a natural ratio, varying between 3.82 and 4.05. In contrast, the ratios in treated leaves were 1.57 at 24...

Prey preferences of adult sea bass Dicentrarchus labrax in the northeastern Atlantic: implications for bycatch of common dolphin Delphinus delphis ArchiMer
Spitz, Jerome; Chouvelon, Tiphaine; Cardinaud, M.; Kostecki,; Lorance, Pascal.
In the northeastern Atlantic, adult sea bass (Dicentrarchus labrax) is one of largest fish living on the shelf, and this species has important commercial value. However, pelagic trawl fisheries that target sea bass have negative operational interactions with common dolphins (Delphinus delphis). Our goal was to determine the diet of adult sea bass in the Bay of Biscay from stomachcontent and stable-isotope analyses, and explore the dietary overlap between sea bass and common dolphins. We found that sea bass primarily target small pelagic fish, most notably mackerel (Scomber scombrus), scads (Trachurus spp.), anchovy (Engraulis encrasicolus), and sardine (Sardina pilchardus). These four species also dominated the diets of common dolphins. This overlap in...

Shift in benthic assemblages and organisms’ diet at salmon farms: community structure and stable isotope analyses ArchiMer
Callier, Myriam; Lefebvre, Sebastien; Dunagan, Mary K.; Bataille, Marie-paule; Coughlan, Jennifer; Crowe, Tasman P..
The extent of the influence of salmon farming on the environment and on the uptake of particulate and dissolved effluents by benthic organisms was assessed using community structure and stable isotope analyses. Sediment cores were collected in 2 directions: perpendicular and parallel to the main residual current, 0, 25 and 200 m from 2 salmon farms (Millstone and Cranford) located in Mulroy Bay, Ireland. In addition, artificial substrates were placed for 2 mo at 1 m depth 0, 25 and 200 m from one farm to trace the uptake of farm-related nutrients by fouling organisms. The extent of measurable change in benthic communities (abundance, diversity, structure, trophic composition) depended on residual current direction. Intraspecific variation in isotopic...

Origine et devenir de la matière organique au sein du micro-estuaire temporaire de Fiume Santu (L’Agriate, Haute-Corse) ArchiMer
Massey, Jean-laurent.
In relation with meteorological, hydrological and topographic characteristics, Corsica has a very large number of temporarily open/closed micro-estuaries. Agriates contains several of them, including the Fiume Santu’s temporarily micro-estuary. These ecosystems are specific because of their sporadic connection with the sea. The opening and closure depends on both the river flow and the presence of the Posidonia oceanica (L.) Delile “banquettes”. Characterization of the global functioning was based on a multiparameter approach (particulate matter and dissolved inorganic matter descriptors, carbon and nitrogen stable isotope, phytoplankton communities), in order to qualify and quantify the organic matter (OM), along an estuary – coastal zone gradient. The OM...

A Multi-Elements Isotope Approach to Assess the Geographic Provenance of Manila Clams (Ruditapes philippinarum) via Recombining Appropriate Elements ArchiMer
Won, Eun-ji; Kim, Seung Hee; Go, Young-shin; Kumar, K. Suresh; Kim, Min-seob; Yoon, Suk-hee; Bayon, Germain; Kim, Jung-hyun; Shin, Kyung-hoon.
The increasing global consumption of seafood has led to increased trade among nations, accompanied by mislabeling and fraudulent practices that have rendered authentication crucial. The multi-isotope ratio analysis is considered as applicable tool for evaluating geographical authentications but requires information and experience to select target elements such as isotopes, through a distinction method based on differences in habitat and physiology due to origin. The present study examined recombination conditions of multi-elements that facilitated geographically distinct classifications of the clams to sort out appropriate elements. Briefly, linear discriminant analysis (LDA) analysis was performed according to several combinations of five stable isotopes...

Influences of geothermal sulfur bacteria on a tropical coastal food web ArchiMer
Pascal, Pierre-yves; Dubois, Stanislas; Goffette, Anais; Lepoint, Gilles.
The activity of the geothermal plant at Bouillante in Guadeloupe (French West Indies) releases thioautotrophic bacteria into the coastal environment. Fish counts reveal that fish abundance increases with higher availability of this bacterial resource. In order to evaluate the trophic role of these bacteria, isotopic compositions (C, N, S) of potential consumers were evaluated on transects at increasing distance from the source of bacteria. The 3 mobile fish species examined (Abudefduf saxatilis, Acanthurus bahianus, and Stegastes partitus) ingested and assimilated chemosynthetic bacteria. Similarly, the isotopic composition of the mobile sea urchin Diadema antillarum was different close to the discharge channel, suggesting a diet mainly composed of sulfur...

Facing the necessity of describing estuarine ecosystems: a review of food web ecology study techniques ArchiMer
Pasquaud, Stéphanie; Lobry, Jeremy; Elie, Pierre.
Estuarine areas are sites of human pressures and degradation. In order to maintain and/or restore the quality of estuarine ecosystems, it is necessary to describe their structure and functioning. For that reason, many recent scientific works focus on food webs, which are depicted as being good indicators of the functioning of aquatic ecosystems. Hence it is necessary to question how estuarine food webs can be described. This paper proposes a pragmatic and practical review of the most widely used techniques (stomach/gut content analysis, stable isotope ratios and biochemical markers) with emphasis on their main advantages, drawbacks and bias according to possible ecological goals (ecological quality objectives). These approaches, although quite different,...

Relative contribution of natural productivity and compound feed to tissue growth in blue shrimp (Litopenaeus stylirostris) reared in biofloc: Assessment by C and N stable isotope ratios and effect on key digestive enzymes ArchiMer
Cardona, Emilie; Lorgeoux, Benedicte; Geffroy, Claire; Richard, Pierre; Saulnier, Denis; Gueguen, Yannick; Guillou, Gael; Chim, Liet.
The aim of this study was to assess the relative contribution of natural productivity and compound food to the growth of the juvenile blue shrimp Litopenaeus stylirostris reared in a biofloc system. Two experiments were carried out based on the same protocol with three treatments: clear water with experimental diet (CW), biofloc with experimental diet (BF) and biofloc unfed (BU). Shrimp survival was significantly higher in biofloc rearing than in CW rearing. The contribution of the biofloc to shrimp diet was estimated through measurement of carbon and nitrogen stable isotope ratios in shrimp and food sources. Different isotopic compositions between feeds were obtained by feeding natural productivity with a mixture rich in fish meal and the shrimps with a...

Trophic role of large benthic sulfur bacteria in mangrove sediment ArchiMer
Pascal, Pierre-yves; Dubois, Stanislas; Boschker, Henricus T. S.; Gros, Olivier.
Large filamentous sulfur-oxidizing bacteria belonging to the Beggiatoacae family can cover large portions of shallow marine sediments surrounding mangroves in Guadeloupe (French West Indies). In order to assess the importance of Beggiatoa mats as an infaunal food source, observations were conducted of the area within mats and at increasing distances from mats. We used natural isotopic compositions and a C-13 enrichment study. Both revealed an ingestion of bacterial mats by associated meiofauna, dominated by rotifers and to a smaller extent by small polychaetes and nematodes. Compared to adjacent sites, sediment covered by bacterial mats presented a higher abundance of diatoms, whereas the total biomass of bacteria did not vary. This constant bacterial...

Devenir des bactéries dans les réseaux trophiques des vasières intertidales : le cas de brouage (Baie de Marennes-Oléron) ArchiMer
Pascal, Pierre-yves.
Bacteria are highly abundant and productive in intertidal mudflat sediments. By analogy with pelagic system, it has been suggested that bacteria play an important role in benthic food web. Due to technical difficulties, numbers of studies dealing with benthic bacterivory are not numerous and give conflicting results. A method using community of 15N enriched bacteria as tracer was developed in order to measure grazing of meiofauna and macrofauna. Since enriched bacteria present characteristics of size, activity and diversity close to natural ones, bias due to selective bacterial ingestion by grazers are limited. Due to tidal cycles, intertidal mudflat constitutes highly variable environment at short time scale. Since those variations may influence...

Trophic relationships between the farmed pearl oyster Pinctada margaritifera and its epibionts revealed by stable isotopes and feeding experiments ArchiMer
Lacoste, Elise; Raimbault, Patrick; Harmelin-vivien, Mireille; Gaertner-mazouni, Nabila.
Bivalve cultures support a host of epibionts, mainly suspension feeders, which can compete for food resources with the cultivated bivalves. However, the magnitude of interspecific competition for food in bivalve aquaculture settings remains inconclusive, especially in tropical areas. We investigated the interactions for food between the farmed pearl oyster Pinctada margaritifera and its epibionts, using stable isotope analysis and feeding experiments. Inter-and intraspecific variations of delta C-13 and delta N-15 stable isotope ratios (SIRs) were determined for oysters in the presence or absence of epibionts. The diet of the most abundant epibionts, Herdmania momus and Didemnum sp., was specified using isotope measurements and flow cytometry during...

Characterization of food web structure of the upper continental slope of the Celtic Sea highlighting the trophic ecology of five deep-sea fishes ArchiMer
Kopp, Dorothee; Robert, Marianne; Pawlowski, Lionel.
In marine ecosystems, the study of trophic relationships has extensively benefited from the development of stable isotope analyses (SIA) as dietary tracers. SIA are particularly useful in elucidating the structure of deep sea food webs given the constraints involved in obtaining gut-content data from deep trawling. We used carbon and nitrogen stable isotope analyses and Stable Isotope Bayesian Ellipses in R (SIBER) and Stable Isotope Analysis in R (SIAR) routines, to determine the trophic ecology of five deep-sea fishes from the upper continental slope of the Celtic Sea. SIA made it possible to deduce some general tendencies in food-web structure and species trophic interactions and confirmed diet determined by gut-content analysis for the same species, in...

Bacterivory by benthic organisms in sediment : Quantification using 15N-enriched bacteria ArchiMer
Pascal, P.y.; Dupuy, C; Mallet, C; Richard, P; Niquil, N.
The fate of benthic bacterial biomass in benthic food webs is a topic of major importance but poorly described. This paper describes an alternative method for evaluation of bacterial grazing rate by meiofauna and macrofauna using bacteria pre-enriched with stable isotopes. Natural bacteria from the sediment of an intertidal mudflat were cultured in a liquid medium enriched with 15NH4Cl. Cultured bacteria contained 2.9% of 15N and were enriched sufficiently to be used as tracers during grazing experiments. Cultured bacteria presented a biovolume (0.21 μm3) and a percentage of actively respiring bacteria (10%) similar to those found in natural communities. The number of Operational Taxon Units found in cultures fluctuated between 56 and 75% of that...

Banc d’Arguin (BA), Mauritania, is a nationally protected shallow gulf > 10,000 km2 between the Sahara desert and the upwelling system off the Mauritanian coast. In the southeast, BA consists of a 500 km2 tidal flat, the most important wintering site for shorebirds using the East Atlantic Flyway. The Mauritanian upwelling-driven phytoplankton production supports the most productive fisheries worldwide, but little is known about its trophic role in the functioning of the inshore BA food web. Using stable isotopes as trophic tracers to distinguish between upwelling-driven phytoplankton, open ocean phytoplankton, and benthic primary producers, we assessed the spatial extent to which the inshore BA food web is fuelled by upwelling-driven phytoplankton...

Late Holocene surface ocean conditions of the Norwegian Sea (Voring Plateau) ArchiMer
Andersson, C; Risebrobakken, B; Jansen, E; Dahl, So.
Late Holocene sea surface ocean conditions of the eastern Norwegian Sea (Voring Plateau) are inferred from planktic stable isotopes and planktic foraminiferal assemblage changes in cores JM97-948/2A and MD95-2011 (66.97degreesN, 7.64degreesE). Strong covariance between the planktic stable oxygen isotopic record and abundance changes of N. pachyderma (sin) show that major changes in surface ocean conditions are reflected both in the geochemical composition of the foraminiferal tests as well as in the composition of the foraminiferal fauna. Surface ocean conditions warmer than present were common during the past 3000 years. During the so-called Medieval Warm Period, surface conditions were highly variable with marked changes in sea surface temperature. The...
