Characterization of a leucine aminopeptidase from Toxoplasma gondii OAK

The M17 family leucine aminopeptidase (LAP) hydrolyze amino acids from the N-terminus of peptides. Many LAPs from parasitic protozoa including Plasmmodium, Trypanosoma and Leishmania, have been intensely investigated because of their crucial roles in parasite biology. In this study, a functional recombinant Toxoplasma gondii LAP (rTgLAP) was expressed in Escherichia coli and its enzymatic activity against synthetic substrates for aminopeptidase, as well as the cellular localization was determined. Our results indicated that TgLAP is a functional aminopeptidase in the cytoplasma of T. gondii.

Purification and partial characterization of Phaseolus vulgaris seed aminopeptidase BJMBR

The aminopeptidase activity of Phaseolus vulgaris seeds was measured using L-Leu-p-nitroanilide and the L-aminoacyl-ß-naphthylamides of Leu, Ala, Arg and Met. A single peak of aminopeptidase activity on Leu-ß-naphthylamide was eluted at 750 µS after gradient elution chromatography on DEAE-cellulose of the supernatant of a crude seed extract. The effluent containing enzyme activity was applied to a Superdex 200 column and only one peak of aminopeptidase activity was obtained. he distribution and aminopeptidase activity of prokaryotes were investigated along a natural continuous salinity gradient in a hypersaline coastal lagoon, the Coorong, South Australia. The abundance of prokaryotes significantly increased from brackish to hypersaline waters and different sub-populations, defined by flow cytometry, were observed along the salinity gradient. The N-terminal amino acid sequence of an aminopeptidase from Japanese edible mushroom, Grifola frondosa, was reported to have high similarity with that of a serine proteinase from basidiomycete, Agaricus bisporous (Nishiwaki and Hayashi, 2001). The full-length cDNA and the corresponding genomic DNA of the enzyme were cloned, based on the reported N-terminal amino acid sequence. The predicted open reading frame (ORF) of the cloned cDNA, encoding a product of 379 amino acids, was expressed in E. coli using pET expression vector. It has been postulated that both digestive capacity and intestinal biochemical features are correlated to dietary habits in birds. Therefore, it would be expected to find biochemical constraint to hydrolyze sugars in those species, which predate exclusively on marine invertebrates. In vitro intestinal activities of these enzymes were studied in Cinclodes nigrofumosus (d' Orbigny) and Cinclodes patagonicus (Gmelling). Due to differences in dietary habits between species I predicted the lack of sucrase activity in C. nigrofumosus but not in C. patagonicus. Modificações na expressão gênica foram observadas nos sistemas esterase, leucina aminopeptidase e x-glicerofosfato desidrogenase, durante o desenvolvimento ontogenético de Anopheles albitarsis. A esterase revelou quatro regiões de atividade, sendo a esterase 1 detectada apenas em larvas de 4º estádio velhas e em pupas, as esterases 2 e 4 foram presentes durante todo o desenvolvimento, e a esterase 3 revelou-se praticamente apenas em larvas e raríssimas vezes em pupas. Dados sobre a leucina aminopeptidase durante o desenvolvimento ontogenético em Anopheles nuñez-tovarìevidenciaram seis zonae de atividade, eendo a LAP1, LAP2, LAP4 e LAP5 presentes nos estádios larvais, pupae e adultos e a LAP3 e LAP6 características desses dois últimos estágios. Diferenças na intensidade de coloração foram detectadas conforme o estágio considerado. Abstract The fermentation of cocoa seeds envolves microbial processes and the action of enzymes. To identify the possible differences in the cocoa fermentation process, with regards to proteolysis, this study has the objective of determining protease activity (under predetermined conditions) and its isoenzymes in two cocoa cultivars (PH-16 and HRT-1188) in different cocoa fermentation times, in addition to establishing the microbial load (molds and yeasts and aerobic mesophilic). Large yellow croaker is an important marine fish species for aquaculture in China. Larval rearing of this species is still based on live prey feeding. The aim of this study was to describe the onset and development of the main digestive enzymes during larval development of this species, in order to obtain essential data for the formulation of a compound diet adapted to larvae. Enzymatic assays were conducted from day I post hatching to day 40 in larvae fed the following live prey sequence: rotifers, Artemia nauplii, copepods, and then frozen copepods. The abundance and composition of microbial communities were investigated during 2007 in Marennes Oleron Bay (France) in order to characterize the biological relationships governing the planktonic food web.
