Long-term cryopreservation of basidiomycetes BJM
Linde,Giani Andrea; Luciani,Alana; Lopes,Ana Daniela; Valle,Juliana Silveira do; Colauto,Nelson Barros.
Abstract Basidiomycetes have several biotechnological and industrial applications such as enzyme production, bioremediation, pharmaceutical and functional food production. Due to climatic features, the preservation of several basidiomycetes is threatened, and to guarantee the preservation of this genetic resource, the development of long-term preservation techniques is necessary once there is no universal protocol for the cryopreservation of basidiomycetes. Cryopreservation is a technique in which microorganisms are submitted to ultralow temperatures. Therefore, this study aimed to collect information on the main conditions for long-term cryopreservation...

MODELING OF THE KINETICS OF CRYOGENIC FREEZING OF LIMA BEAN SEEDS AS A FUNCTION OF INITIAL WATER CONTENT REA
Luciano,Karina M da F.; Mata,Mario E. R. M. Cavalcanti; Fortes,Mauri; Duarte,Maria E. M..
ABSTRACT Cryopreservation is a technique used to preserve genetic material at ultra-low temperatures in cryogenic germplasm banks. In order to enable the cryopreservation of a seed, it is necessary to study its appropriate freezing temperature and its water content, because seeds reach maximum physiological quality with high water contents, and they tolerate drying when they are orthodox seeds, but they may not tolerate cryopreservation. Therefore, the objective of this study was to analyze the influence of initial water content (50 to 10% wet basis) on the kinetic behavior of the lima bean seed at the temperature of −150 °C, in addition to determining...

Riesco, Marta F.; Felix, Francisca; Matias, Domitilia; Joaquim, Sandra; Suquet, Marc; Cabrita, Elsa.
Sperm cryopreservation is a widely employed technique that promotes alternative techniques to contribute to broodstock management or restoration programs for species of commercial interest, endangered species or species with an interesting genotype. The preservation of genetic material from improved stocks or from the original population is extremely important for the oyster aquaculture industry to prevent the potential impacts of epidemic diseases and natural disasters. The Portuguese oyster, Crassostrea angulata, was the most important species commercialized by the shellfish industry. However, inadequate management of this industry and pathology occurrences resulted in a significant decrease in natural populations. For...

Short-term storage and cryopreservation of turbot (Scophthalmus maximus) sperm ArchiMer
Chereguini, Olvido; Cal, Rosa Maria; Dreanno, Catherine; Ogier De Baulny, Bénédicte; Suquet, Marc; Maisse, Gérard.
Short-term storage over several days as well as cryopreservation of turbot (Scophthalmus maximus) sperm were studied. Two extenders, Ringer 200 and artificial seminal liquid (ASL2), are suggested for semen collection in order to avoid the deleterious effect of urine contamination, and for the purpose of short-term storage between 0 and 15 degrees C. Oxygen atmosphere is not suitable for turbot sperm storage. Turbot spermatozoa undergo cryopreservation with a high rate of success especially in a sucrose solution with 10% dimethyl sulfoxide (DMSO) and 10% egg yolk.

Cryopreservation has an immunomodulating effect on tracheal tissue as a result of class II antigen depletion due to epithelium exfoliation. However, not all epithelium is detached. We evaluated the role of apoptosis in the remaining epithelium of 30 cryopreserved tracheal grafts. Caspase-3 immunoreactivity of tracheal epithelium was studied in canine tracheal segments cryopreserved with F12K medium, with or without subsequent storage in liquid nitrogen at -196°C for 15 days. Loss of structural integrity of tracheal mixed glands was observed in all cryopreserved tracheal segments. Caspase-3 immunoreactivity in tracheal mucosa and in mixed glands was significantly decreased, in contrast to the control group and to...

Orchid cryopreservation Ciência e Agrotecnologia
Vendrame,Wagner; Faria,Ricardo Tadeu de; Sorace,Mauren; Sahyun,Sandra Aparecida.
Orchids are lush and highly valuable plants due to their diversity and the beauty of their flowers, which increases their commercialization. The family Orchidaceae comprises approximately 35,000 species, distributed among more than 1,000 distinct genera and 100,000 hybrids, totaling approximately 8% to 10% of all flowering plants. With the advance of agriculture and the constant destruction of their natural habitat, orchid species are collected in an indiscriminate manner by collectors and vendors, and this extractive activity threatens many species with extinction, drastically reducing their genetic variability in nature. Therefore, it is essential to seek alternatives that make the preservation of such species feasible using techniques with low...

LONDE,LUCIANA C.N.; VENDRAME,WAGNER A.; SANAEI,MASSY; OLIVEIRA,ALEXANDRE B. DE.
ABSTRACT The preservation of banana genetic material is usually performed through seedlings. However, most banana cultivars do not produce seed and are propagated vegetatively. Therefore, cryopreservation is a feasible technique that allows the preservation of banana genotypes indefinitely. For the success of cryopreservation protocols, the selection of cryoprotectants and pre-freezing techniques are important factor. Therefore, the objective of this study was to verify the effects of different cryoprotectants with and without 1% phloroglucinol and pre-cooling periods on the development of a protocol for cryopreservation of in vitro rhizomes ofMusa...

Cryopreservation of sperm in marine fish ArchiMer
Suquet, Marc; Dreanno, Catherine; Fauvel, Christian; Cosson, J; Billard, R.
Since the first work of Blaxter in 1953, fish sperm cryopreservation has been attempted on about 30 marine species. The present paper reviews the techniques used and the results published in these species. Particular attention is paid to the handling procedure of sperm before freezing, the problems of semen ageing and semen contamination with urine. The quality of frozen-thawed semen was evaluated using previously standardized biotests, such as a two-step motility activation technique adapted for the different species and fertilization assays using a discriminating insemination technique. Most extenders used in marine fish are saline or sugar solutions. From the investigated cryoprotectants, dimethyl sulphoxide (DMSO)...
