Polyphenolic compounds are measured in carrots. Beside measurements of total phenolic contents (TPC) as a sum parameter, HPLC methods are developed and validated to detect the single constituents. The methods are applied for carrot samples from different field experiments as well as farming systems. The aim of the experiments is to test if: 1. carrot samples from different treatments can be differentiated 2. the sum parameters are correlated with the HPLC data (single constituents) 3. the application of multivariate statistics can help to classify the samples according to their treatment

Using 31-phosphorus nuclear magnetic resonance spectroscopy ((31)P-NMR) to characterize phosphorus (P) in animal manures and litter has become a popular technique in the area of nutrient management. To date, there has been no published work evaluating P quantification in manure/litter samples with (31)P-NMR compared to other accepted methods such as high performance liquid chromatography (HPLC). To evaluate the use of (31)P-NMR to quantify myo-inositol hexakisphosphate (phytate) in ileal digesta, manure, and litter from broilers, we compared results obtained from both (31)P-NMR and a more traditional HPLC method. The quantification of phytate in all samples was very consistent between the...

The identification of molecules present in plants by chromatographic techniques such as HPLC, can bring promising alternative to the control of plant diseases, since brassicas have glucosinolates, which act in plant defense. The objective of this work was to verify the presence of allyl isothiocyanate by HPLC (High Performance Liquid Chromatography Reverse Phase), using different methods of extraction. The plant material was leaves of Crambe abyssinica, dried at 45 °C for 48 h. The extraction methods used consisted of 200 mg L-1 of plant material for each method: infusion, aqueous solution, and cetonic, hydroalcoholic, methanol, hexane and chloroform solutions. The identification and...

The aim of this study was to detect and quantify the main residual monomers released from composites, using high performance liquid chromatography (HPLC). Discs were made with dental composites (Herculite XRV, Tetric Ceram and Filtek Z250) and immersed in deionized water at 37ºC for 28 days, with water changes in 1, 7, 14 and 21 days. The mean concentration of residual monomers were subject to the Kruskal-Wallis test (p<0.05). Tetric Ceram exhibited significantly higher concentrations of leached monomers. Bis-GMA was the monomer released in lower concentrations for all the materials. There was no statistical difference between the amounts of TEGDMA and UDMA. Most of the monomers demonstrated maximal concentration at the...

Deflazacort (DFZ) is a glucocorticoid used as an anti-inflammatory and immunosuppressant drug. No official methods are available for DFZ determination in pharmaceutical formulations. The objective of this study was to develop, validate and compare spectrophotometric (UV and colorimetric) and high-performance liquid chromatography (HPLC) methods, for the quantitative determination of DFZ in tablets and oral suspension. For the UV method, ethanol was used as the solvent, with detection at 244 nm. The colorimetric method was based on the redox reaction with blue tetrazolium in alkaline medium, with detection at 524 nm. The method by HPLC was carried out using a C18 column, mobile phase...

Rutin is a flavonoid glycoside, mainly consists of phenolic compounds, responsible for many biological activities. The objective of the present study was to develop and validate a precise, simple, robust, rapid and reliable reverse phase high -performance liquid chromatography (RP-HPLC) technique by using Qbd approach for evaluating the rutin in nanoparticles. Central composite design (CCD) was employed for optimizing the experimental conditions of RP-HPLC method. Buffer pH, methanol content in the mobile phase composition, flow rate, and wavelength were selected as independent variables whereas retention time, peak area, and asymmetry factor was selected as dependent variables. The...
