Seminal Plasma Proteins 17
P Perumal.
The ejaculated semen consists of two major components viz. sperm cells (spermatozoa) and the fluid part obtained after centrifugation called seminal plasma. The spermatozoa originate from the semniferous tubule and are suspended in the seminal plasma. The seminal plasma is composed of secretions contributed by the testis, epididymis, seminal vesicles, ampullae, prostate and bulbourethral glands. About 60-80 % of the ejaculated semen of the bull originates from these sources. Seminal plasma is a highly complex biological fluid containing proteins, amino acids, enzymes, fructose and other carbohydrates, lipids, major...

Development and validation of a simple and rapid way to generate low volume of plasma to be used in point-of-care HIV virus load technologies 55
Vasconcellos,Isabelle; Mariani,Diana; Azevedo,Marcelo C.V.M. de; Ferreira Jr.,Orlando C.; Tanuri,Amilcar.
ABSTRACT A new point-of-care HIV viral load, mPIMA HIV-1/2 VL, Abbott, USA, has been recently developed. This point-of-care viral load requires no skilled person to run and uses a small plasma volume (50 µL). However, obtaining 50 µL of plasma can be a challenge in limited resource settings. We validated a simple and easy method to obtain enough amount of plasma to run a point-of-care viral load. The study utilized 149 specimens from patients failing antiretroviral therapy. At least 250 µL of whole blood was collected in a microtube/EDTA from fingerstick (fs-plasma) and immediately centrifuged. Parallel collection of...

Spectrofluorimetric determination of amlodipine in human plasma without derivatization 60
Kadioglu,Yucel; Ozturk,Murat.
A rapid and sensitive spectrofluorimetric method was developed for the determination of amlodipine (AD), a calcium channel blocker, in the plasma. The type of solvent, the wavelength range, and the range of AD concentration were selected to optimize the experimental conditions. The calibration curves were linear (r² >0.997) in the concentration range of 0.1-12.5 ppm of AD. The limit of quantitation and limit of detection values for the method for plasma samples were 0.1 ppm and 0.07 ppm, respectively. The precision calculated as the relative standard deviation was less than 3.5%, and the accuracy (relative error) was better than 5.5% (n=6). The method developed in this study can be...

Seminal plasma components and their interactions with spermatic cells 195
de Souza, Fabiana Ferreira; Lopes, Maria Denise; Bicudo, Sony Dimas.
With the increasing use of artificial insemination techniques, little importance has been given to seminal plasma, since it is substituted by other diluters. However, natural sexual intercourse would not work without seminal plasma as diluter for spermatozoa in the epididimus. The study of seminal plasma brings important information for the research on diluters, which are made to substitute the forme. Thus, if the seminal plasma composition were known, it would be easier to preserve semen samples, specially those of species that present cryopreservation problems. Beside, knowledge on "fertility markers" that may be present...

Changes of Progesterone and Endothelin Concentrations in the Peripheral Plasma of Female Calves and Cycling Cows: Effects of PGF2α Injection 19
OHTANI, Masayuki; OKI, Naoko; TANIMURA, Miyune; KOBAYASHI, Shu-ichi; ACOSTA, Tomas J.; HAYASHI, Kanako; MIYAMOTO, Akio; 宮本, 明夫.
It is well known that an injection of prostaglandin F2α (PGF2α) or its analogue during the mid luteal phase of the estrous cycle induces a rapid decrease in plasma progesterone (P4) concentration, followed by luteolysis in the cow. There is evidence that a potent vasoactive peptide, endothelin-1 (ET-1), is produced in the bovine corpus luteum (CL), and that it is directly involved in luteolysis. We previously found that ET-1 concentrations in the peripheral plasma increase during the period of luteolysis and ovulation in cows. However, it is not clear whether the elevation of peripheral plasma ET-1 concentration observed during luteolysis and ovulation...

A new method for analysis of plasma carotenoids in poultry 45
Teixeira,M.; Meleiro,C.H.A..
A study was carried out to evaluate a new quantitative analytical technique for determination of plasma carotenoids in poultry. Plasma carotenoides were extracted and measured from 30 blood samples of male Cobb broilers. The new technique was efficient in detecting plasma carotenoids of chickens and also showed advantages upon those techniques currently in use such as the form of presentation of results, use of a compatible solvent, and the scanning method for reading. The new technique did not increase the cost of the analysis.

CRUZ,WANISE M. SOUZA; GUIMARÃES,SHEILA M.S.; MACIEL,GABRIELLE C.; HUGUENIN,ANA BEATRIZ A.; CARVALHO,MONICQUE E.M. DE; COSTA,BÁRBARA O.; SILVA,GEISIANE A. DA; COLAFRANCESCHI,ALEXANDRE S.; SCALCO,FERNANDA B.; RIBEIRO,MARCIA.
Abstract Coronary artery bypass grafting reduces plasma L-carnitine and may impair the production of myocardial energy. L-carnitine supplementation may elevate plasma L-carnitine and increase cardiac mechanical efficiency. The objective of this study was to verify the recovery of preoperative plasma LC in patients with heart failure undergoing coronary artery bypass grafting supplemented with a daily oral dose of 50 mg / kg. Volunteers with ischemic heart failure who underwent surgery were randomized into a supplemented group (A-received 50 mg / kg L-carnitine) or placebo group (B) for 60 days. Supplementation was started on the third postoperative day....

Platelets can be a biological compartment for the Hepatitis C Virus 58
Ariede,Jovita Ramos; Pardini,Maria Inês de Moura Campos; Silva,Giovanni Faria; Grotto,Rejane Maria Tommasini.
Although HCV has hepatic tropism, the presence of the virus in extra-hepatic compartments has been well documented. Platelets have been described as carriers of the virus in the circulation and may be a natural reservoir for the virus. However, few studies have been performed to evaluate the levels of HCV RNA in plasma and platelets are equal or differ in some way. Therefore, the aim of this study was to perform a comparative evaluation of the stability of HCV RNA in plasma and isolated platelets. Four aliquots of whole plasma obtained from patients infected with HCV were incubated at 37 °C for 0, 48, 96 and 144 h. After incubation, the

Evaluation of four different strategies to characterize plasma membrane proteins from banana roots 64
Garcia,Suzana Antunes Lourençoni; Panis,Bart; Swennen,Rony; Carpentier,Sebastien Christian.
Plasma membrane proteins constitute a very important class of proteins. They are involved in the transmission of external signals to the interior of the cell and selective transport of water, nutrients and ions across the plasma membrane. However, the study of plasma membrane proteins is challenging because of their poor solubility in aqueous media and low relative abundance. In this work, we evaluated four different strategies for the characterization of plasma membrane proteins from banana roots: (i) the aqueous-polymer two-phase system technique (ATPS) coupled to gelelectrophoresis (gel-based), and (ii) ATPS coupled to...

Fucoidan prevents concanavalin A-induced liver injury through induction of endogenous IL-10 in mice 19
Saito, Ako; Yoneda, Masashi; Yokohama, Shiro; Okada, Mitsuyoshi; Haneda, Masakazu; Nakamura, Kimihide; 中村, 公英.
Fucoidan is a complex of sulfated polysaccharides derived from non-mammalian origin such as marine brown algae and induces cytokine expression. We investigated the effect of fucoidan on concanavalin A (Con A)-induced liver injury in mice. Liver injury was induced by an intravenous injection of Con A (18.5 mg/kg). Various doses of fucoidan (1-30 mg/kg) were intravenously administered 30 min before Con A injection. The plasma alanine aminotransferase (ALT) and several cytokines levels were determined, and hepatic histological changes were also assessed. The effect of fucoidan administration by itself on induction of interleukin (IL)-10 in plasma and liver tissue was investigated. Con A...

Mimotopes selected by biopanning with high-titer HIV-neutralizing antibodies in plasma from Chinese slow progressors 55
Zhang,Xiaoli; Han,Xiaoxu; Dai,Di; Mingjia,Bao; Zhang,Zining; Zhang,Min; Bice,Tristan; Zhao,Min; Cao,Yaming; Shang,Hong.
OBJECTIVE: One approach to identifying HIV-1 vaccine candidates is to dissect the natural antiviral immune response in treatment-naïve individuals infected for over ten years, considered slow progressor patients (SPs). It is suspected that SP plasma has strongly neutralizing antibodies (NAb) targeting specific HIV viral epitopes. METHODS: NAbs levels of 11 HIV-1-infected SPs were detected by PBMC-based neutralization assays. To investigate SP NAb epitope, this study used a biopanning approach to obtain mimotopes of HIV-1 that were recognized by SP plasma NAbs. IgG was purified from hightiter NAb SP plasma, and used as the ligand for three rounds of...
