生乳中の酵母の動態を明らかにすることを目的として,帯広畜産大学と近郊の酪農家3戸で飼養している健康なホルスタイン種乳牛(体細胞数1×105cfu/ml以下)の乳頭表皮,分房乳,バルク乳およびローリー乳中の酵母数の月別変動について調べた.各試料は1999年7月から2000年6月までの1年間,それぞれ毎月1回ずつ採取した.生乳中の酵母の検出率と平均酵母数はそれぞれ順に,乳頭表皮では70.1%,52cfu/cm2,分房乳は47.9%,1.8cfu/ml,バルク乳は100%,28cfu/mlおよびローリー乳では100%,66cfu/mlであった.乳頭表皮および各種類の生乳中において90%以上を占める酵母数の水準は,乳頭表皮が150cfu/cm2以下(90.8%),分房乳が10cfu/ml以下(94.4%),バルク乳が100cfu/ml以下(97.8%)およびローリー乳が150cfu/ml以下(91.7%)であると考えられた.生乳中の酵母数は夏から秋期に高く,冬から春期に低くなる季節的な変動傾向が認められた.健康な乳牛の分房乳中では,主な乳房炎起因菌(C. parapsilosis, C. lambica, C. catenulata, C. rugosa)は夏期に検出されたが,冬から春期には検出されなかった. Microflora of lactic acid bacteria and yeasts of Laban which had been made with bovine or ovine milk by traditional natural fermentation in Yemen,were studied. The mean of viable cell counts of microorganisms in Laban was 6.4 x10⁸ cfu/ml. The 60 strains of lactic acid bacteria and 24 strains of yeasts were isolated and identified. Using a combination of PAGE and mass spectrom etry for protein identification,we obtained evidence that a putative odorant-binding protein,designated hypothetical protein LOC517854,occurs in bovine colostrum. This protein,termed as a putative bovine colostral odorant-binding protein (bcOBP), consists of 172 AA residues,including a putative 16-AA signal peptide. The theoreticalis oelectric point value and molecular mass of the full-length sequence of bcOBP were calculated to be 4.57 and 19604.18,respectively. ペンシルバニア紀(>3億1000万年前)に,有羊膜類であるSynapsidは他の系統から分化し,腺を発達させた。乳腺は毛包と会合した祖先型アポクリン腺から発達した。乳腺と毛の関係は単孔類の乳腺に保持されている。また,有袋類における乳腺毛の初期個体発生時の存在とその後の退化に,その証拠が残されている。単孔類の乳頭のない乳腺区分にあるmammo-ピロ-脂腺単位の密集した集団は,透水性を示す卵への水分や他の成分の補給のための構造的進化を反映しているかもしれない。ラクトースなどの成分は,初期Synapsidの祖先アポクリン腺から分泌されていたかもしれない。三畳紀のTherapsidは,複雑な栄養成分を含む乳を分泌し,卵サイズを小さくし,孵化時の新生子の未熟化を引き起こした。乳腺区分に生じた乳頭は,乳腺の毛を退化させた。 乳腺分泌は,羊皮状の卵殻をもつ卵を産むSynapsidで開始されたと考えられる。羊皮状の卵殻をもつ卵は,低湿度の空気にふれた際,急速に水分損失を被る。Synapsidは抱卵嚢の中で抱卵することで卵の乾燥を防いだが,これは母親の行動を制限した。羊皮状の卵殻をもつ卵は,殻を通して液体水分を補給することができる。乳腺分泌は,本来卵に水分を供給する手段として開始され,Cynodont (キノドン類)において内温性獲得の要因となった。Synapsidの卵は,ナトリウムやカルシウムイオンなどの栄養素を摂取した可能性がある。孔のある卵殻と2層性の卵黄膜は,子宮内での分泌物と抱卵時での乳腺分泌物の摂取を促進したと考えられる。 |

A thermophilic lactic acid bacterium, Lactobacillus fermentum TDS030603, produced about 100 mg/L of EPS in purified form when grew in de Man-Rogosa-Sharpe (MRS) broth. The 1% (w/v) solution of the purified EPS was highly viscous, exhibiting an apparent 22 viscosity (ηapp) of 0.88 Pa s at a shear rate of 10/s. To investigate the impact of carbohydrate source on the production yield and chemical structure of EPS and the viscosity of EPS solution, a chemically defined medium (CDM) has been developed. The efficacy of the use of acidic electrolyzed water (AEW) for premilking teat cleaning and disinfection (TCD) was evaluated using a split-udder experimental design. The efficacy of AEW for TCD was compared with that of NaClO solution contained 200 ppm available chlorine (HClO). The viable cell counts levels of SPC,lactic acid bacteria,thermotrophilic bacteria and anaerobe (group A) which contaminated on teat skin were over 10³ cfu/cm²,whereas psychrotrophilic bacteria and coliforms (group B) were less 10² cfu/cm². Carbohydrates were extracted from a sample of milk from a mink, Mustela vison (Family Mustelidae). Free neutral and acidic oligosaccharides were isolated from the carbohydrate fraction and their chemical structures were compared with those of white-nosed coati (Nasua narica, Procyonidae) and harbour seal (Phoca vitulina, Phocidae) that we had studied previously. The ratio of free lactose to milk oligosaccharides was similar to that in milk of the white-nosed coati; in both species, this ratio was much lower than that in the milk of most eutherians. A neutral exopolysaccharide produced in large amounts by Lactobacillus fermentum TDS030603 was purified from culture supernatants and its structure determined by monosaccharide analysis, methylation analysis and 1H-NMR spectroscopy. Optimum conditions for the production of this exopolysaccharide by the above bacterial strain, when cultured using cheese whey medium or MRS broth, were also investigated. The activity of α3-D-mannoside-β-1,2-N-acetylglucosaminyltransferase I (GnT I; EC 2.4.1.101), which catalyzes the first step in the conversion of oligomannose to complex or hybrid N-glycans of glycoproteins, was detected in bovine follicular fluid (bFF). The GnT I activity in bFF had a pH optimum of 5.8 and an absolute requirement for either Co2+, Mn2+, or Mg2+, the activity being stimulated by these cations in the above order. The apparent Km value for α1-3α1-6 mannopentaose of GnT I in bFF was 2.17 mM. Twenty-two strains of Saccharomyces cerevisiae isolated from tape-mash for the production of brem, a traditional rice wine from Bali, Indonesia, were divided into three groups (I, II, and III) based on the sequences of the rDNA spacer region and SUC2 gene. DNA analyses suggested that the seventeen strains in groups II and III were taxonomically closer to baking strains than the five strains in group I. These differences were reflected in their leavening ability in dough with and without addition of 5% sucrose, and their α-glucosidase activity, although most of the strains did not leaven dough with addition of 30% sucrose.
