Lactic acid bacteria (LAB) are commonly used in food industries,especially for the production of fermented dairy products. During fermentation process,some dairy LAB produce exopolysaccharide(EPS),providing good mouthfeel properties to the products by modifying their textures. In general, EPS is divided into two groups based on their monosaccharide composition; homo-EPS and hetero-EPS. It is believed that the major physiological function of EPS is the first line of biological defense against phagocytosis,phage attack, antibiotics,toxic metal ions,and physical stresses such as desiccation and osmotic stress. Short communication : Evidence for the presence of a putative odorant-binding protein in bovine colostrum OAK

Using a combination of PAGE and mass spectrom etry for protein identification,we obtained evidence that a putative odorant-binding protein,designated hypothetical protein LOC517854,occurs in bovine colostrum. This protein,termed as a putative bovine colostral odorant-binding protein (bcOBP), consists of 172 AA residues,including a putative 16-AA signal peptide. The theoreticalis oelectric point value and molecular mass of the full-length sequence of bcOBP were calculated to be 4.57 and 19604.18,respectively. The highest sequence similarity (83%) was observed with a potential pheromone transporter,Allergen Bos d 2. Human and great ape milks contain a diverse array of milk oligosaccharides, but little is known about the milk oligosaccharides of other primates, and how they differ among taxa. Neutral and acidic oligosaccharides were isolated from the milk of three species of Old World or catarrhine monkeys (Cercopithecidae: rhesus macaque (Macaca mulatta), toque macaque (Macaca sinica) and Hamadryas baboon (Papio hamadryas)) and three of New World or platyrrhine monkeys (Cebidae: tufted capuchin (Cebus apella)and Bolivian squirrel monkey (Saimiri boliviensis); Atelidae: mantled howler (Alouatta palliata)). Bactrian camel milk and colostrum are commonly used as foods in Mongolia, whose people believe that these products promote human health. It has been hypothesized that milk oligosaccharides are biologically significant components of human milk, acting as receptor analogs that inhibit the attachment of pathogenic microorganisms to the colonic mucosa, and as prebiotics, which stimulate the growth of bifidobacteria within the infant colon. Lactobacillus fermentum TDS030603株の生産する中性多糖の構造研究 OAK

A neutral exopolysaccharide produced in large amounts by Lactobacillus fermentum TDS030603 was purified from culture supernatants and its structure determined by monosaccharide analysis, methylation analysis and 1H-NMR spectroscopy. Optimum conditions for the production of this exopolysaccharide by the above bacterial strain, when cultured using cheese whey medium or MRS broth, were also investigated. Effects of carbohydrate source on physicochemical properties of the exopolysaccharide produced by Lactobacillus fermentum TDS030603 in a chemically defined medium OAK

A thermophilic lactic acid bacterium, Lactobacillus fermentum TDS030603, produced about 100 mg/L of EPS in purified form when grew in de Man-Rogosa-Sharpe (MRS) broth. The 1% (w/v) solution of the purified EPS was highly viscous, exhibiting an apparent 22 viscosity (ηapp) of 0.88 Pa s at a shear rate of 10/s. To investigate the impact of carbohydrate source on the production yield and chemical structure of EPS and the viscosity of EPS solution, a chemically defined medium (CDM) has been developed. Results of TLC, HPLC, 25 and 1H NMR spectroscopy indicated that the chemical structures of EPS released in MRS and in the CDM supplemented with glucose, galactose, lactose or sucrose were very similar. ウシをはじめとする家畜ミルクオリゴ糖研究の最近の進歩 OAK

人乳にはラクトース以外のミルクオリゴ糖が初乳で22~24 g/L,常乳で12~13 g/L含まれ,これまでに115種類もの構造が解析されている.一方でウシでは分娩直後の初乳で1 g/L以上のミルクオリゴ糖が含まれ,そのうちの70%は3'-シアリルラクトースなどの酸性オリゴ糖が占めるが,とくに酸性オリゴ糖の含有量は分娩後48時間以降の乳では急減し,常乳中にはごく少量しか含まれていない.ウシミルクオリゴ糖の構造研究は,含まれる量がごく少量であることが原因でヒトミルクオリゴ糖の構造研究に比べて著しく遅れていたが,近年親水性相互作用高速液体クロマトグラフィーやエキソグリコシダーゼ消化、質量分析の組み合わせによる構造解析方法の進歩によって微量での解析が可能となった.一方で,新たな各ウシミルクオリゴ糖の定量分析方法も開発された.本解説では,ウシを中心とする家畜ミルクオリゴ糖構造研究の歴史的な展開,新たな構造情報や定量分析方法・定量分析値,ならびにウシミルク糖タンパク質糖鎖の解析方法・構造情報を紹介するとともに,将来的な家畜ミルクオリゴ糖やミルク複合糖質の産業面での利用可能性について展望する.
