Linares,Rita de Cássia; Barry,Aline Fernandes; Alfieri,Alice Fernandes; Médici,Kerlei Cristina; Feronato,César; Grieder,Wladimir; Alfieri,Amauri Alcindo.
Neonatal diarrhea is one of the main causes of morbidity and mortality in piglets, and it leads to significant economic losses for pig farmers worldwide. The aim of this study was to determine the frequency of diagnosis, age group, and association of group A rotavirus (GARV) infection with diarrhea in piglets from pig herds in two (south and center-west)Brazilian geographical regions. The frequency of GARV diagnosis was evaluated between 2004 and 2007, using SS-PAGE on 681 fecal samples (428 diarrheic and 253 with normal consistency) from 1-4 week-old piglets. The animals were selected from 130 pig herds and 80 counties in the states of Rio Grande do Sul, Santa Catarina, Paraná, Mato Grosso do Sul, and Mato Grosso, Brazil. None of the herds were vaccinated...

Neurological and epidemiological aspects of a BoHV-5 meningoencephalitis outbreak BABT
Lunardi,Michele; Claus,Marlise Pompeo; Lisbôa,Júlio Augusto Naylor; Amude,Alexandre Mendes; Headley,Selwyn Arlington; Alfieri,Alice Fernandes; Alfieri,Amauri Alcindo.
Bovine herpesvirus 5 is a DNA virus that has been associated with meningoencephalitis in young cattle. While its clinical diagnosis is obscured by other major diseases that also produce similar neurological disease in cattle, the use of conventional virological techniques is hampered by the establishment of a lifelong latent infection in the host and the difficulty in differentiating BoHV-1 and BoHV-5. The aim of the current report is to describe the clinical and epidemiological aspects observed in a natural outbreak of BoHV-5 meningoencephalitis in a dairy cattle herd from Brazil. In the outbreak, the affected animals consisted of nine calves, which presented three possible forms of the neurological disease, subjectively classified as peracute, acute, and...

An outbreak of winter dysentery caused by bovine coronavirus in a high-production dairy cattle herd from a tropical country BABT
Takiuchi,Elisabete; Barry,Aline Fernandes; Alfieri,Alice Fernandes; Filippsen,Patrícia; Alfieri,Amauri Alcindo.
Bovine coronavirus (BCoV) is a known cause of winter dysentery (WD) in adult cattle. The morbidity of the disease is high, that results in a significant decrease in milk production and consequently, economic losses. In the present study, we report on a classical outbreak of WD that affected a high-production Holstein dairy herd raised in a tropical country. The lactating batch included 154 cows, and 138 (90%) presented diarrhea in a short (nine days) period of time. Three (2%) cows died. The other batches of animals did not become ill. The evolution of the disease in the herd, including the clinical signs and epidemiological features, strongly suggested a WD case. Semi-nested PCR and RFLP confirmed that BCoV was the cause of the infection. Samples tested...

Molecular epidemiology of the human group A rotavirus in the Paraná State, Brazil BABT
Santos,Jucélia Stadinicki dos; Alfieri,Alice Fernandes; Leite,José Paulo Gagliardi; Skraba,Irene; Alfieri,Amauri Alcindo.
From January/2000 to December/2003, 550 diarrheic fecal samples from the children and adults were collected in several geographical regions of Paraná State, Brazil. The enzyme immunoassay showed 120 (21.8%) samples positive for the group A rotaviruses. One hundred and fourteen samples were genotyped by multiplex-nested-PCR assay. The highest frequency (77.5%) of the positive samples (n=93) was observed in the children under 5 years old. Rotavirus diarrhea was more frequent in the cold and dry seasons of the four evaluated years. The most frequent genotypes were: G1 (50.9%), G4 (9.6%), G9 (7.0%), G2 (1.7%), G3 (0.9%), P[ 8] (71.9%), and P[ 4] (3.5%). The P[ 8] G1 (46.5%) and P[ 8] G4 (9.6%) were the main combinations found to P and G genotypes. The mixed...

A bovine teat papilloma specimen harboring Deltapapillomavirus (BPV-1) and Xipapillomavirus (BPV-6) representatives BABT
Claus,Marlise Pompeo; Lunardi,Michele; Alfieri,Amauri Alcindo; Otonel,Rodrigo Alejandro Arellano; Ferracin,Lara Munique; Fungaro,Maria Helena Pelegrinelli; Alfieri,Alice Fernandes.
The common occurrence of multiple papillomavirus infections has been shown in several studies involving the human host. However, investigations with the aim of identifying mixed papillomavirus infections in cattle have been conducted only recently. In the current work we describe a co-infection with two different bovine papillomavirus (BPV) types that was identified in a bovine teat papilloma. The skin wart was obtained from a cow belonging to a Brazilian beef herd. A PCR assay was carried out with the FAP primer pair, which amplifies a partial segment of the L1 gene (approximately 478 bp), and the amplicon was submitted to direct sequencing. Because nucleotide sequences with satisfactory quality scores were not obtained, the amplicon was cloned and...

Multiple bovine papillomavirus infections associated with cutaneous papillomatosis in brazilian cattle herds BABT
Claus,Marlise Pompeo; Lunardi,Michele; Alfieri,Amauri Alcindo; Otonel,Rodrigo Alejandro Arellano; Sartori,Daniele; Fungaro,Maria Helena Pelegrinelli; Alfieri,Alice Fernandes.
Cutaneous papillomatosis is a pathological condition commonly found in cattle and is characterized by the presence of benign proliferative tumors caused by bovine papillomavirus (BPV) infection. While multiple infections with human papillomavirus (HPV) are common in healthy and immunodeficient humans, studies with the aim of identifying mixed infections are still sporadic in veterinary medicine. The aim of this study is to describe the occurrence of multiple BPV infections in cattle affected by cutaneous papillomatosis. Fifteen skin warts were collected from at least two diverse anatomical regions of six bovines with papillomatosis belonging to three cattle herds from the Paraná state in Brazil. The BPV types present in the skin wart samples were...

Bovine coronavirus detection in a collection of diarrheic stool samples positive for group a bovine rotavirus BABT
Barry,Aline Fernandes; Alfieri,Alice Fernandes; Stipp,Danilo Tancler; Alfieri,Amauri Alcindo.
Neonatal diarrhea is an important cause of economic losses for cattle farmers. The main viral etiologies of enteric diseases are group A rotaviruses (GARV) and the bovine coronavirus (BCoV). Although both viruses infect calves of the same age, the occurrence of mixed infections is still under studied. The present study describes the co-infection of BCoV and GARV in stool samples. Forty-four diarrheic fecal samples from calves up to 60 days old that had previously tested positive for GARV by SS-PAGE were analyzed using semi-nested PCR for BCoV. A product with 251 bp of the BCoV nucleoprotein gene was amplified in 15.9% (7/44) of the samples, demonstrating that co-infection is not an unusual event. These results reinforce the need for testing for both GARV...

Isolation in HRT-18 cells and molecular analysis of a BCoV strain from diarrheic feces of naturally infected calves BABT
Stipp,Danilo Tancler; Barry,Aline Fernandes; Alfieri,Alice Fernandes; Bodnar,Lívia; Alfieri,Amauri Alcindo.
Bovine coronavirus (BCoV) may cause acute diarrhea in newborn calves, leading to significant economic losses for cattle farmers. There are several diagnostic techniques used to detect BCoV in calf fecal samples, but virus isolation still has advantages for antigenic and genomic characterization. This study describes the isolation in HRT-18 cells and molecular characterization of Brazilian BCoV wild-type strains. Three fecal samples from diarrheic 30 day-old calves were inoculated in HRT-18 cell monolayers, which were then evaluated for HA titers and tested using semi-nested PCR followed by RFLP and sequencing. Two samples were successfully isolated and presented HA titers of 16 and 32 units per 25 mL. The results were confirmed using semi-nested PCR and...

Segmented double-stranded genomic RNA viruses in fecal samples from broiler chicken BJM
Tamehiro,Claudia Yurika; Alfieri,Alice Fernandes; Médici,Cristina; Alfieri,Amauri Alcindo.
Segmented double-stranded RNA (dsRNA) viruses were identified by polyacrylamide gel electrophoresis (PAGE) technique in fecal samples from broiler chicken. A total of 378 fecal samples from 1-7 weeks old chickens were analyzed. dsRNA with migration profile characteristic of avian rotavirus (AvRV), reovirus (ARV) or picobirnavirus (PBV) was identified in 32 (8.5%), 7 (1.8%) and 13 (3.4%) samples, respectively. AvRV and ARV occurred more frequently in chickens up to 1 month old and were related with enteritis signs. Considering only fecal samples of chickens with diarrhea, the AvRV was detected in 37.8% (14/37) and the ARV in 13.5% (5/37) of analyzed samples. AvRV was identified in only 1.5% (4/274) and ARV was not detected in normal feces collected from...

Bovine herpesvirus 5 detection by virus isolation in cell culture and multiplex-PCR in central nervous system from cattle with neurological disease in Brazilian herds BJM
Claus,Marlise Pompeo; Alfieri,Alice Fernandes; Médici,Kerlei Cristina; Lunardi,Michele; Alfieri,Amauri Alcindo.
Bovine herpesvirus 5 (BoHV-5) is an important cause of meningoencephalitis in young and adult cattle. The multiple etiology of neurological disturbances in cattle makes the quick and conclusive diagnosis of BoHV-5 infection important for animal and public health, mainly because of herbivore rabies that is endemic in Brazilian cattle herds. The objective of this retrospective study was to use a multiplex-polymerase chain reaction (multiplex-PCR) for BoHV-5 and BoHV-1 glycoprotein C gene detection from stored central nervous system (CNS) tissue fragments of cattle with neurological clinical signs. Forty-seven frozen CNS samples of young and adult cattle from 31 herds in three Brazilian geographical regions (South, Southeast, and Center-west) were evaluated....

Identification of a mutation in the spike protein cleavage site in Brazilian strains of wild-type bovine coronavirus BJM
Takiuchi,Elisabete; Barreiros,Marco Antônio Bacellar; Alfieri,Alice Fernandes; Alfieri,Amauri Alcindo.
The spike (S) protein of coronaviruses, a type I membrane glycoprotein, is primarily responsible for entry into susceptible cells by binding with specific receptors on cells and mediating subsequent virus-cell fusion. The bovine coronavirus (BCoV) S protein is cleaved into two subunits, the N-terminal S1 and the C-terminal S2. The proteolytic cleavage site of S protein is highly conserved among BCoV strains and is located between amino acids 763 and 768 (KRRSRR). This study describes a single mutation in the S protein cleavage site of three Brazilian strains of BCoV detected in diarrheic fecal samples from calves naturally infected. The sequenced PCR products revealed that amino acid sequence of the cleavage site of our strains was KRRSSR, indicating a...

Virus isolation and molecular characterization of canine distemper virus by RT-PCR from a mature dog with multifocal encephalomyelit BJM
Amude,Alexandre Mendes; Carvalho,Glei dos Anjos; Alfieri,Amauri Alcindo; Alfieri,Alice Fernandes.
A case of multifocal distemper encephalomyelitis in a mature dog is described. In the presented case the ante mortem clinical diagnosis of canine distemper virus (CDV) infection could not be ideally performed due to the absence of typical signs of distemper, such as myoclonus and systemic signs accompanying the nervous signs. The definitive diagnosis of distemper encephalomyelitis was only carried out at post mortem through virus isolation in cell culture from fresh central nervous system (CNS) fragments and CDV nucleoprotein gene detection in the CNS by RT-PCR.

Histopathological and molecular characterization of encephalitic listeriosis in small ruminants from northern Paraná, Brazil BJM
Headley,Selwyn Arlington; Bodnar,Lívia; Fritzen,Juliana T.T.; Bronkhorst,Dalton Evert; Alfieri,Alice Fernandes; Okano,Werner; Alfieri,Amauri Alcindo.
Listeriosis is a disease primarily of ruminants caused by the Gram-positive bacterium Listeria monocytogenes. Ruminants either demonstrate manifestations of the encephalitic, septicemic, or reproductive form of listeriosis. The pathological and molecular findings with encephalitic listeriosis in a 5.5-month-old, male, mixed-breed goat and a 3-year-old Texel-crossed sheep from northern Paraná, Brazil are described. Clinically, the kid demonstrated circling, lateral protrusion of the tongue, head tilt, and convulsions; the ewe presented ataxia, motor incoordination, and lateral decumbency. Brainstem dysfunctions were diagnosed clinically and listeriosis was suspected. Necropsy performed on both animals did not reveal remarkable gross lesions; significant...
