Paralytic shellfish poison accumulation yields and feeding time activity in the Pacific oyster (Crassostrea gigas) and king scallop (Pecten maximus) ArchiMer

Pacific oysters and king scallops placed individually in a recirculating flume were fed for 2 weeks with a constant concentration (120 cell ml(-1)) of a toxic strain of Alexandrium minutum. Fluorescence at the outlet of each experimental unit was measured continuously, and biodeposits were recovered twice daily to evaluate feeding time activity (FTA) and rates of organic filtration (OFR), ingestion (OIR) and organic absorption (OAR). Ion-pairing high performance liquid chromatography (IP-HPLC) was performed concurrently to quantify paralytic phycotoxins both (i) individually in shellfish at the end of the contamination period and (ii) in A. minutum cultures to estimate cellular toxin concentration. Paralytic phycotoxin uptake by scallops (Pecten maximus) ArchiMer

It is difficult to apply sanitary standards for mollusc contamination by paralytic shellfish poisoning to animals not consumed whole. This problem is illustrated by the 1990 embargo on a Japanese shipment of frozen scallop muscle and gonad: the sanitary threshold applied to digestive gland (in Japan) does not guarantee that muscle and especially gonad are toxin-free. Accordingly, we performed experimental contaminations of scallops (Pecten maximus) from Port-en-Bessin (Normandy, France) using a toxic Japanese strain of Alexandrium tamarense. Modélisation de la contamination des bivalves par les toxines d'Alexandrium spp. Rapport final ArchiMer

L'objectif général de l'étude est de simuler les épisodes de contamination de certains bivalves (huîtres et moules) par les toxines d'Alexandrium minutum et A. calenella à partir d'un module expérimental adapté, et en vue de construire un modèle de contamination applicable à ces espèces. Ce modèle sera ensuite susceptible d'être couplé aux modèles biologiques locaux (estuaire de Penzé et étang de Thau) de déterminisme des blooms, ainsi qu'aux modèles hydrodynamiques développés par différentes équipes (Ifremer, Université de Montpellier). La première année (2001-2002) était consacrée à la simulation des contaminations en estuaire de Penzé (Alexandrium minulum) et au développement de cultures en masse d'A. calenella de Thau en bioréacteurs.

Le maximum saisonnier de Dynophysis cf acuminata à Antifer (Normandie, France) ArchiMer

Since 1987 Antifer oil tanker terminal (Normandy, France) was weIl known, as a preferential area for summer highest blooms of Dinophysis cf acuminata, a toxic dinoflagellate. Three oceanographic cruises were made in August 1989, before, in the course of and after the seasonal max. of this species. Results obtained from these cruises (hydrological parameters and phytoplankton) as weIl as from a daily survey at a reference station in Antifer harbour, corrobora te the prevailing role of coastal up flowing of Seine estuarine waters, pushed to the coast by a dominant S - SW wind, on intensity and duration of Dinophysis bloom. Such observations are in agreement with those of Summer 1987 at reference station.

Cinétiques de décontamination en milieu contrôlé de moules toxiques (Diarrheic Shellfish Poison). Premiers résultats ArchiMer

Laboratory assays have been performed, using mussels contamined with Dinophysis, in order to assess potential depurations kinetics. Mussels samples from Seine bay, Douarnenez bay and Vilaine bay have been tested . From these assays, which are to be performed in 1989; two main results are stressed : 1) when the toxicity level is near 0,5 MU/g of hepatopancreas*, depuration is undedectable during twelve days, 2) when the toxicity level exceeds 1 MU/g,depuration is important during the first eight days. *(quarantine level used by Japanese authorities) .

Etude de la décontamination de moules toxiques (toxines diarrhéiques) en laboratoire et en milieu naturel ArchiMer

Two studies of depuration, one in laboratory conditions and the other one in an oyster's pond were performed using 2 sets of contaminated mussels with diarrhetic toxins at a high and a low level (3 and 1 M.U.). Depuration kinetics were followed both by the adult mouse test and okadaïc acid analysis. Results are - in the pond, high toxic mussels need about 20 days to become eatable (toxicity level : 0.5 M.U.g-1 digestive glands), -this time decreases when the initial toxicity is low but it increases when mussels are underfed, - a good correlation is observed between the total toxicity and okadaïc acid concentration.

Shellfish production, and production of marine bivalve molluscs in particular, is the main form of marine aquaculture in the European Union, with a production estimated at 1,200,000 tons in 1999 (Food and Agricultural Organization). Moreover, aquaculture production is rapidly expanding throughout Europe and is becoming a major economic resource in developing areas such as Northern Greece, Ireland, and northwestern Spain. These shellfish, mainly mussels, oysters, and clams, are produced in a natural environment. The occurrence of new phytoplankton species in a coastal area may be explained by the import of shellfish containing whole live algal cells in their digestive tracts. Indeed, shellfish containing toxic algal cells can induce both primary contaminations in safe areas ( initially free from toxic microalgae), and secondary contaminations of other shellfish growing in the same area via the faeces of the imported animals. To mitigate this problem, shellfish need to be placed in a separate holding tank and their intestinal content purged. Several experiments using a self-regulated system were conducted to define the factors likely to influence the uptake of paralytic shellfish poison (PSP) by oysters in the Penze estuary (France, Brittany). Each 4-day experiment was carried out in a recirculated sea water system using 15 Pacific oysters (Crassostrea gigas) separated from each other and supplied with unfiltered natural seawater containing alternatively toxic (Alexandrium minutum) or non-toxic (Skeletonema costatum) algal diets. Effect of the PSP-causing dinoflagellate, Alexandrium sp. on the initial feeding response of Crassostrea gigas ArchiMer

The effects of toxic and non-toxic strains of Alexandrium sp. on the initial feeding responses of the Pacific oyster Crassostrea gigas were determined by continuous monitoring of the clearance rate of individual oysters. This was determined fluorometrically by loss of fluorescence of live microalgae introduced as food. Feeding oysters with toxic or non-toxic A. tamarense and toxic A. fundyense resulted in stop/start clearance behaviour. In contrast, control oysters fed a reference microalga, Isochrysis sp., known to support their growth, did not exhibit this behavior. Comet assay in phytoplankton as biomarker of genotoxic effects of environmental pollution ArchiMer

The alkaline comet assay was tested on different microalgae: the clinofiagellates, Karenia mikimotoi and Alexandrium minuturn, and the diatom, Chaetoceros gracilis. The microalgae were exposed during their exponential growth to the model direct genotoxicant, hydrogen peroxide (1 h, 5 and 100 pM H2O2). Following H2O2 exposure, the comet assay was validated only for K. mikimotoi for which genotoxicity was observed from the lowest tested concentration of 5 pM with a concentration-dependent effect. C. gracilis was too small in size (4 mu m) to be correctly analysed. For A. minuturn, our lysis buffer was not strong enough to digest the cellulosic thecal plates. Modelling of paralytic shellfish toxin biotransformations in the course of Crassostrea gigas detoxification kinetics ArchiMer

The aim of this study was to evaluate the importance of biotransformation of paralytic shellfish toxins during the detoxification process in contaminated oysters. Mathematical models based upon the detoxification patterns of digestive gland and other tissues were developed. It was demonstrated that biotransformations do not seem to play an important role in digestive gland or other tissue detoxification kinetics with our data set. Moreover, different toxin transfers from digestive gland toward other tissues were investigated. No significant transfer was highlighted in our data set. Continuous fluorescence recording as a way to improve Pacific oyster (Crassostrea gigas) models of paralytic shellfish toxin accumulation ArchiMer

Dinophysis cf. acuminata concentrations have been monitored from April to July 1985 in Vilaine Bay with 20 sampling stations and at several depths. Besides, variations in biomasses for other species have been analysed against Temperature/Salinity patterns. This study confirms 1) The importance of stratification and coastal currents in Dinophysis distribution, 2) the influence of bottom morphology on cumulative phenomenons as observed in the inner part of the Bay. In the same way, waters stratification would play an important role in species succession.

Influence of Crassostrea gigas (Thunberg) sexual maturation stage and ploidy on uptake of paralytic phycotoxins ArchiMer

The purpose of this study was to assess paralytic phycotoxin uptake in diploid and triploid oysters at two stages of their sexual cycle corresponding to their status in early summer (June) and winter (November). Samples of diploid and triploid oysters were exposed to a toxic culture of Alexandrium minutum for 4 days in each season. No significant differences in filtration or clearance rates were observed during either November or June experiments. When diploid oysters were at resting stage (November), toxin uptake showed no significant difference between the ploidy classes. Pour tenter d’apporter des réponses aux questions soulevées par une précédente étude commanditée pour approfondir les connaissances sur Vulcanodinium rugosum, l’organisme producteur de la Pinnatoxine G (PnTX G) (Hess et al., 2012), les objectifs de cette nouvelle étude sont les suivants : - Elargissement de la zone initiale de prélèvement d’Ingril à toute la lagune méditerranéenne et mise à profit de l’augmentation du nombre d’échantillons pour vérifier l’impact de l’effet matrice dans l’analyse LC-SM/MS. - Mise en culture en masse de Vulcanodinium rugosum et - Conduite d’une expérience de contamination in vivo sur des moules pour vérifier l’accumulation de la PnTX-G et comprendre les mécanismes de la métabolisation des PnTXs et de ses composés associés.....
