Genetic distance and its association with heterosis in cacao BABT

The efficiency of cacao breeding program can be increased by choosing superior crosses to be made between divergent clones. We assessed the genetic distance among five clones with RAPD data (genetic distance - GD) and with yield component data (Mahalanobis distance - MD). The clones were evaluated in a diallel, during five years, for five yield components. A total of 130 RAPD bands were scored. GD and MD were used to determine the correlation between genetic distances among clones and the performance of their hybrids. The correlation between GD and MD was 0.67 (P=0.03). Use of RAPD-PCR to identify true hybrid plants from crosses between closely related progenitors Braz. J. Genet.

RAPD-PCR molecular markers were used to identify common bean and soybean hybrid plants derived from crosses between closely related progenitors, with no apparent phenotypic differences. Primers OP-F12 and OP-0O3 were used to identify true hybrids derived from crosses between common bean cultivars Rudá (A 285) and AN 910408, and soybean cultivars Cristalina and Bossier, respectively. Each primer generated one polymorphic DNA band which was present in the male progenitor and absent in the female progenitor. As RAPD bands are normally inherited as dominant characters, the presence of these bands in the F1 plants confirmed their status.

The objective of the present study was to test three different procedures for DNA extraction of Melipona quadrifasciata based on existing methods for DNA extraction of Apis, plants and fungi. These methods differ in the concentrations of specific substances in the extraction buffer. The results demonstrate that the method used for Apis is not adequate for DNA extraction from M. quadrifasciata. On the other hand, with minor modifications this method and the methods for plants and fungi were adequate for DNA extraction of this stingless bee, both for adults and larvae

The soybean is a protein source of high biological value. However, the presence of anti-nutritional factors affects its protein quality and limits the bioavailability of other nutrients. The effect of heat-treatment, 150 ºC for 30 minutes, on hulled and hull-less soybean flour from the cultivar UFVTN 105AP on urease, trypsin inhibitor activity, protein solubility, amino acid profile, and in vivo protein quality was investigated. The treatment reduced the trypsin inhibitor activity and urease, but it did not affect protein solubility. ABSTRACT: The objective of this paper was to estimate the genetic divergence among 49 soybean ( Glycine max L. Merrill .) genotypes to assist grain quality-focused breeding programs in the choice of progenitors. The genetic divergence was estimated using the Mahalanobis generalized distance from the percentages of protein, oil, and fatty acids oleic, linoleic and linolenic after cultivation of genotypes in different environments. Co-evolution model of Colletotrichum lindemuthianum (melanconiaceae, melanconiales) races that occur in some Brazilian regions Genet. Mol. Biol.

Colletotrichum lindemuthianum, the causal agent of anthracnose in the common bean (Phaseolus vulgaris L.), displays a high level of virulence diversity, which explains the large number of existing pathotypes. Several lines of evidence indicate that such diversity is, at least in part, due to plant and pathogen co-evolution. A co-evolution model based on the binary classification of 25 races identified in Brazil by inoculation of differential cultivars and random amplified polymorphic DNA (RAPD) data is proposed. In this model, races 8 and 64 that infected bean cultivar Cornell 49-242 (Are gene) and Mexico 222 (Mexico I gene) are considered to be sources of two important evolutionary routes. Inferences about undescribed races from Brazil could be made.

Abstract Two retrotransposons from the superfamilies Copia and Gypsy named as Copia-LTR_SS and Gypsy-LTR_SS, respectively, were identified in the genomic bank of Sclerotinia sclerotiorum. These transposable elements (TEs) contained direct and preserved long terminal repeats (LTR). Domains related to codified regions for gag protein, integrase, reverse transcriptase and RNAse H were identified in Copia-LTR_SS, whereas in Gypsy-LTR_SS only domains for gag, reverse transcriptase and RNAse H were found. The abundance of identified LTR-Solo suggested possible genetic recombination events in the S. sclerotiorum genome. Our work was designed to estimate the informativeness of the cotton (Gossypium hirsutum L.) simple sequence repeat (SSR) microsatellite locus and to estimate the genetic distance between 53 cotton cultivars as well as to select a set of SSR primers able to differentiate between the 53 cotton cultivars studied. The genetic reduction of linolenic acid levels increases the quality and stability of soybean oil. The objective of this study was to determine the inheritance and evaluate the nature and magnitude of gene effects on soybean seed linolenic acid level. Means and variances of F1, F2, and F3 generations were made from the cross between accession BARC-12 (low linolenic acid content) and the commercial Brazilian cultivar CAC-1 (normal linolenic acid content). We used data simulation to determine the influence of population size, the effect of the degree of marker saturation of the genome, and the number of individuals required for mapping of recombinant inbred lines (RILs). Identification of proteins whose expression is transient, those that are of low-abundance, and of natural peptides not described in databases, is still a great challenge. Plant extracts are in general complex, and contaminants interfere with the identification of proteins involved in important physiological processes, such as plant defense against pathogens. This review discusses the challenges and strategies of separomics applied to the identification of low-abundance proteins and peptides in plants, especially in plants challenged by pathogens.
