RAPD-PCR molecular markers were used to identify common bean and soybean hybrid plants derived from crosses between closely related progenitors, with no apparent phenotypic differences. Primers OP-F12 and OP-0O3 were used to identify true hybrids derived from crosses between common bean cultivars Rudá (A 285) and AN 910408, and soybean cultivars Cristalina and Bossier, respectively. Each primer generated one polymorphic DNA band which was present in the male progenitor and absent in the female progenitor. As RAPD bands are normally inherited as dominant characters, the presence of these bands in the F1 plants confirmed their status. |

This work aimed to identify the quantitative trait loci (QTL) associated with photosynthesis and growth and productivity traits of soybean and to study possible associations between these traits by the analysis of coincidence of QTL in linkage groups (LGs). Thus, populations of recombinant inbred lines (RILs) of the F7 and F8 generations derived from the cross between the varieties BARC-8 and Garimpo were used. The traits evaluated were net assimilation rate of CO2 under saturating light (Asat), potential photosynthesis rate (Pmax), leaf area (A), specific leaf area (SLA), specific leaf nitrogen (N); root (W R), nodule (W N), stem (W ST), leaf (W L), pod (W P) and plant dry mass (W T); nodule (nN), seed (n s), and pod number (nP); seed fresh mass per plant... |

The soybean is a protein source of high biological value. However, the presence of anti-nutritional factors affects its protein quality and limits the bioavailability of other nutrients. The effect of heat-treatment, 150 ºC for 30 minutes, on hulled and hull-less soybean flour from the cultivar UFVTN 105AP on urease, trypsin inhibitor activity, protein solubility, amino acid profile, and in vivo protein quality was investigated. The treatment reduced the trypsin inhibitor activity and urease, but it did not affect protein solubility. Protein Efficiency Coefficient (PER) values of the flours were similar, and the PER of the hull-less soybean flour did not differ from casein. The Net Protein Ratio (NPR) did not differ between the experimental groups. The... |

ABSTRACT: The objective of this paper was to estimate the genetic divergence among 49 soybean ( Glycine max L. Merrill .) genotypes to assist grain quality-focused breeding programs in the choice of progenitors. The genetic divergence was estimated using the Mahalanobis generalized distance from the percentages of protein, oil, and fatty acids oleic, linoleic and linolenic after cultivation of genotypes in different environments. Genotypes were grouped by agglomerative methods and the two and three-dimensional projections of the distance matrix were obtained. The average protein and oil contents in the four environments ranged from 34.25 to 45.18% and from 16.48 to 23.01%, respectively. The average contents of the fatty acids oleic, linoleic and linolenic... |

Colletotrichum lindemuthianum, the causal agent of anthracnose in the common bean (Phaseolus vulgaris L.), displays a high level of virulence diversity, which explains the large number of existing pathotypes. Several lines of evidence indicate that such diversity is, at least in part, due to plant and pathogen co-evolution. A co-evolution model based on the binary classification of 25 races identified in Brazil by inoculation of differential cultivars and random amplified polymorphic DNA (RAPD) data is proposed. In this model, races 8 and 64 that infected bean cultivar Cornell 49-242 (Are gene) and Mexico 222 (Mexico I gene) are considered to be sources of two important evolutionary routes. Inferences about undescribed races from Brazil could be made. |

Abstract Two retrotransposons from the superfamilies Copia and Gypsy named as Copia-LTR_SS and Gypsy-LTR_SS, respectively, were identified in the genomic bank of Sclerotinia sclerotiorum. These transposable elements (TEs) contained direct and preserved long terminal repeats (LTR). Domains related to codified regions for gag protein, integrase, reverse transcriptase and RNAse H were identified in Copia-LTR_SS, whereas in Gypsy-LTR_SS only domains for gag, reverse transcriptase and RNAse H were found. The abundance of identified LTR-Solo suggested possible genetic recombination events in the S. sclerotiorum genome. Furthermore, alignment of the sequences for LTR elements from each superfamily suggested the presence of a RIP (repeat-induced point mutation)... |

Seven soybean cultivars (Bossier, Cristalina, Davis, Kent, Lincoln, Paraná and Uberaba), with different levels of resistance to Cercospora sojina, race 04, were crossed according to a diallel design, with no reciprocals, to determine the general and the specific combining abilities for the resistance. The evaluations of the reaction to the disease were performed 20 days after the inoculation of the fungus on the most infected leaflet of the plant, in the parents and in the F1 hybrids. To quantify the resistance, the following characteristics were evaluated: infection degree (ID); number of lesions per leaflet (NLL); lesion mean diameter (LMD); lesioned leaf area (LLA); percentage of lesioned leaf area (PLLA); number of lesions per square centimeter (NLC)... |

The soybean (Glycine max (L.) Merrill) frogeye leaf spot is caused by the fungus Cercospora sojina Hara and is a widespread disease in Brazil and other countries, causing severe losses in grain yield and also affecting seed quality. The availability of DNA markers linked to genes for resistance to this disease would accelerate breeding programs, particularly when other traits are also being evaluated. Bulked segregant analysis was applied to 3 F2 populations derived from crosses between the resistant cultivars Parana, Cristalina and Uberaba, and the susceptible cultivar Bossier. In the cross 'Parana' x 'Bossier', 2 RAPD markers were identified, CSOPA1(800C) and CSOPA2(1,250C), located at 4.4 ± 1.8 centiMorgans (cM) and 3.4 ± 1.7 cM respectively from the... |

Various population sizes and number of markers have been used to obtain genetic maps. However, the precise number of individuals and markers needed for obtaining reliable maps is not known. We used data simulation to determine the influence of population size, the effect of the degree of marker saturation of the genome, and the number of individuals required for mapping of recombinant inbred lines (RILs). Three genomes with 11 linkage groups were generated with saturation levels of 5, 10 and 20 cM. For each saturation level populations were generated with 50, 100, 154, 200, 300, 500 and 800 individuals with 100 replications for each population size. A total of 2100 populations was generated and mapped. Small marker numbers and small population sizes... |

The enrichment and isolation of proteins are considered limiting steps in proteomic studies. Identification of proteins whose expression is transient, those that are of low-abundance, and of natural peptides not described in databases, is still a great challenge. Plant extracts are in general complex, and contaminants interfere with the identification of proteins involved in important physiological processes, such as plant defense against pathogens. This review discusses the challenges and strategies of separomics applied to the identification of low-abundance proteins and peptides in plants, especially in plants challenged by pathogens. Separomics is described as a group of methodological strategies for the separation of protein molecules for proteomics.... |
