Taking advantage of the massive genome sequencing effort made on thermophilic prokaryotes, thermal adaptation has been extensively studied by analysing amino acid replacements and codon usage in these unicellular organisms. In most cases, adaptation to thermophily is associated with greater residue hydrophobicity and more charged residues. Both of these characteristics are positively correlated with the optimal growth temperature of prokaryotes. In contrast, little information has been collected on the molecular 'adaptive' strategy of thermophilic eukaryotes. The Pompeii worm A. pompejana, whose transcriptome has recently been sequenced, is currently considered as the most thermotolerant eukaryote on Earth, withstanding the greatest thermal and chemical... |

Three species of man.-rove oysters, Crassostrea rhizophorae, C. brasiliana, and C. gasar, have been described along the Atlantic shores of South America and Africa. Because the distribution of these molluscs is of great biological and commercial interest, their taxonomy and distribution deserve further clarification. Therefore, 15 populations were sampled from both continents. Their 16S mitochondrial polymorphism was studied by sequencing and PCR-RFLP analysis. Two haplotypes were identified. Haplotype a was the only one observed in Africa, but it was also observed in South America together with haplotype b. Because C. gasar is the only mangrove oyster identified on the west coast of Africa, haplotype a was attributed to this species, which has thus been... |

Euryhaline teleosts possess the capacity to osmoregulate under various environmental conditions (freshwater to hypersaline water). This physiological capacity is generally monitored using enzyme activity assays (Na+/K+-ATPase...), hormones quantification (prolactine, growth hormone) or their mRNAs expression. To date, few studies addressed the genetic correlates of adaptation to varying salinity at a molecular level in such fish. In the sea bass Dicentrarchus labrax, genetic differentiation was observed at specific allozyme loci between lagoon- and open-sea populations. In the present study, we investigated transcriptomic response of D. labrax to salt- and freshwater acclimation in two organs involved in osmoregulation, gill and intestine. By using... |

Summer mortality of Pacific oysters is known in several countries. However no specific pathogen has been systematically associated with this phenomenon. A complex combination of environmental and biological parameters has been suggested as the cause and is now starting to be identified. A high genetic basis was found for survival in oysters when a first generation (G1) was tested in three sites during summer. This paper presents a synthesis on physiological characteristics of two selected groups ('R' and 'S', from families selected for resistance and susceptibility to summer mortality respectively), of the second and third generations. R and S showed improvement or reduction of survival compared with the control in both field and laboratory trials... |

Marine bivalve metabolism can be perturbed by hydrocarbon and pesticide pollution in coastal ecosystems. In this study, in the Pacific oyster, Crassostrea gigas, full-length cDNAs encoding two non-P450 phase I enzymes, flavin-containing monooxygenase 2 (FMO-2) and monamine oxidase A (MAO A), were characterized. Both sequences contained the co-factor fixation motifs characteristic of their respective enzyme families. Using reverse transcription polymerase chain reaction (RT-PCR), the messenger RNA (mRNA) transcription levels of these two enzymes in tissues of oysters exposed, under experimental conditions, to hydrocarbons and two pesticide treatments were investigated. The pesticide treatments were exposure to either glyphosate or to a mixture composed of... |

For a long time, population geneticists have had to content themselves with analysing neutral markers to infer selection processes indirectly. The recent improvement of sequencing tools now enables them to analyse the variations of a large number of genes and therefore to spot the genes, or the amino acids, which are under the direct influence of the selection (Yang & Bielawski, 2000). In order to analyse the selection processes acting on the mutations and discriminate the different factors acting on their evolution (genetic selection or drift), it is essential to compare polymorphism data between genomes of the same species/population and divergence data between species/populations (Kimura, 1983). Recently, polymorphism data has become available for a... |

Phosphoglucomutase is a key enzyme in glycolysis and has been widely studied in vertebrates and some invertebrates but no molecular information is available in marine invertebrates despite the importance of this marker in ecological and genetical studies. In this work, we isolated a cDNA and the corresponding genomic sequence that encode PGM-2 locus in the Pacific oyster Crassostrea gigas. We used sequences drawn from the database to construct an evolutionary framework for examining the position of mollusc PGM sequences among prokaryotic and eukaryotic homologues and showed that oyster PGM gene organization was closer to vertebrates PGM genes than other invertebrates as previously found in other Lophotrochozoa species. We also investigated PGM mRNA... |

Mangrove oysters include various species that are also present on different coasts. In the southern part of the Atlantic Ocean, Crassostrea rhizophorae has been observed along the southern coast of America and Crassostrea gasar along the western coast of Africa. Fifteen populations of these oysters were sampled from these two geographic areas. Their polymorphism was studied on the 16S mitochondrial fragment both by par sequencing and RFLP analysis. Some samples' caryotype were also analysed. Two haplotypes were identified: C. gasar was found in Africa, but also for the first time in South America, whereas C. rhizophorae was only found on the coasts of South America. |
