The present study proposes a modification of the technique described by PURSER & MOIR (1959) for the quantitative evaluation of rumen ciliate based on an adaptation described by DEHORITY (I984). The modifying process includes: the replacement of two drops of brilliant green dye, for at least four hours, by three drops of lugol solution, for at least 15 minutes. It was made a comparative evaluation of these stainings. It was concluded that lugol solution can replace the brilliant green dye showing the following advantages: staining time reduction and subsequent speeding of sample processing; evidence of skeletal plates of entodiniomorphs making its identification easier; improved observation of small ciliates and inconspicuous structures; improved... The hemocyte composition of adults of Amblyomma cajennense (Fabricius, 1787) Koch, 1844 adults consisted of three basic cellular types: prahemocyte I (PR I), plasmatocyte (PL) and spherulocyte (ES II). Among the ixodids observed (A. cajennense; A. dissimile Koch, 1844; Rhipicephalus sanguineus Latreille, 1806; Anocentor nitens (Neumann, 1897) Schulze, 1937; Boophilus microplus Canestrini, 1887 and Haemaphysalis sp.) only A. cajennense was different in the adult hemolymph colour. It changed from amber to deep blue as the ixodid age advanced. With the purpose of characterize morphologically the hemocytes of larvae and nymphs of Rhipicephalus sanguineus on different nutritional phases, it was obtained samples of haemolymph were obtained by sectioning the forelegs and collecting the drop there formed. After dried, the samples were fixed by methanol and stained by Giemsa. Microscopical observation resulted in the characterization of five basic cellular types: prohemocytes, plasmatocytes, granulocytes, spherulocytes and oenocytoids.
