One Perkinsus species may hide another: characterization of Perkinsus species present in clam production areas of France ArchiMer

Although clam populations in France are known to be infected with protozoans of the genus Perkinsus, no molecular characterization was previously performed on these parasites. Considering that several members of this genus have been associated with mortalities of molluscs worldwide, a study was undertaken in order to characterize these parasites in France. For that purpose, clams, Ruditapes philippinarum and R. decussatus, collected from different production areas and found to be infected with Perkinsus sp. in thioglycolate culture medium, were selected for PCR-RFLP tests and sequencing. Created in 1986, the REPAMO network (Network of mollusc pathology), is a national zoosanitary surveillance network of shellfish health status along the French coasts. Its activities are in keeping with two European Directives, 91 /67/EEC and 95/70/EC and are a patt of the institutional tasks ofIfremer. The aims of the network are to prevent the introduction and spread of exotic pathogens and to survey the evolution of notifiable pathogens already present in France. Bonamiosis is a parasitic disease (causative agent: Bonamia ostreae) affecting the European flat oyster Ostrea edulis, responsible for a drastic decline in its aquaculture production. Selective breeding programs for resistance to bonamiosis have been undertaken to counter this disease. Nous disposons aujourd'hui d'o utils diagnostiques sensibles et spécifiques (PCR, HIS ... ) et d'un protocole de transmission de l' infection qui permettent d'assurer un suivi des productions larvaires en écloserie de bivalves pour les espèces considérées comme sensibles aux infections à herpèsvirus, mais également pour de nouvelles espèces. En effet, des mortalités massives (100 %) ont été rapportées dans une écloserie de coquille St-Jacques en Bretagne et ont pu être associées à la détection de OsHV -1. Les résultats obtenus concernant cette espèce, Pecten maximus, complètent et reflètent l'ensemble des travaux réalisés.

Bonamia ostreae is an intracellular protozoan which is recognized as a cause of mortality in European populations of flat oysters (Ostrea edulis). Based on the recent characterization of actin genes of B. ostreae, specific primers were designed for real-time PCR using SYBR® Green chemistry. Specificity was demonstrated by the unique melting temperature peak observed in positive samples and by the lack of amplification in samples of oysters infected by closely related parasites, including Bonamia exitiosa. A calibration curve using a cloned template was defined to estimate copy number. Marteilia in American oysters Crassostrea virginica reared in France ArchiMer

Progeny of Crassostrea virginica oysters, introduced into France in 1992, were reared at 3 different locations. A survey was taken of these young C. virginica oysters during 1993. Histological and electron microscopical examination revealed a parasite interpreted as a Martelia-like protozoan in 1 oyster. The structure and the initial stages of the developmental cycle of this parasite are described for the first time in American oysters. However, no mature spores were detected. Thus, exact identification of the protozoan is difficult. This study indicates that American oysters could be infected with Marteilia spp. during cohabitation with Marteilia-infected native bivalves in France.

Bonamia exitiosa is an intracellular parasite (Haplosporidia) that has been associated with mass mortalities in oyster populations in the Southern hemisphere. This parasite was recently detected in the Northern hemisphere including Europe. Some representatives of the Bonamia genus have not been well categorized yet due to the lack of genomic information. In the present work, we have applied Whole-Genome Amplification (WGA) technique in order to characterize the actin gene in the unculturable protozoan B. exitiosa. In the present study, we have used molecular approaches to identify this parasite in samples of the dwarf oyster Ostrea stentina after reports of massive mortality along the Tunisian coasts.
