Gamete cryopreservation, an asset for a durable pearl farming in French Polynesia ArchiMer
Hui, Belinda; Demoy-schneider, Marina; Vonau, Vincent; Le Moullac, Gilles; Moriceau, Jacques; Le Pennec, Marcel; Cochard, Jean-claude.
Cryopreservation is a useful tool for genetic improvement which has been applied to several bivalve mollusk species. It would allow to keep the gametes of selected individuals and to preserve ex-situ the biodiversity of wild populations of the black-lip pearl oyster Pinctada margaritifera threatened of standardization by the seed transfers between the islands in French Polynesia. The ability to cryopreserve spermatozoa would bring about significant benefits to the cultured black pearl industry. Sperm freezing supposes the control of different steps: preparation of breeders, sperm collection, evaluation of sperm quality and the freezing process itself. Broodstock conditioning in hatchery allows to obtain gametes from the pearl oyster. Sperm quality is...

Dao, Jean-claude; Buestel, Dominique; Gerard, Andre; Halary, C.; Cochard, Jean-claude.
The king scallop (Pecten maximus L.) has been the object of some in-depth studies since 1973 with the double aim of natural resources management and extensive aquaculture in mind. The results obtained during the first ten years enabled the launch of a pluri-annual 1983-1987 program which aims to demonstrate the re-populating of the roadsted of Brest and the introduction of new management methods in the Bay of Saint-Brieuc. The technical results of the resource management and of the aquaculture operations are described. They enabled the stabilisation of the annual production of the Saint-Brieuc deposit as well as to foresee a new biomass of 80-100 tonnes of spawners per year in the roadsted of Brest. However, the implementation of all these developments is...

Pearl Oysters Pinctada margaritifera grazing on natural plankton in Ahe atoll lagoon (Tuamotu archipelago, French Polynesia) ArchiMer
Fournier, Jonathan; Dupuy, Celie; Bouvy, Marc; Couraudon-reale, Marine; Charpy, Loic; Pouvreau, Stephane; Le Moullac, Gilles; Le Pennec, Marie-annick; Cochard, Jean-claude.
In atoll lagoons of French Polynesia, growth and reproduction of pearl oysters are mainly driven by plankton concentration. However, the actual diet of black-lip pearl oysters P. margaritifera in these lagoons is poorly known. To fill this gap, we used the flow through chamber method to measure clearance rates of P. margaritifera in Ahe atoll lagoon (Tuamotu Archipelago, French Polynesia). We found : (i) that pearl oysters cleared plankton at a rate that was positively related to plankton biovolume, (ii) that nanoflagellates were the main source of carbon for the pearl oysters, (iii) that the quantity and origin of carbon filtrated by pearl oysters was highly dependent on the concentration and composition of plankton. These results provide essential...

Hamida, Leila; Medhiouband, Mohamed; Cochard, Jean-claude; Romdhane, Mohamed; Le Pennec, Marcel.
A comparative study of the reproductive cycle of Ruditapes decussatus under natural (South of Tunisia) and controlled conditions (hatchery). The aim of this study was to describe and compare the gametogenic activity of Ruditapes decussatus in situ and in hatchery. Criteria of maturity were characterized and the positive role of conditioning in the success of artificial reproduction was underlined. Specimens sampled in the South of Tunisia were conditioned at the experimental station of Monastir (Tunisia). The parameters studied were the variation of the condition index and the frequency distribution of oocyte diameters. Results showed that the reproductive cycle was continuous in situ. Gametogenesis started in January and February. Gonadic activity...

Cochard, Jean-claude; Goarant, Cyrille; Boglio, Eric; Lemonnier, Hugues.
The influence of the soil quality of culture pond on the physiological state of Penaeus stylirostris reared in earthen shrimp pond in New Caledonia was investigated by the measurement of the osmoregulatory capacity and magnesium hemolymph concentration of samples of 20 of these shrimp captured in the pond and maintained for 24 or 48h in cages laid on the bottom. A first experiment was affected by the occurrence of a pathology caused by Vibrio penaeicida probably due to initial handling stress. The second experiment showed that the osmoregulatory capacity is depresssed as a function of the soil quality : the more it is reduced the more the depression is important.

Widowati, L; Cochard, Jean-claude; Dorange, G; Le Pennec, M.
To date, it has been impossible to fertilize the oocytes of ripe Pecten maximus obtained by gonad stripping as it is routinely done in hatchery with Crassostrea gigas . The events occurring between artificial induction of spawning and normal fertilization were studied experimentally in order to explain this difference. Fertilization and rearing trials were performed on oocytes obtained from the gonads of both species by scarification, by aspiration from genital ducts (of P. maximus only) and after spawning. Every batch of oocytes was fixed for histological study. Serial sections of the gonad of a fixed spawning P. maximus were made to reveal modifications of the nucleus of the oocytes during their expulsion. In P. maximus the stripped oocytes can be...

Calcein staining of calcified structures in pearl oyster Pinctada margaritifera and the effect of food resource level on shell growth ArchiMer
Linard, Clementine; Gueguen, Yannick; Moriceau, Jacques; Soyez, Claude; Hui, Belinda; Raoux, Aurore; Cuif, Jean Pierre; Cochard, Jean-claude; Le Pennec, Marcel; Le Moullac, Gilles.
Marine mollusc shell growth has been widely measured using fluorochrome marking. In order to test the efficiency and reliability of calcein staining on P. margaritifera shells and pearls, the present study examined two administration methods, different concentrations and several immersion times.Immersion in a 150 mg.L-1 calcein solution for 12 h to 24 h appeared to be the best method for marking P. margaritifera shells. For pearl marking, injection of a 200 mg.L-1 calcein solution into the pearl pouch was the optimal method. Calcein marking was then used to measure the influence of food resource levels on the shell growth. Groups of 23-month-old P. margaritifera were fed at three trophic levels for two months. The two highest food levels tested (6 000...

Influence of sediment characteristics on shrimp physiology: pH as principal effect ArchiMer
Lemonnier, Hugues; Bernard, E; Boglio, E; Goarant, Cyrille; Cochard, Jean-claude.
Penaeid shrimp reared in earthen ponds are exposed to sediment, which can, in some instances, induce a stress. In seawater, the osmoregulatory capacity (hyporegulation) is a useful tool to compare the physiological condition of shrimp exposed to various stressors. By keeping some shrimp in cages at different locations of a single pond heterogeneous in terms of sediment quality, it was possible, using osmotic pressure (OP), to identify some locations where the stress was maximum and some others where it was minimum. Simultaneously, sediment samples were taken and analysed in order to evaluate some physico-chemical parameters that could be related to the stress observed in the shrimps kept in the cages. This approach allowed to show a significant positive...

Effect of food conditioning on gonadic activity in the oyster Pinctada margaritifera ArchiMer
Le Moullac, Gilles; Hui, Belinda; Vonau, Vincent; Levy, Peva; Cochard, Jean-claude.
Preliminary experiments allowed defining basic requirements of Pinctada margaritifera for somatic and gonadic growth in controlled conditions. This study was continued by observation of the gametogenesis at a cellular level. In April/May 2008, 125 oysters were conditioned for 30 days in 25 tubular tanks (30 L). Water was renewed 4 times/hour. A mixed (v:v) diet of Isochrysis galbana (T-Iso) and Chaetoceros gracilis was supplied continuously. A control batch of 125 oysters was maintained in trays in the lagoon. An initial sample of 30 individuals was made. Gonadic changes were first characterized using a gonadal development index (GDI) based on the ratio of the gonad area to the total visceral mass area on a sagittal section of the body. Histological...

Investigation of marine bivalve morphology by in vivo MR imaging: first anatomical results of a promising technique ArchiMer
Pouvreau, Stephane; Rambeau, Marc; Cochard, Jean-claude; Robert, Rene.
Classically, investigation of the internal morphology and composition of molluscs and especially bivalves relies on destructive method (biometry, biochemistry and histology). These techniques have given essential information, but in contrast are time consuming and lead to the irreversible loss of the animal while they don't allow integration of the various levels of molecular-to-organism functioning. The aim of this study is to analyze for the first time the potential of NMR (nuclear magnetic resonance) imaging (MRI) to depict, with sufficient resolution and satisfactory contrast, the anatomy of a bivalve model, the Pacific oyster, Crassostrea gigas, without opening it. MRI experiments were carried out at 19 °C in several non-anaesthetized adult Pacific...

Buestel, Dominique; Cochard, Jean-claude; Gerard, Andre.

The limits of morphometric features for the identification of black-lip pearl oyster (Pinctada margaritifera) larvae ArchiMer
Paugam, A; D' Ollone, Christine; Cochard, Jean-claude; Garen, Pierre; Le Pennec, Marcel.
As with most cultivated bivalves, culture of the Tahitian pearl oyster Pinctada margaritifera is particularly dependent on the natural environment, especially for spat supply. The ability to track in real time the abundance and the development of pearl oyster larvae in the plankton would help optimize spat collection in atolls of French Polynesia. However no identification criteria are available for the larvae of several bivalves species present in the lagoons and it is not yet possible to specifically monitor pearl oyster larvae. The aim of this study is to determine the most pertinent morphological identification criteria, to specifically identify the larvae of P. margaritifera and differentiate them from those of three other abundant species: Pinctada...

This study aimed to model the food intake of P. margaritifera in order to examine the relationship between food level and reproductive activity. The effect of microalgae concentration on ingestion rate and assimilation efficiency was studied over a broad concentration range, using a mixture of Isochrysis galbana and Chaetoceros gracilis. Reproductive effort was assessed using three microalgae concentrations of 0.5, 7 and 18 cell μL-1. Reproductive status was assessed by gonad development index (GDI) – the ratio of the gonad surface to the visceral mass surface – and histological analysis of the gonad based on the presence (continuous or discontinuous) or the absence of gonial cells (GC). Ingestion is a saturating function of seston concentration for...

Soyez, Claude; Tiapari, Jerome; Teissier, Hinano; Cochard, Jean-claude.
The growth of the abalone Haliotis Tuberculata L. in Bay of Brest ArchiMer
Cochard, Jean-claude.
The growth of H. tuberculata L. was studied by analysing the distribution of the growth checks on the shell of 640 individuals. The average lengths of the seven first rings including the first one appear to fit weIl to the Von Bertalanffy model. The value of the growth parameter K = 0,33 is rather similar to those cited in Litterature for the same species but the average attainable size L%= 97,5 seems to be low in this area.

Growth and gonad development of the tropical black-lip pearl oyster, Pinctada margaritifera (L.), in the Gambier archipelago (French Polynesia) ArchiMer
Le Moullac, Gilles; Tiapari, Jerome; Teissier, Hinano; Martinez, Elodie; Cochard, Jean-claude.
The growth and reproductive cycle of cultured black-lipped pearl oysters, Pinctada margaritifera (L.), were studied in the Gambier Islands (134°52' W, 23°07' S) from September 2002 to August 2003. Temperatures were recorded throughout the year, revealing seasonal temperature variations between 22.3 °C and 27.8 °C. The mean annual chlorophyll a value, as computed from satellite data, was 0.188 ± 0.075 µg L-1. To study growth and reproduction, 720 two-year-old individuals were ear hung on longlines suspended at a depth of 7 m. Samples were taken twice a month to obtain the following measurements: shell height; wet weight of flesh and total oyster; dry weight of adductor muscle, mantle and visceral mass; and glycogen content. Chemical induction of larval metamorphosis of Pecten maximus with a note on the nature of naturally occurring triggering substances ArchiMer
Chevolot, Lionel; Cochard, Jean-claude; Yvin, Jean-claude.
Delayed metamorphosis usually occurs when scallop Pecten maximus (L.) larvae are reared in still water. Metamorphosis rates may be increased by adding aqueous extracts of certain marine red algae, including one [Delesseria sanguinea (Linne) Lamouroux (Delesseriaceae, Rhodomelaceae)] from which various chemical inducers have been isolated. Of these, jacaranone (a simple quinol) is by far the most active component; other components (p-OH phenylacetic acid methyl ester, p-OH benzaldehyde, p-OH benzyl alcohol) are only weak inducers. All are catabolic derivatives of tyrosine. Homogentisic acid (biosynthetically related to jacaranone) and some catecholamines (L-DOPA, epinephrine) can also induce metamorphosis of P. maximus larvae. All these compounds can be...
