The Mediterranean Thau Lagoon is an important oyster farming area in Europe. Oyster growth rates are among the highest in France, although chlorophyll a concentration is low. Previous studies have demonstrated that picophytoplankton, nano-microphytoplankton, dinoflagellates and loricate ciliates such as tintinnids are abundant. However, heterotrophic flagellates and aloricate ciliates have not been investigated. The influence of organic nutrients on the evolution of Pseudo-nitzschia delicatissima cultures was investigated in an enrichment experiment with high-molecular-weight dissolved organic matter (HMWDOM) and in an uptake assay with N-15-ammonium and N-15-urea. HMWDOM was extracted from seawater collected at a nearby shore station during the decline of a diatom bloom. Environmental conditions ultimately leading to blooms of the toxic dinoflagellate Alexandrium catenella/tamarense were investigated at two Mediterranean sites (Annaba Bay, Algeria and Thau lagoon, France). Three years were examined in details: 1992 (a pre-Alexandrium period), 2002 (a year with the first bloom in Annaba) and 2010 (a year with a major bloom in Annaba). Most conditions were similar, but ammonium concentrations were much higher in Annaba (up to 100 μM) than in Thau (up to 10 μM). Since 1998, blooms of Alexandrium catenella/tamarense in the lagoon of Thau developed regularly each autumn, reaching a maximum of several millions cells per liter in 2004. By contrast, spring blooms occurred only twice (in 2000 and 2007). During these periods, sea surface temperatures (SST) and the wind patterns appear to impact the bloom occurrences much more than the apparent limiting resources such as inorganic nutrients. The analysis of SST and wind from April to June and September to November (from 2000 to 2007) indicates first that there has to be an initial wind stress in order to resuspend the cysts buried in the sediment. The oyster pond under study has a natural sediment bottom and water is changed only once or twice every month depending on tidal height. Between successive water renewals, the pond is a closed system, equivalent to a batch culture in which sinking and biodeposition of particulate matter are the only sources of organic matter to the bottom. Laboratory experiments were designed to study the toxin content and profile of the Alexandrium catenella strain ACT03 (isolated from Thau Lagoon, French Mediterranean) in response to abiotic environmental factors under nutrient-replete conditions. This dinoflagellate can produce various paralytic shellfish toxins with concentrations ranging from 2.9 to 50.3 fmol/cell. The toxin profile was characterized by carbamate toxins (GTX3, GTX4 and GTX5) and N-sulfocarbamoyl toxins (C1, C2, C3 and C4). A cross-ecosystem comparison of data obtained from 20 French Mediterranean lagoons with contrasting eutrophication status provided the basis for investigating the variables that best predict chlorophyll a (Chl a) concentrations and nutrient limitation of phytoplankton biomass along a strong nutrient enrichment gradient. The dilution technique was used to estimate chlorophyll and pheopigment, net and gross production as well as zooplankton grazing over a 12-month period in a coastal lagoon in Southern France. Uptake rates of three potential N-sources (ammonium, nitrate and N-urea) and two potential C-sources (HCO(3)(-) and C-urea) were estimated during growth of Alexandrium catenella in both light and dark phases. Alexandrium tamarense (M. Lebour) Balech strains isolated in spring 2007 from a single bloom in Thau lagoon have been grown in nonaxenic artificial media. Time series data have been examined in Thau lagoon (Southern France) from 1972 to 2006 for water temperature, salinity, nutrients and from 1987 to 2006 for phytoplankton. A first main trend identified is an increase in mean annual water temperature (1.5 degrees C over 33 years or 0.045 degrees C/year) that was not evenly distributed among seasons.
