A total of 120 Pekin ducks were distributed at random into four experimental groups, vaccinated or not against Newcastle disease (ND): G1 (Ulster 2C strain), G2 (B1 strain), G3 (LaSota strain), and G4 (nonvaccinated group). At 60 days of age, all groups were challenged with a pathogenic ND virus (NDV) suspension, and a group of specific pathogen free (SPF) chicks (G5) was also inoculated. Cloacal and tracheal swabs from all birds were collected after six, 14, 20, and 30 days post-challenge for virus isolation. NDV was isolated in 100% of SPF chicks. Pekin ducks from all groups, vaccinated or not, did not show any ND clinical signs, demonstrating that these birds are not susceptible to ND clinical disease. Reservoir competence for the Newcastle Disease virus (NDV) was evaluated in sparrows (Passer domesticus, Linnaeus 1758) captured on a commercial poultry farm and a chicken hatchery in the State of Pernambuco, Northeastern Brazil. A total number of 103 birds collected from a poultry farm (24/103) and a chicken hatchery (79/103) were examined. Hemagglutination inhibition tests, isolation, and viral characterization were performed in all samples collected from each bird. Titers ranging from 1:2 to 1:64 were detectable in 10.68% of sparrows, but positive serology and viral isolation were obtained only from sparrows captured at the hatchery. Intracerebral pathogenicity index (ICPI) and mean death time (MDT) were determined using commercial live vaccines against Newcastle disease available in Brazil. The ICPI profiles obtained for B1 vaccine strains were nonvirulent and varied from 0 to 0.19, and their MDT was 104-116 hours. The LaSota strains had an ICPI varying between 0.02 and 0.37 and MDT from 92 to 116 hours. ICPI and MDT for the Clone 30 were 0.11 and 104 hours, respectively. For Ulster vaccines, ICPI and MDT were 0 and >150 hours; for VG-GA was 0.03 and 140 hours; and for C2, 0.04 and >144 hours.
