Survival and development of reintroduced Cattleya intermedia plants related to abiotic factors and herbivory at the edge and in the interior of a forest fragment in South Brazil Acta Botanica

ABSTRACT Biotic and abiotic factors, such as luminosity, temperature, air humidity, and herbivory, can affect the establishment of reintroduced plants in natural habitats. This study evaluated the effects of these factors on the survival and growth of Cattleya intermedia plants reintroduced into a forest fragment in South Brazil. Plants of C. intermedia were obtained from in vitro seed germination in asymbiotic culture. Eighty-eight plants were reintroduced at both the forest edge and forest interior. Plants with greater shoot heights and number of leaves and pseudobulbs suffered more damage from herbivores at the edge. Anathallis adenochila is a small epiphytic orchid, endemic to the Atlantic Forest in the southern and southeastern regions of Brazil. Information on the species is scarce and limited to distribution and occurrence data, with no reports about its development. In the present study, plantlets were propagated in vitro, and the influence of different concentrations of macronutrient salts and sucrose on the survival and development of the species was assessed. Asymbiotic culture of Cattleya intermedia Graham (Orchidaceae): the influence of macronutrient salts and sucrose concentrations on survival and development of plantlets Acta Botanica

In this study, C. intermedia plantlets were micropropagated in an asymbiotic culture and the influence of different concentrations of sucrose (15, 30, 45 and 60 g L-1, plus a zero sucrose medium) and macronutrient salts (complete Murashige and Skoog (MS) medium and half MS medium (with half-strength macronutrients)) on survival and development of the plantlets was evaluated. ABSTRACT In order to successfully establish themselves in their natural environment, ferns need habitats with abiotic conditions that are suitable for spore germination and gametophyte development. The objective of this study was to assess the influence of abiotic factors on the initial development of Cyathea phalerata cultivated in vitro. Spore germination and gametophyte development were assessed under varying conditions of surface sterilization, pH, temperature and photoperiod. Germination and sporophytic development of Regnellidium diphyllum Lindm. (Marsileaceae) in the presence of copper Acta Botanica

Regnellidium diphyllum Lindm. is a heterosporous fern growing in wetlands and humid soils that are being converted to agricultural activities. Many products that are used in agriculture contain copper, resulting in surface and groundwater contamination. Germination and initial development tests were performed using Meyer's solution containing copper sulphate at concentrations of 0 (control), 1, 5, 10, 50 and 100 mg L-1. The experiment was conducted in a growth chamber at 25 ± 1ºC for 28 days, with a 12/12-hour light/dark cycle and a photon flux density of 100 µmol m-2 s-1. Influence of antibiotics on embryogenic tissue and Agrobacterium tumefaciens suppression in soybean genetic transformation Bragantia

The influence of different antibiotic treatments in soybean genetic transformation was evaluated. First, an assay was performed to verify how different antibiotic treatments affect soybean embryogenic tissues. The effect of carbenicillin at 500 mg L-1 was genotype-dependent. This antibiotic did not affect embryo survival of cv. IAS5, but a three-fold increase of embryo proliferation was observed for cv. Bragg, when compared to the control. On the other hand, cefotaxime at 350 and 500 mg L-1 caused death of embryogenic tissues of both cultivars. Finally, the association of cefotaxime (250 mg L-1) + vancomycin (250 mg L-1) did not affect negatively the somatic embryos of tested cultivars until 63 days of treatment. In vitro culture of Vriesea gigantea and Vriesea philippocoburgii: two vulnerable bromeliads native to Southern Brazil BABT

Micropropagation studies were carried out using the seeds for establishing an in vitro culture of Vriesea gigantea and Vriesea philippocoburgii. Germination rate of V. gigantea was higher than of V. philippocoburgii. Plantlets of V. philippocoburgii gave rise to many adventitious shoots when cultivated in Knudson basal medium. In contrast, for V. gigantea, a higher salts-concentration was needed, so that the number of shoots was increased by Murashige and Skoog medium. Addition of activated charcoal and naphthaleneacetic acid in regeneration medium allowed the growth of shoots and the formation of roots, confirming the success of in vitro culture. The genotoxicity of untreated and treated sewage from two municipal wastewater treatment plants (WTP BN and WTP SJN) in the municipality of Porto Alegre, in the southern Brazilian state of Rio Grande do Sul, was evaluated over a one-year period using the Tradescantia pallida var. purpurea (Trad-MCN) bioassay. Inflorescences of T. pallida var. purpurea were exposed to sewage samples in February (summer), April (autumn), July (winter) and October (spring) 2009, and the micronuclei (MCN) frequencies were estimated in each period. The aim of this work was to identify Brazilian soybean (Glycine max) genotypes with potential to respond to in vitro culture stimuli for primary somatic embryo induction, secondary embryo proliferation and plant regeneration. Differences among eight tested cultivars were observed at each stage. Two cultivars, IAS-5 and BRSMG 68 Vencedora, were selected for the evaluation of the capacity for embryo differentiation and plant regeneration. Regnellidium diphyllum has its distribution restricted to Southern Brazil and adjoining localities in Uruguay and Argentina. Currently it is on the list of threatened species of Rio Grande do Sul. The conversion of wetlands into agricultural areas or soil contamination by the introduction of waste products and fertilizers may compromise the establishment and survival of this species. Among the pollutants are heavy metals, such as cadmium (Cd). Regeneration of soybean via embryogenic suspension culture Scientia Agricola

In an attempt to establish an alternative plant regeneration system for soybean [Glycine max (L.) Merrill] cultivars used in Brazilian breeding programs, ten genotypes were tested for their embryogenic potential. Cotyledons were removed as explants from immature seeds harvested from field-grown plants. After 45 days on induction medium, the number of responding cotyledons and the number of somatic embryos per immature cotyledon were evaluated. The percentage of explants that produced somatic embryos varied from 1 to 70% among cultivars. The average number of somatic embryos produced per cotyledon pair ranged from 0.01 to 10.3 with a mean of 3.4.
