Arnaud, Christophe; Faury, Nicole; Bouquet, Jean-paul.
The Marennes-Oléron is a well known cupped oyster (Crassostrea gigas) and mussel (Mytilus edulis) reproduction area. Every year, a monitoring of the reproduction of these two species is carried out on behalf of the Regional Shellfish Culture Section of Marennes-Oléron. This monitoring aims to determine the periods of larval emission in order to let the professionals know about the best collection times This study presents the main results obtained in 1999 and compares them with those obtained in 1997 and 1998. The year 1999 showed good results as far as larval emission and development are concerned in both oysters and mussels. The comparison of the three consecutive years shows the disparity of larval evolutions.

Haemocytes from Crassostrea gigas and OsHV-1: A promising in vitro system to study host/virus interactions ArchiMer
Morga, Benjamin; Faury, Nicole; Guesdon, Stephane; Chollet, Bruno; Renault, Tristan.
Since 2008, mass mortality outbreaks associated with the detection of particular variants of OsHV-1 have been reported in Crassostrea gigas spat and juveniles in several countries. Recent studies have reported information on viral replication during experimental infection. Viral DNA and RNA were also detected in the haemolymph and haemocytes suggesting that the virus could circulate through the circulatory system. However, it is unknown if the virus is free in the haemolymph, passively associated at the surface of haemocytes, or able to infect and replicate inside these cells inducing (or not) virion production. In the present study, we collected haemocytes from the haemolymphatic sinus of the adductor muscle of healthy C. gigas spat and exposed them in...

Faury, Nicole; Razet, Daniel; Soletchnik, Patrick; Goulletquer, Philippe; Ratiskol, Jacqueline; Garnier, Jacqueline.
An hydrobiological monitoring survey has been carried out in the Bay of Marennes-Oléron since 1977. Five stations, distributed over the Bay, were sam pied twice a month. Spatial and temporal trends of the physical characteristics (i.e., seawater, temperature, salinity, dissolved oxygen and suspended matter), nutrients (azote, nitrite, nitrate, silicate) and organic variables (chlorophyll a and pheopigments) were assessed by using a statistical time-series analysis mode\. It was used to distinguish seasonal variability from long term trends. Pluviometric data as weil as the Charente River fluxes were similary analyzed by the model. The seasonal variability showed an homogeneous pattern over the five stations for all parameters, but the site-dependent...

In situ localization and tissue distribution of ostreid herpesvirus 1 proteins in infected Pacific oyster, Crassostrea gigas ArchiMer
Martenot, Claire; Segarra, Amelie; Baillon, Laury; Faury, Nicole; Houssin, Maryline; Renault, Tristan.
Immunohistochemistry (IHC) assays were conducted on paraffin sections from experimentally infected spat and unchallenged spat produced in hatchery to determine the tissue distribution of three viral proteins within the Pacific oyster, Crassostrea gigas. Polyclonal antibodies were produced from recombinant proteins corresponding to two putative membrane proteins and one putative apoptosis inhibitor encoded by ORF 25, 72, and 87, respectively. Results were then compared to those obtained by in situ hybridization performed on the same individuals, and showed a substantial agreement according to Landis and Koch numeric scale. Positive signals were mainly observed in connective tissue of gills, mantle, adductor muscle, heart, digestive gland, labial palps, and...

A comparative field study of growth, survival and reproduction of Crassostrea gigas, C-angulata and their hybrids ArchiMer
Soletchnik, Patrick; Huvet, Arnaud; Le Moine, Olivier; Razet, Daniel; Geairon, Philippe; Faury, Nicole; Goulletquer, Philippe; Boudry, Pierre.
This paper reports a comparative study of two cupped oysters, Crassostrea gigas and C angulata, and their hybrids reared in the Marennes-Oleron Sound (France) under usual farming conditions. The relative proficiency of these species was characterized in terms of growth, survival and reproduction, three major concerns for oyster farming. Four populations of each taxon were sampled (C gigas: Japan and France; C angulata: Taiwan and Spain) and used as progenitors to produce six different progenies under common hatchery and nursery conditions (transfer of 5 to 6 g of seed oysters at sea and rearing in bags for 2 years). Mortality profiles differed greatly among the progenies. The C angulata progeny of Taiwanese origin gave the poorest results, with 100%...

Soletchnik, Patrick; Le Moine, Olivier; Faury, Nicole; Razet, Daniel; Geairon, Philippe; Goulletquer, Philippe.
The summer mortality patterns was studied in 1996 on the Ronce-les-Bains oyster bank (175 ha), located on the southern part of the Bay Marennes-Oleron (Atlantic coastline). Studies on growth, sexual maturation, survival rates and environmental variables were compared at fifteen experimental sites (on- and off-bottom culture). After 7 months, cumulative mortality reached 8-19 and 23-33% for off-bottom and on-bottom culture, respectively which is significantly higher for the latter case. A seasonal mortality trend was noted, with a significant increase related to air temperature greater than 20 degrees C and with a significant pre-spawning glycogen catabolism. Based on soft tissue flesh growth results from both types of culture, several empirical models were...

Bougrier, Serge; Masson, Daniel; Geairon, Philippe; Fouche, Dominique; Leroy, Anne; Faury, Nicole; Ratiskol, Jacqueline.
One of the problems faced by oyster farmers in France is the immediate or delayed mortality of oysters in the storage tanks prior to shipping. This may be due to overstocking and/or improper water management. To investigate the problem, experiments were performed in commercial storage tanks stocked with oysters at different densities. The aim of this study was to provide some basic experimental data and guidelines for better handling of oysters prior to shipping

Antiviral immunity in the Pacific oyster, Crassostrea gigas: last development and perspective ArchiMer
Renault, Tristan; Faury, Nicole; Barbosa, Valerie; Moreau, Kevin; Haffner, Philippe; Pepin, Jean-francois.
Herpes-like viruses have been reported last decades in various marine mollusc species in association with mortality outbreaks throughout the world (Renault & Novoa 2004). One of these viruses isolated during French outbreaks has been characterised as an unassigned member of the Herpesviridae family and named ostreid herpes virus 1 (OsHV-1) or Pacific oyster herpes virus (Davison et al. 2005). Studying anti-viral non-specific defence mechanisms (innate immunity) is of great interest, because they constitute the only one in molluscs. Therefore, innate immunity has be investigated in the Pacific cupped oyster, Crassostrea gigas. The main aim of the work was focused on the identification and the characterisation of genes induced by OsHV-1 in the Pacific...

Detection and description of a particular Ostreid herpesvirus 1 genotype associated with massive mortality outbreaks of Pacific oysters, Crassostrea gigas, in France in 2008 ArchiMer
Segarra, Amelie; Pepin, Jean-francois; Arzul, Isabelle; Morga, Benjamin; Faury, Nicole; Renault, Tristan.
Ostreid herpesvirus 1 (OsHV-1) infections have been reported around the world and are associated with high mortalities of the Pacific oyster (Crassostrea gigas). In the summer 2008, abnormal mortality rates ranging from 80% to 100% were reported in France and affected only Pacific oysters. Analyses of oyster samples collected during mortality outbreaks demonstrated a significant detection of OsHV-1 (75% of analysed batches), which appeared stronger than previous years. DNA sequencing based on C and IA regions was carried out on 28 batches of OsHV-1 infected Pacific oysters collected in 2008. Polymorphisms were described in both the C and IA regions and characterized a genotype of OsHV-1 not already reported and termed OsHV-1 μVar. A microsatellite zone...

Long dsRNAs promote an anti-viral response in Pacific oyster hampering ostreid herpesvirus 1 replication ArchiMer

Double-stranded RNA (dsRNA)-mediated genetic interference (RNAi) is a widely used reverse genetic tool for determining the loss-of-function phenotype of a gene. Here, the possible induction of an immune response by long dsRNA was tested in a marine bivalve (Crassostrea gigas), as well as the specific role of the subunit 2 of the nuclear factor kappa B inhibitor (I kappa B2). This gene is a candidate of particular interest for functional investigations in the context of oyster mass mortality events, as Cg-I kappa B2 mRNA levels exhibited significant variation depending on the amount of ostreid herpesvirus 1 (OsHV-1) DNA detected. In the present study, dsRNAs targeting Cg-I kappa B2 and green fluorescent protein genes were injected in vivo into oysters...

Identification of the autophagy pathway in a mollusk bivalve, Crassostrea gigas ArchiMer
Picot, Sandy; Faury, Nicole; Arzul, Isabelle; Chollet, Bruno; Renault, Tristan; Morga, Benjamin.
The Pacific oyster, Crassostrea gigas, is a mollusk bivalve commercially important as a food source. Pacific oysters are subjected to stress and diseases during culture. The autophagy pathway is involved in numerous cellular processes, including responses to starvation, cell death, and microorganism elimination. Autophagy also exists in C. gigas, and plays a role in the immune response against infections. Although this process is well-documented and conserved in most animals, it is still poorly understood in mollusks. To date, no study has provided a complete overview of the molecular mechanism of autophagy in mollusk bivalves. In this study, human and yeast ATG protein sequences and public databases (Uniprot and NCBI) were used to identify protein members...
