Molecular characterization of Paecilomyces fumosoroseus (Deuteromycotina: Hyphomycetes) isolates Scientia Agricola
Azevedo,Andréia Cristiane Souza; Furlaneto,Márcia Cristina; Sosa-Gómez,Daniel Ricardo; Fungaro,Maria Helena Pelegrinelli.
ITS and RAPD analyses were used to investigate molecular variations within samples of Paecilomyces isolates and to resolve five morphologically atypical isolates resembling P. fumosorosus, obtained from whitefly in Northern Paraná State. The ITS4-ITS5 amplicon was 700 base pairs (bp) long in all isolates. The five isolates of Paecilomyces not assigned to species produced restriction profiles identical to all the reference strains of P. fumosoroseus. The extent of fingerprint variability observed by RAPD was sufficient to discriminate all the isolates. The genetic similarity among unidentified isolates and strains of P. fumosoroseus was even higher than that observed among reference strains of this species, allowing us to conclude that isolates CNPso-P77,...

Molecular identification of Aspergillus spp. isolated from coffee beans Scientia Agricola
Magnani,Marciane; Fernandes,Thiago; Prete,Cássio Egidio Cavenaghi; Homechim,Martin; Ono,Elisabete Yurie Sataque; Vilas-Boas,Laurival Antonio; Sartori,Daniele; Furlaneto,Márcia Cristina; Fungaro,Maria Helena Pelegrinelli.
Some species belonging to the genus Aspergillus are potential producers of ochratoxin A (OA), a mycotoxin with nephrotoxic, immunosuppressive, teratogenic and carcinogenic effects. The aim of the present study was to identify the species of Aspergillus that contaminate the inside of coffee beans collected in the stage of maturation and drying, from 16 producing areas located in the northern region of the State of Paraná, in the South of Brazil. A total of 108 isolates of Aspergillus spp. was identified at the species level, by sequencing the internal transcribed spacer (ITS1-5.8S-ITS2) of ribosomal DNA (rDNA). The results revealed the presence of potentially ochratoxigenic species in 82% of the geographic regions studied, among which Aspergillus niger was...

Antimicrobial activity of enterocin obtained from Enterococcus durans on Shiga-like toxin-producing Escherichia coli Ciência Rural
Tosoni,Natara Favaro; Perini,Hugo Felix; Terra,Márcia Regina; Katsuda,Marly Sayuri; Furlaneto,Márcia Cristina; Maia,Luciana Furlaneto.
ABSTRACT: Shiga-like toxin-producing Escherichia coli (STEC) is an important source of food contamination that presents risks to human health. Several industrial food processes eliminate this microorganism; however, these processes can alter the characteristics of the product. Alternative methods of preservation have been identified as an option to control these foodborne pathogens. The purpose of this study was to evaluate the action of bacteriocins produced by Enterococcus durans MF5 in STEC cells. Cell-free supernatant (CFS) containing enterocins from the MF5 isolate was tested over different time points (6, 18, and 24 h). Enterocins present in the crude CFS showed inhibition against STEC at all time points. In the investigation of cell integrity,...

Production of cuticle-degrading proteases by Nomuraea rileyi and its virulence against Anticarsia gemmatalis Ciência Rural
Nunes,Ana Rita Fonseca; Martins,Juliana Nascimento; Furlaneto,Márcia Cristina; Barros,Neiva Monteiro de.
Nomuraea rileyi represents an important natural control agent of Anticarsia gemmatalis preventing populations from reaching economic threshold levels in soybean. During the processes of host infection, entomopathogenic fungi produce extracellular proteases, which degrade the host cuticle and are suggested to be virulence determinants. It was examined the production of subtilisin-like (Pr1) and trypsin-like (Pr2) proteases in two strains (NR458 and CG434) of N. rileyi and its possible role in the process of pathogenicity to this caterpillar. Fungal growth was performed in a mineral medium containing nitrate, and supplemented with the cuticle or exuviae from A. gemmatalis, or with the non-cuticular substrate casein. In medium containing nitrate as sole...
