One Perkinsus species may hide another: characterization of Perkinsus species present in clam production areas of France ArchiMer

Although clam populations in France are known to be infected with protozoans of the genus Perkinsus, no molecular characterization was previously performed on these parasites. Considering that several members of this genus have been associated with mortalities of molluscs worldwide, a study was undertaken in order to characterize these parasites in France. For that purpose, clams, Ruditapes philippinarum and R. decussatus, collected from different production areas and found to be infected with Perkinsus sp. in thioglycolate culture medium, were selected for PCR-RFLP tests and sequencing. Created in 1986, the REPAMO network (Network of mollusc pathology), is a national zoosanitary surveillance network of shellfish health status along the French coasts. Its activities are in keeping with two European Directives, 91 /67/EEC and 95/70/EC and are a patt of the institutional tasks ofIfremer. The aims of the network are to prevent the introduction and spread of exotic pathogens and to survey the evolution of notifiable pathogens already present in France. Depuis 2008, la filière ostréicole française doit faire face à des surmortalités exceptionnelles des naissains d’huîtres creuses, Crassostrea gigas, (huîtres de moins d’un an) comprises entre 60 et 90% dans tous les sites d’élevage. Des cas de mortalité similaires ont été décrits en Irlande, dans les îles anglo-normandes, au Portugal en 2009 et au Royaume-Uni en 2010. En 2008, des mortalités massives d'huîtres creuses (Crassostrea gigas) ont été recensées en France pendant l'été. Ce phénomène, observé de façon récurrente sur les côtés françaises depuis le milieu des années 70, a présenté un caractère exceptionnel en 2008 car il a été observé simultanément dans tous les bassins ostréicoles. De plus, les taux de mortalité calculés ont été particulièrement élevés (80 à 100 % pour de nombreux lots). Molecular detection and quantification of the protozoan Bonamia ostreae in the flat oyster, Ostrea edulis ArchiMer

Bonamia ostreae is an intracellular protozoan which is recognized as a cause of mortality in European populations of flat oysters (Ostrea edulis). Based on the recent characterization of actin genes of B. ostreae, specific primers were designed for real-time PCR using SYBR® Green chemistry. Specificity was demonstrated by the unique melting temperature peak observed in positive samples and by the lack of amplification in samples of oysters infected by closely related parasites, including Bonamia exitiosa. A calibration curve using a cloned template was defined to estimate copy number. Bonamia ostreae and Ostrea edulis : a stable host-parasite system in France? ArchiMer

Bonamiosis due to the intrahaemocytic protistan parasite Bonamia ostreae is a European endemic disease affecting flat oysters Ostrea edulis. After its first description in June 1979 in L'Ile Tudy, Brittany, the parasite rapidly spread to all French oyster farming areas and in other European countries through transfers of live molluscs. Bonamiosis and marteiliosis, another protozoan disease appeared in the seventies, drastically reduced the French flat oyster production from 20 000 tonnes in 1970 to less than 2000 tonnes after 1981. A Large-Scale Epidemiological Study to Identify Bacteria Pathogenic to Pacific Oyster Crassostrea gigas and Correlation Between Virulence and Metalloprotease-like Activity ArchiMer

A 4-year bacteriological survey (2003-2007) of four molluscs cultivated in France and faced with mortality episodes was performed by the French shellfish pathology network. The more abundant bacteria isolated during 92 mortality episodes, occurring mainly in Pacific oyster Crassostrea gigas, were identified by genotyping methods. It allowed us both to confirm the representativeness of Vibrio splendidus and Vibrio aestuarianus bacterial strains and to identify both a large number of Vibrio harveyi-related strains mainly detected during 2007 oyster mortality outbreaks and to a lesser extent bacterial strains identified as Shewanella colwelliana. Ce document compile l’ensemble des bulletins de la surveillance de la santé des mollusques marins émis au cours de l’année 2017 par le réseau REPAMO. Ces bulletins, édités à fréquence mensuelle, relatent les événements de mortalité notifiés au réseau ainsi que les résultats des analyses effectuées lors des prélèvements d’animaux réalisés sur les lieux des mortalités. Les bulletins sont diffusés aux instances professionnelles concernées (CRC, CNC, CDPMEM, CNPMEM, centres techniques), à la DGAL, à la DPMA, aux laboratoires d’analyses, à Ifremer ainsi que sur le site internet du REPAMO. Ils ont pour objectif de tenir les destinataires informés des épisodes de mortalités anormales de mollusques marins rencontrés sur le littoral français métropolitain et corse.

Mass mortality outbreaks (90-100%) in both juvenile and adult blue mussels were reported in 2014 along the Pertuis Charentais area of the French Atlantic coast. A series of studies were carried out to learn more about the phenomenon through detecting pathogens, describing environmental conditions and establishing the physiological status of the animals. Bacteria identif ied as belonging to the V. splendidus species were detected in dying mussels and seem able to cause mortality in blue mussels in the laboratory. Unusual environmental conditions (large amounts of fresh water, re-suspension of sediments during a series of storms, and a high renewal rate of specific water masses in Brittany’s Pertuis area) may be partially responsible for sparking the phenomen

At the present time losses due to diseases are one of main factors limiting the aquaculture development. Unceasing increase of exchanges between different countries favours the risk of introduction and propagation of new pathogen agents. First description of French V. tubiashii strains pathogenic to mollusk: I. Characterization of isolates and detection during mortality events ArchiMer

Nine dominant bacterial isolates were obtained from different batches of Crassostrea gigas spat experiencing high mortality rates in a French experimental hatchery/nursery in 2007. Using phenotypic analysis combined with multilocus sequence analysis, the isolates were shown to be genetically close to the Vibrio tubiashii type strain. Based on (1) analyses of the recA gene sequences; (2) the results of DNA–DNA hybridization assays between 07/118 T2 (LMG 27884 = CECT 8426), which is a representative strain, and the V. tubiashii type strain (69%); and (3) phenotypic traits, the bacteria were classified in a group close to American V. tubiashii strain. First results of an epidemiological study on oyster (Crassostrea gigas) mortality events in France during summer 2008 ArchiMer

Abnormal mortalities occurred in most oyster producing areas in France during the summer of 2008. The mortalities were sudden and severe (up to 100%) and mainly affected 6 to 18 month old juveniles. Only Crassostrea gigas species was affected. The cause of the mortality was unclear but OsHV-1 virus and bacteria belonging to Vibrionaceae family were frequently detected in affected populations. An epidemiological study was commenced in autumn 2008 in order to identify factors associated with, and the cause (or causes) of these mortalities.
