In the context of the fish farming industry needing to substitute the animal food (meal and oil) by vegetable ingredients, the main objective of this thesis was to study the long-term effects of a diet exclusively composed by plant products on the metabolism of the european sea bass (Dicentrarchus labrax) and more particularly, on its capacity to biosynthesis the Highly Unsaturated Fatty Acids (HUFA) from the PolyUnsaturated Fatty acids (PUFA) brought by this vegetal diet. Our transcriptomic approaches showed that the use of a plant food disrupted the lipids metabolism (cholesterol and fatty acids) and the energy metabolism (carbohydrates, amino acids) in the liver, but also affected other physiological processes such as the immune system. Characteristics of fads2 gene expression and putative promoter in European sea bass (Dicentrarchus labrax): Comparison with salmonid species and analysis of CpG methylation ArchiMer

Marine fish species exhibit low capacity to biosynthesise highly unsaturated fatty acid (HUFA) in comparison to strict freshwater and anadromous species. It is admitted that the Delta(6) desaturase (FADS2) is a key enzyme in the HUFA biosynthetic pathway. We investigated by quantitative PCR the relative amounts of FADS2 mRNA in European sea bass (Dicentrarchus labrax) in comparison with a salmonid species, the rainbow trout (Oncorhynchus mykiss L.). Fish are the main source of the n-3 highly unsaturated fatty acids, which are crucial for human health. Their synthesis from C18 precursors is mediated by desaturases and elongases, but the activity of these enzymes has not been conclusively established in marine fish species. This study reports the cloning, tissue expression, and functional characterization of a sea bass (Dicentrarchus labrax L.) Δ6-desaturase and one of its splicing variants. Two cDNAs with open reading frames of 1,346 and 1,354 bp were cloned and named D6D and D6D-V, respectively. Both deduced protein sequences (445 and 387 amino acids, respectively) contained two transmembrane regions and the N-terminal cytochrome b5 domain with the HPGG motif characteristic of microsomal desaturases....

It is known in vertebrates that endothelin-1 (ET-1) plays a key role ill morphogenesis whose expression level greatly influences the development of craniofacial malformations. In the Present study, the complete cDNA fragment encoding a precursor of endothelin-1, the preproendothelin-1 (PPET1), was cloned by RACE-PCR from European sea bass. The cDNA encoded a 199 amino acid polypeptide that was composed of the "mature" and "big" ET-1. Supplies of marine fish oils are limited, and continued growth in aquaculture production dictates that lipid substitutes in fish diets must be used without compromising fish health and product quality. In this study, the total substitution of a fish meal and fish oil by a blend of vegetable meals (corn, soybean, wheat and lupin) and linseed oil in the diet of European sea bass (Dicentrachus labrax) was investigated. Two groups of European sea bass were fed with fish diet (FD) or vegetable diet (VD) for 9 months.
