A re-appraisal of Harknessia (Diaporthales), and the introduction of Harknessiaceae fam. nov. Naturalis
Crous, P.W.; Summerell, B.A.; Shivas, R.G.; Carnegie, A.J.; Groenewald, J.Z..
Harknessiaceae is introduced as a new family in the ascomycete order Diaporthales to accommodate species of Harknessia with their Wuestneia-like teleomorphs. The family is distinguished by having pycnidial conidiomata with brown, furfuraceous margins, brown conidia with hyaline, tube-like basal appendages, longitudinal striations, and rhexolytic secession. Six species occurring on Eucalyptus are newly introduced, namely H. australiensis, H. ellipsoidea, H. pseudohawaiiensis, and H. ravenstreetina from Australia, H. kleinzeeina from South Africa, and H. viterboensis from Italy. Epitypes are designated for H. spermatoidea and H. weresubiae, both also occurring on Eucalyptus. Members of Harknessia are commonly associated with leaf spots, but also occur as...

Riding with the ants Naturalis
Duarte, A.P.M.; Attili-Angelis, D.; Baron, N.C.; Groenewald, J.Z.; Crous, P.W.; Pagnocca, F.C..
Isolates of Teratosphaeriaceae have frequently been found in the integument of attine ants, proving to be common and diverse in this microenvironment. The LSU phylogeny of the ant-isolated strains studied revealed that they cluster in two main lineages. The first was associated with the genus Xenopenidiella whereas the other represented two ant-isolated lineages sister to the taxa Penidiella aggregata and P. drakensbergensis, which are allocated to the new genus Penidiellomyces. The genus Penidiella is limited to the lineage containing P. columbiana, which is not congeneric with Penidiellomyces or Penidiellopsis, nor with Simplicidiella, a novel genus introduced here to accommodate a strain isolated from ants. For species level analysis, the final 26...

Why everlastings don’t last Naturalis
Crous, P.W.; Groenewald, J.Z..
The Cape Floral Region represents one of the world’s biodiversity hot spots, with a high level of plant, animal and insect endemism. The fungi occurring in this region, however, remain poorly studied. It is widely postulated that each plant species should harbour at least five to six unique fungal species, a number that we regard to be a huge underestimate. To test this hypothesis, we decided to study a single senescent flower of Phaenocoma prolifera (‘everlasting’; Asteraceae) collected in South Africa, and posed the question as to how many different species of fungi could be isolated and cultivated from 10 leaf bracts. Using a damp chamber technique, numerous microfungi could be induced to sporulate, enabling most of them to be successfully isolated on...

A case for re-inventory of Australia’s plant pathogens Naturalis
Hyde, K.D.; Chomnunti, P.; Crous, P.W.; Groenewald, J.Z.; Damm, U.; Ko Ko, T.W.; Shivas, R.G.; Summerell, B.A.; Tan, Y.P..
Several filamentous oomycete species of the genus Halophytophthora have recently been described from marine environments, mostly from subtropical and tropical ecosystems. During a survey of oomycetes from leaf litter of Spartina alterniflora in salt marshes of southeastern Georgia, isolates of four taxa were recovered that bore similarity to some members of Halophytophthora but were highly divergent from isolates of Halophytophthora s.str. based on a combined sequence analysis of two nuclear loci. In phylogenetic analyses, these isolates were placed basal to a monophyletic group comprised of Pythium of the Pythiaceae and the Peronosporaceae. Sequence and morphology of these taxa diverged from the type species Halophytophthora vesicula, which was placed...

Species of Mycosphaerella and related anamorphs on Eucalyptus leaves from Thailand Naturalis
Crous, R. Cheewangkoon: P.W.; Hyde, K.D.; Groenewald, J.Z.; To-anan, C..
Species of Mycosphaerella and their related anamorphs represent potentially serious foliar pathogens of Eucalyptus. The fungi treated in the present study were isolated from symptomatic Eucalyptus leaves collected in Thailand during June –October 2007. Species were initially identified based on morphological and cultural characteristics. Identifications were confirmed using comparisons of DNA sequence data of the internal transcribed spacers (ITS1, 5.8S nrDNA, ITS2) and the 28S nrDNA (LSU) regions. To help distinguish species of Pseudocercospora, the dataset was expanded by generating partial sequences of the translation elongation factor 1-α and actin genes. By integrating the morphological and molecular datasets, five new taxa were distinguished, namely...

Multiple gene genealogies and phenotypic characters differentiate several novel species of Mycosphaerella and related anamorphs on banana Naturalis
Arzanlou, M.; Groenewald, J.Z.; Fullerton, R.A.; Abeln, E.C.A.; Carlier, J.; Zapater, M.-F.; Buddenhagen, I.W.; Viljoen, A.; Crous, P.W..
Three species of Mycosphaerella, namely M. eumusae, M. fijiensis, and M. musicola are involved in the Sigatoka disease complex of bananas. Besides these three primary pathogens, several additional species of Mycosphaerella or their anamorphs have been described from Musa. However, very little is known about these taxa, and for the majority of these species no culture or DNA is available for study. In the present study, we collected a global set of Mycosphaerella strains from banana, and compared them by means of morphology and a multi-gene nucleotide sequence data set. The phylogeny inferred from the ITS region and the combined data set containing partial gene sequences of the actin gene, the small subunit mitochondrial ribosomal DNA and the histone H3...

Reassessing Vermisporium (Amphisphaeriaceae), a genus of foliar pathogens of eucalypts Naturalis
Barber, P.A.; Crous, P.W.; Groenewald, J.Z.; Pascoe, I.G.; Keane, P..
The genus Vermisporium presently accommodates 13 species, 11 of which are associated with leaf spots of eucalypts in the Southern Hemisphere. Vermisporium is chiefly distinguished from Seimatosporium (Amphisphaeriaceae) on the basis of a short exogenous basal appendage, and the absence of a recognisable apical appendage. Due to the increasing importance of these species in native forests, and confusion pertaining to their taxonomy, a revision of the genus was undertaken based on fresh collections and dried herbarium specimens. Results from DNA sequence data analyses of the nrDNA-ITS and 28S nrRNA genes for species of Vermisporium indicated the genus to be a synonym of Seimatosporium. New combinations are introduced in Seimatosporium for several species: S....

In the present study, sequence data for the ITS nrDNA region were used in conjunction with morphological observations to resolve the taxonomy of species of Botryosphaeriaceae associated with Proteaceae. Neofusicoccum luteum was confirmed from Buckinghamia and Banksia in Australia, and on Protea cynaroides in South Africa. A major pathogen of Banksia coccinea in Australia was shown to be N. australe and not N. luteum as previously reported. Neofusicoccum protearum was previously reported on Proteaceae from Australia, Madeira, Portugal and South Africa, and is shown here to also occur in Hawaii and Tenerife (Canary Islands). Furthermore, several...

Species of Leucadendron, Leucospermum and Protea (Proteaceae) are in high demand for the international floriculture market due to their brightly coloured and textured flowers or bracts. Fungal pathogens, however, create a serious problem in cultivating flawless blooms. The aim of the present study was to characterise several of these pathogens using morphology, culture characteristics, and DNA sequence data of the rRNA-ITS and LSU genes. In some cases additional genes such as TEF 1-α and CHS were also sequenced. Based on the results of this study, several novel species and genera are described. Brunneosphaerella leaf blight is shown to be caused by three species, namely B. jonkershoekensis on Protea repens, B. nitidae sp. nov. on Protea nitida and B....
