Growth-related characters, condition factor, and genetic differentiation were investigated for a single cohort of young-of-the-year (YOY) sole within and among nurseries with differing levels of heavy metals (Cd, Cu and Zn) contamination in the two Charentais Straits, Bay of Biscay, France. Fasting and refeeding occur naturally in predators but this is largely ignored when dealing with farmed fish. Therefore, the effects of 3-week fasting and re-alimentation (2.5% of the individual body mass) were investigated using two genetically selected populations (F2 generation) of 250 g juvenile sea bass (Dicentrarchus labrax L.). Spatial and temporal population genetic structures of the common sole, Solea solea, were studied in Northeastern Atlantic and Mediterranean Sea populations, using three polymorphic exon-primed intron-crossing (EPIC) markers. Results demonstrated significant multilocus differentiation among Eastern Mediterranean and a group composed by Western Mediterranean and Atlantic populations (theta = 0.150, P < 0.001), but also suggested unrecorded genetic differentiation of the Adriatic Sea population. Feed efficiency is a major goal for aquaculture sustainability, and selecting fish to genetically enhance this trait would be highly valuable. However, no selective breeding program specifically targeted to feed efficiency exists for farmed fish, mostly because of the difficulty of measuring individual feed intake. We report the isolation, development and multiplex optimisation of 12 new microsatellite loci for the great scallop, Pecten maximus. Diversity was moderate to high, with number of alleles ranging from 4 to 20 and observed heterozygosity between 0.28 and 0.88. Progeny produced in a commercial hatchery was used to test locus power for parentage assignment. The percentage of offspring that was unambiguously assigned to a unique pair of parents was 97% (software package CERVUS-COLONY). Parentage assignment revealed that 22% of the studied progeny resulted from unplanned crosses. Effective population size of the study progeny was also estimated. Our study illustrates the power of microsatellites for the genetic monitoring of hatchery-produced great scallops. |
