Programme Pathologie - Rapport scientifique Année 1994 - Contrat de Plan Etat Région Poitou-Charentes 1994-1998 - Convention 94 RPC R 62 ArchiMer
Renault, Tristan; Cochennec, Nathalie; Le Deuff, Rose-marie; Chollet, Bruno; Maffart, Patricia; Haffner, Philippe; Berthe, Franck; Thuillier, Benoit.

Segarra, Amelie; Faury, Nicole; Haffner, Philippe; Pepin, Jean-francois; Tourbiez, Delphine; Trancart, Suzanne; Travers, Marie-agnes; Renault, Tristan.

Renault, Tristan; Berthe, Franck; Thebault, Anne; Lipart, Cecile; Chollet, Bruno; Cochennec, Nathalie; Haffner, Philippe.

Mass mortalities in hatchery-reared sea bass (Lates calcarifer) larvae associated with the presence in the brain and retina of virus-like particles ArchiMer
Renault, Tristan; Haffner, Philippe; Baudin Laurencien, F.; Breuil, Gilles; Bonami, Jean-robert.
Since 1986, an unknown disease causing high mortalities appeared in Ifremer's Pacific OceanologicaI Center (Tahiti) nursery of sea bass (Lates calcarifer). opathology of lesions involving both centraI nervous system (CNS) and eye retina, and the tuation exists in South-East Asian countries (Indonesia, Philippines, Malaysia and Singapore), and in northem Australia. This paper describes the histmorphology of a new fish virus, which is believed to be the aetiological agent of high mortalities among sea bass larvae.

Antiviral immunity in the Pacific oyster, Crassostrea gigas: last development and perspective ArchiMer
Renault, Tristan; Faury, Nicole; Barbosa, Valerie; Moreau, Kevin; Haffner, Philippe; Pepin, Jean-francois.
Herpes-like viruses have been reported last decades in various marine mollusc species in association with mortality outbreaks throughout the world (Renault & Novoa 2004). One of these viruses isolated during French outbreaks has been characterised as an unassigned member of the Herpesviridae family and named ostreid herpes virus 1 (OsHV-1) or Pacific oyster herpes virus (Davison et al. 2005). Studying anti-viral non-specific defence mechanisms (innate immunity) is of great interest, because they constitute the only one in molluscs. Therefore, innate immunity has be investigated in the Pacific cupped oyster, Crassostrea gigas. The main aim of the work was focused on the identification and the characterisation of genes induced by OsHV-1 in the Pacific...

First evidence of a potential antibacterial activity involving a laccase-type enzyme of the phenoloxidase system in Pacific oyster Crassostrea gigas haemocytes ArchiMer
Luna-acosta, Andrea; Saulnier, Denis; Pommier, Mylene; Haffner, Philippe; De Decker, Sophie; Renault, Tristan; Thomas-guyon, Helene.
Phenoloxidases (POs) are a group of copper proteins including tyrosinase, catecholase and laccase. In several insects and crustaceans, antibacterial substances are produced through the PO cascade, participating in the direct killing of invading microorganisms. However, although POs are widely recognised as an integral part of the invertebrate immune defence system, experimental evidence is lacking that these properties are conserved in molluscs, and more particularly in the Pacific oyster Crassostrea gigas. In the present study, Vibrio splendidus LGP32 and Vibrio aestuarianus 02/041 growths were affected, after being treated with C. gigas haemocyte lysate supernatant (HLS), and either a common substrate of POs, l-3,4-dihydroxyphenylalanine (l-DOPA), to...

A Large-Scale Epidemiological Study to Identify Bacteria Pathogenic to Pacific Oyster Crassostrea gigas and Correlation Between Virulence and Metalloprotease-like Activity ArchiMer
Saulnier, Denis; De Decker, Sophie; Haffner, Philippe; Cobret, Laetitia; Robert, Maeva; Garcia, Celine.
A 4-year bacteriological survey (2003-2007) of four molluscs cultivated in France and faced with mortality episodes was performed by the French shellfish pathology network. The more abundant bacteria isolated during 92 mortality episodes, occurring mainly in Pacific oyster Crassostrea gigas, were identified by genotyping methods. It allowed us both to confirm the representativeness of Vibrio splendidus and Vibrio aestuarianus bacterial strains and to identify both a large number of Vibrio harveyi-related strains mainly detected during 2007 oyster mortality outbreaks and to a lesser extent bacterial strains identified as Shewanella colwelliana. Because metalloprotease has been reported to constitute a virulence factor in a few Vibrio strains pathogenic for...

Infectious diseases are mostly explored using reductionist approaches despite repeated evidence showing them to be strongly in fluenced by numerous interacting host and environmental factors. Many diseases with a complex aetiology therefore remain misunderstood. By developing a holistic approach to tackle the complexity of interactions, we decipher the complex intra-host interactions underlying Pacific oyster mortality syndrome affecting juveniles of Crassostrea gigas, the main oyster species exploited worldwide. Using experimental infections reproducing the natural route of infection and combining thorough molecular analyses of oyster families with contrasted susceptibilities, we demonstrate that the disease is caused by multiple infection with an initial...

Suivi bactérien d'un élevage larvaire de crevettes péneïdes - Ecloserie CAMCAR, 12 janvier - 12 février 1991 ArchiMer
Haffner, Philippe; Berthe, Franck.

Mass mortality outbreaks (90-100%) in both juvenile and adult blue mussels were reported in 2014 along the Pertuis Charentais area of the French Atlantic coast. A series of studies were carried out to learn more about the phenomenon through detecting pathogens, describing environmental conditions and establishing the physiological status of the animals. Bacteria identif ied as belonging to the V. splendidus species were detected in dying mussels and seem able to cause mortality in blue mussels in the laboratory. Unusual environmental conditions (large amounts of fresh water, re-suspension of sediments during a series of storms, and a high renewal rate of specific water masses in Brittany’s Pertuis area) may be partially responsible for sparking the phenomen

Nine dominant bacterial isolates were obtained from different batches of Crassostrea gigas spat experiencing high mortality rates in a French experimental hatchery/nursery in 2007. Using phenotypic analysis combined with multilocus sequence analysis, the isolates were shown to be genetically close to the Vibrio tubiashii type strain. Based on (1) analyses of the recA gene sequences; (2) the results of DNA–DNA hybridization assays between 07/118 T2 (LMG 27884 = CECT 8426), which is a representative strain, and the V. tubiashii type strain (69%); and (3) phenotypic traits, the bacteria were classified in a group close to American V. tubiashii strain. Species-specific mechanisms of cytotoxicity toward immune cells determine the successful outcome of Vibrio infections ArchiMer

Vibrio species cause infectious diseases in humans and animals, but they can also live as commensals within their host tissues. How Vibrio subverts the host defenses to mount a successful infection remains poorly understood, and this knowledge is critical for predicting and managing disease. Here, we have investigated the cellular and molecular mechanisms underpinning infection and colonization of 2 virulent Vibrio species in an ecologically relevant host model, oyster, to study interactions with marine Vibrio species. All Vibrio strains were recognized by the immune system, but only nonvirulent strains were controlled. We showed that virulent strains were cytotoxic to hemocytes, oyster immune cells. By analyzing host and bacterial transcriptional...

A field report on vibrio disease of seabass (Dicentrarchus labrax ) in the South of France ArchiMer
Breuil, Gilles; Haffner, Philippe.
Vibrio anguillarum was isolated from diseased seabass reared in aquatic farms (hatcheries and grow-out farms) located along the French Mediterranean coast. A representative strain (V62) differs from the French V. anguillarum strain (V408) by its ability to grow on a 0% salinity medium. The pathogenicity of the representative strain for sea bass was confirmed by injections and bath challenge. The V62 strain reacted with V. anguillarum antiserum (V408) and 84% of the isolated strains from diseased sea bass agglutinated with anti V62 serum.

Vibrio splendidus O-antigen structure: A trade-off between virulence to oysters and resistance to grazers ArchiMer
Oyanedel, Daniel; Labreuche, Yannick; Bruto, Maxime; Amraoui, Hajar; Robino, Etienne; Haffner, Philippe; Rubio, Tristan; Charrière, Guillaume; Le Roux, Frederique; Destoumieux-garzón, Delphine.
A major debate in evolutionary biology is whether virulence is maintained as an adaptive trait and/or evolves to non‐virulence. In the environment, virulence traits of non‐obligatory parasites are subjected to diverse selective pressures and trade‐offs. Here we focus on a population of Vibrio splendidus that displays moderate virulence for oysters. A MARTX (Multifunctional‐autoprocessing repeats‐in‐toxin) and a type‐six secretion system (T6SS) were found to be necessary for virulence toward oysters, while a region (wbe) involved in O‐antigen synthesis is necessary for resistance to predation against amoebae. Gene inactivation within the wbe region had major consequences on the O‐antigen structure, conferring lower immunogenicity, competitive advantage and...
