Several molecular characteristics of the Ribgrass mosaic virus - Impatiens (RMV-Imp) were investigated in this thesis including the evaluation of three mutation constructs in virulence and virus accumulation on Nicotiana (N.) benthamiana and N. tabacum. Regarding a strategy of expressing foreign genes in plants, two diverged RMV-based expression vectors containing a green fluorescent protein were cloned into the binary vector pPZP200 for subsequent agroinfiltration work. An infectious full-length clone of RMV and three RMV-based mutants already existed. They vary depending on which of the two putative ORFs is inactivated. An additional mutation site of one of these mutation constructs was discovered and reverse mutated. After the inoculation of the... |

The Tansley review by Ryan & Graham (2018) provided a welcome critical perspective on the role of arbuscular mycorrhizal (AM) fungi in large‐scale industrial agriculture, with a focus on cereals (wheat, Triticum aestivum). They conclude that there is little evidence that farmers should consider the abundance or diversity of AM fungi when managing crops. We welcome many of the points made in the paper, as they give an opportunity for self‐reflection, considering that the importance of AM fungi in agroecosystems is often taken for granted. However, we suggest that it is too early to draw the overall conclusion that the management of AM fungi by farmers is currently not warranted. We offer the following points to contribute to the discussion. The first... |

Introduction Mixed cropping, i.e. mixing different crops in the same field, provides agronomic advantages as increased productivity under low inputs conditions (e.g. for organic farming: Bedoussac et al. 2015) and higher yield stability (Raseduzzaman and Jensen 2017). In mixed cropping, choosing the right cultivars is critical for the performance of the mixture, as shown for pea-barley mixtures (Hauggaard-Nielsen and Jensen 2001) and maize-bean mixtures (Hoppe 2016). As performance in pure stand can strongly diverge from performance in mixture, estimating the ability of a cultivar to be mixed with another crop is therefore of utmost importance. For this purpose, concepts of General and Specific Combining Ability in hybrid breeding (Griffing 1956) have... |

Three isolates of different Trichoderma species were selected for this study. T. hamatum (LU592) and T. harzianum (LU686), were known to stimulate growth and improve establishment of P. radiata seedlings, and T. atroviride (LU132) had no stimulatory activity. The ability of these isolates to establish and survive in bulk soil, the rhizosphere, rhizoplane or endorhizosphere of P. radiata seedlings was determined. In addition, the effect of each isolate on P. radiata seedling development and growth was assessed and is currently being confirmed in a large scale commercial experiment. From the nursery experiment, the Trichoderma isolate which was the most effective at colonising all P. radiata root subsystems (LU592) was selected for more detailed ecological... |

Crop mixtures consisting of cereals and legumes have proven as a well-adapted arrangement due to their complementarity towards important resources, especially nitrogen. Crop mixtures combine high yield performance and yield stability. They can contribute to a diversified cropping landscape and adaptation to climate change. The search for alternatives to protein imports from overseas and investments in post-harvest separation technologies are currently fostering their adoption by farmers in Western-Europe, especially under organic and lowinput farming conditions. However, screening and breeding for mixed cropping has hardly been explored for arable crops. Thus, the objective was to develop novel breeding strategies and tools specifically for mixed cropping... |

Introduction and methods Crop mixtures of cereals and legumes have received recent attention in the scope of climate change and local protein supply. Cereal-legume mixtures are known to increase yield and yield stability compared with pure crops. Thus dedicated breeding strategies for mixed cropping might become necessary. As a model crop, we compared 32 current European spring pea (Pisum sativum L.) and 8 current spring barley (Hordeum vulgare L.) cultivars for their yield performance in mixed and pure stands, with pea as the focal species. Trials were done as an incomplete factorial design (Fig. 1) at two locations in Switzerland with two replications (Fig. 2). We calculated variance components for the general and specific mixing ability (GMA and... |
