Ouabain exacerbates botulinum neurotoxin-induced muscle paralysis via progression of muscle atrophy in mice OAK
Fujikawa, Ryu; Muroi, Yoshikage; Unno, Toshihiro; Ishii, Toshiaki; 室井, 喜景; 石井, 利明.
Botulinum neurotoxin serotype A (BoNT/A) inhibits acetylcholine release at the neuromuscular junction in isolated muscles, and ouabain can partially block its effect. However, it is not clear whether ouabain attenuates BoNT/A-induced neuromuscular paralysis in vivo. In this work, we investigated the effects of ouabain on BoNT/A-induced neuromuscular paralysis in mice. Ouabain was administered to mice intraperitoneally immediately after a single injection of BoNT/A into skeletal muscle. The effects of ouabain on BoNT/A-induced muscle paralysis were assessed by quantitative monitoring of muscle tension and digit abduction via the digit abduction scoring (DAS) assay. A single administration of ouabain significantly prolonged BoNT/A-induced neuromuscular...

The mesencephalic trigeminal sensory nucleus is involved in the control of feeding and exploratory behavior in mice OAK
Ishii, Toshiaki; Furuoka, Hidefumi; Ito, Takuya; Kitamura, Nobuo; Nishimura, Masakazu.
The mesencephalic trigeminal nucleus (Me5), which receives input from oral proprioceptors and projects to higher brain regions, is involved in mastication-induced modulation of satiation. To investigate how the Me5 is involved in the control of feeding and exploratory behavior, we examined the effect of bilateral electrolytic lesions of the Me5 on feeding and exploratory behavior in mice. Mouse feeding and exploratory behaviors were analyzed using a food- search-compulsion-apparatus (FSCA), which was designed to distinguish between the two behaviors under standard living conditions. To assess anxiety in mice in an unfamiliar environment, exploratory activity was analyzed in an automated hole-board apparatus. Mice with bilateral Me5 lesions had unique...

Comparison of growth and exploratory behavior in mice fed an exclusively milk formula diet and mice fed a food-pellet diet post weaning OAK
Ishii, Toshiaki; Ito, Takuya; Nishimura, Masakazu; 石井, 利明.
An exclusively milk formula diet stunted the growth of mice immediately following weaning. Milk-fed mice displayed a low-frequency profile of exploratory behavior, while pellet-fed mice showed high-frequency exploration. In contrast to exploratory behavior, feeding behavior did not differ significantly between milk- and pellet-fed mice. Despite showing low-frequency exploratory behavior, mice on an exclusively milk formula diet showed no difference in behavioral activities analyzed by an automatic hole-board apparatus compared to pellet-fed mice. These results suggest that the growth stunt caused by an exclusively milk formula diet retards the acquisition of active exploratory behavior without affecting the emotional state of mice.

Integrin-linked kinase controls neurite outgrowth in N1E-115 neuroblastoma cells OAK
Ishii, Toshiaki; Satoh, Eiki; Nishimura, Masakazu; 石井, 利明; 佐藤, 栄輝.
Mouse N1E-115 cells grown on a laminin matrix exhibit neurite outgrowth in response to serum deprivation. Treatment of cells with an antibody against beta (1) integrin inhibits neurite outgrowth. Thus, beta (1) integrin is involved in the neuritogenesis of N1E-115 cells on a laminin matrix. Integrin-linked kinase (ILK), a recently identified cytoplasmic serine/threonine protein kinase that binds to the cytoplasmic domain of beta (1) integrin, has an important role in transmembrane signal transduction via integrins. We report that ILK is expressed in N1E-115 cells, the expression levels of which are constant under both normal and differentiating conditions. A stable transfection of a kinase-deficient mutant of ILK (DN-ILK) results in inhibition of neurite...

Integrin-linked kinase is involved in lactoferrin-induced anchorage-independent cell growth and survival in PC12 cells OAK
Ishii, Toshiaki; Uto, Tomoka; Mori, Kaori; Fujikawa, Ryu; 石井, 利明.
Aims: Bovine lactoferrin (bLf) causes anchorage-independent cell growth in PC12 cells. The present study investigated the mechanisms involved in bLf-induced anchorage-independent cell growth and survival in PC12 cells. Main methods: The number of adherent cells and suspended cells was estimated separately by using a methyl thiazol tetrazolium (MTT) assay, and the sum of both optical density (O.D.) (570 nm) values was used as a measure of the total number of cells. Key findings: Integrin-linked kinase (ILK) plays an important role in integrin and growth factor signaling pathways. Stable transfection of PC12 cells with a dominant negative kinase-deficient mutant of ILK (DN-ILK) inhibited bLf-induced anchorage-independent cell growth. The ILK activity in the...

Inactivation of integrin-linked kinase induces aberrant tau phosphorylation via sustained activation of glycogen synthase kinase 3 beta in N1E-115 neuroblastoma cells OAK
Ishii, Toshiaki; Furuoka, Hidefumi; Muroi, Yoshikage; Nishimura, Masakazu; 石井, 利明; 古岡, 秀文; 室井, 喜景.
Integrin-linked kinase (ILK) is a focal adhesion serine/ threonine protein kinase with an important role in integrin and growth factor signaling pathways. Recently, we demonstrated that ILK is expressed in N1E-115 neuroblastoma cells and controls integrin-dependent neurite outgrowth in serum-starved cells grown on laminin (Ishii, T., Satoh, E., and Nishimura, M. ( 2001) J. Biol. Chem. 276, 42994 - 43003). Here we report that ILK controls tau phosphorylation via regulation of glycogen synthase kinase-3beta (GSK-3beta) activity in N1E-115 cells. Stable transfection of a kinase-deficient ILK mutant (DN-ILK) resulted in aberrant tau phosphorylation in N1E-115 cells at sites recognized by the Tau-1 antibody that are identical to some of the phosphorylation...

Rolipram improves facilitation of contextual fear extinction in the 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-induced mouse model of Parkinson's disease OAK
Kinoshita, Ken-ichi; Muroi, Yoshikage; Unno, Toshihiro; Ishii, Toshiaki.
Cognitive impairment often occurs in Parkinson's disease (PD), but the mechanism of onset remains unknown. Recently, we reported that PD model mice produced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) show facilitation of hippocampal memory extinction, which may be the cause of cognitive impairment in PD. When we examined the cAMP/CREB signaling in the hippocampus, decreased levels of cAMP and phosphorylated CREB were observed in the dentate gyrus (DG) of MPTP-treated mice. Administration of rolipram improved the memory deficits with concomitant recovery of cAMP and phosphorylated CREB levels, suggesting that reduced cAMP/CREB signaling in the DG leads to cognitive impairment in MPTP-treated mice. The mesencephalic trigeminal sensory nucleus is involved in acquisition of active exploratory behavior induced by changing from a diet of exclusively milk formula to food pellets in mice OAK
Ishii, Toshiaki; Furuoka, Hidefumi; Kitamura, Nobuo; Muroi, Yoshikage; Nishimura, Masakazu; 古岡, 秀文; 北村, 延夫; 室井, 喜景; 石井, 利明.
Post-weaning mice fed exclusively milk display low-frequency exploratory behavior [Ishii, T., Itou, T., and Nishimura, M. (2005) Life Sci. 78, 174-179] compared to mice fed a food pellet diet. This low-frequency exploratory behavior switched to high-frequency exploration after a switch from exclusively milk formula to a food pellet diet. Acquisition of the high-frequency exploratory behavior was irreversible. Recently, we demonstrated that the mesencephalic trigeminal nucleus (Me5) is involved in the control of feeding and exploratory behavior in mice without modulating the emotional state [Ishii, T., Furuoka, H., Itou, T., Kitamura, N., and Nishimura, M. (2005) Brain Res. 1048, 80-86]. We therefore investigated whether the Me5 is involved in acquisition...

Volatile female odors activate the accessory olfactory system of male mice without physical contact OAK
Muroi, Yoshikage; Ishii, Toshiaki; Komori, Seiichi; Kitamura, Nobuo; Nishimura, Masakazu; 室井, 喜景; 石井, 利明; 北村, 延夫.
We previously reported that male mice are more attracted to volatile odors from intact female mice than from ovarlectornized female mice. In the present study, we investigated male attraction to volatile odors from soiled bedding collected from the cages of estrous or ovariectomized female mice. There was no difference in the total time spent sniffing volatile odors from estrous and ovariectomized female mice, suggesting that female mice emit volatile odors which are not excreted into bedding. To test this possibility, we investigated c-Fos expression in the mitral cell layer and granule cell layer of the accessory olfactory bulb 60 min after exposure of male mice to volatile odors without physical contact. Volatile odors from an estrous female mouse...

Role of Integrin-Linked Kinase in Neuronal Cells OAK
Ishii, Toshiaki.
Integrin-linked kinase (ILK) is a focal adhesion serine/threonine protein kinase that binds to the cytoplasmic domain of β1 integrin and has an important role in integrin and growth factor signaling pathways. Clustering of integrins on the cell surface in contact with the extracellular matrix induces focal adhesion that recruits numerous mitogenic signaling proteins other than ILK, such as growth factor receptors, mitogenactivated protein kinase, and small GTP-binding proteins, to integrin receptors and forms signaling centers where adhesive and mitogenic pathways can integrate. ILK is highly expressed in neuronal cells and its enzyme activity is activated by cell adhesion on the extracellular matrix in a phosphatidylinositol 3-kinasedependent manner....

Bilateral lesions of the mesencephalic trigeminal sensory nucleus stimulate hippocampal neurogenesis but lead to severe deficits in spatial memory resetting OAK
Ishii, Toshiaki; Suenaga, Ryuta; Iwata, Wataru; Miyata, Ryouhei; Fujikawa, Ryu; Muroi, Yoshikage.
The mesencephalic trigeminal sensory nucleus (Me5), which receives signals originating from oral proprioceptors, becomes active at weaning and contributes to the acquisition of active exploratory behavior [Ishii, T., Furuoka, H., Kitamura, N., Muroi, Y., and Nishimura, M. (2006) Brain Res. 1111, 153-161]. Because cognitive functions play a key role in animal exploration, in the present study we assessed the role of Me5 in spatial learning and memory in the water maze. Mice with bilateral Me5 lesions exhibited severe deficits in both a reversal learning and a reversal probe test compared with sham-operated mice. In spite of these reversal tests, Me5 lesions had no effect on a hidden platform test. These results suggest that Me5-lesioned mice show a...

A novel neuropeptide Y neuronal pathway linking energy state and reproductive behavior OAK
Muroi, Yoshikage; Ishii, Toshiaki.
Animals consume energy for reproduction, as well as survival. Excess or insufficient energy investment into reproduction, respectively, threatens the survival of parents or leads to the failure of reproduction. Management of energy consumption in reproduction is important, not only for the success of the process, but also for the survival of the parents. Reproductive behaviors, such as mating and parental behavior, are indispensable for achieving each event of reproduction including gametogamy, parturition, and lactation. Therefore, reproductive behavior is one of the important factors in managing energy consumption for reproduction. Orexigenic and anorexigenic molecules in the hypothalamus have been implicated in the regulation of reproductive functions....
