Proteomic and endophytic changes during storage of organically and conventionally grown carrots Organic Eprints
LOUARN, Sébastien; NAWROCKI, Arkadiusz; Thorup-Kristensen, Kristian; Søgaard Lund, Ole; NØRREGAARD JENSEN, Ole; Collinge, David B.; Jensen, Birgit.
This study utilises physiologically independent assays to evaluate whether there are differences in the way that organically and conventionally grown carrots develop during storage. Thus quantitative proteomics and sampling the endophytic micromycota of stored carrots provide two different assessments of the physiological state of stored carrots.

Selection for resistance against root pathogens in a pea composite cross Organic Eprints
Knudsen, Jens; Larsen, John; Bødker, Lars; Jørnsgård, Bjarne; Jensen, Birgit.
The possibility of improving resistance in pea against the root pathogen Aphanomyces euteiches using composite cross as a breeding and selection method was examined. In order to maintain acceptable agricultural features and high yield 6 out of the 8 parental varieties in the present composite-cross were commercially grown varieties. Populations of the composite cross were grown up to five generations with selection pressure in soil heavily infested with pea root pathogens or without selection pressure on soil free of pea root pathogens. Yield of populations of the F9 and F10 generations of the composite cross grown with selection pressure was on average 35% higher than that of the population obtained without selection pressure as well as the average yield...

Specificity of soil-borne pathogens on grain legumes Organic Eprints
Jensen, Birgit; Bødker, Lars; Larsen, John; Knudsen, Jens Chr.; Jørnsgaard, Bjarne.
Specificity of soil-borne legume pathogens on pea, lupin and faba bean is currently investigated in fields where grain legumes are intensively cultivated. The study has so far lead to the following conclusions -Legume host-pathogen interactions demonstrate specificity of pathogen populations particularly in pea and lupin. -A. euteiches rot root was specified to pea in Denmark as root rot symptoms and oospores of the pathogen never were observed in roots of faba bean and lupin -F. oxysporum followed by F. solani were most frequently isolated from plant roots in plots highly infested by lupin pathogens -F. avenaceum was most frequently isolated from plant roots in plots highly infested by pea pathogens -Pathogenicity tests showed F. solani followed...

Jensen, Birgit; Jørnsgaard, Bjarne; Knudsen, Jens Chr..

Functional Compost Organic Eprints
Luxhøi, Jesper; Brøgger, Morten; Knudsen, Inge M.B.; Poulsen, Pernille H.B.; Møller, Jacob; Jensen, Birgit; Jensen, Dan F.; Magid, Jakob.
The aim of the research program Functional Compost is to develop and test compost, which have been enriched with chitin, for plant growth promoting properties and to recognise specific mechanisms. Two types of compost were included in the program: source separated biodegradable municipal solid waste compost (DM = 62 %) and garden and park waste compost (DM = 66 %). Chitin was added in trace amounts during the maturity phase, combined with two levels of trace amounts immediately before adding the compost to the growth medium. The research program includes several parallel experiments. In experiment I, compost (20 vol. %) was added to soil (no plants) and incubated at 15 C for 5 month, under regular determination of microbial respiration and gross and net N...

Gulerodens farefulde vej fra marken til forbrugeren Organic Eprints
Knudsen, Inge M.B.; Jensen, Dan Funck; Jensen, Birgit.

Resistance against Fusarium spp. in narrowed leafed lupin (Lupinus angustifolius L.) Organic Eprints
Jørnsgård, Bjarne; Kuptsov, Vladislav; Jensen, Birgit; Bødker, Lars; Knudsen, Jens Christian; Nielsen, John.
L. angustifolius genotypes susceptible to Fusarium root rot and wilt can be grown in new lupin land without disease symptoms, however, lupin root rot and wilt pathogens build up during repeated cropping of susceptible lupins in the same field. After four lupin crops symptoms of root rot and wilt began in an inhomogeneous pattern over the field, and after another three lupin crops susceptible lupins are destroyed before flowering homogeneously over the field and caused total crop failure. The dominating pathogens isolated from diseased lupins with root rot and wilt symptoms in this field are in descending order F. oxisporum, F. solani, F. culmorum, and F. avenaceum, the latter only being isolated from young autumn sown plants. A core collection of lupin...

The influence of the fungal pathogen Mycocentrospora acerina on the proteome and polyacetylenes and 6- methoxymellein in organic and conventionally cultivated carrots (Daucus carota) during post harvest storage Organic Eprints
LOUARN, Sébastien; NAWROCKI, Arkadiusz; Edelenbos, Merete; JENSEN, Dan Funck; NØRREGAARD JENSEN, Ole; Collinge, David B.; Jensen, Birgit.
Carrots may have to be discarded during post harvest cold storage due to development of fungal infections, caused by, e.g., liquorice rot caused by Mycocentrospora acerina. Control of this disease is mainly based on cultural practice. We compared the susceptibility of carrots grown under conventional and organic agricultural practices. We have developed a bioassay for infection studies of M. acerina on carrots and observed that organic roots were more susceptible after one month storage than conventional ones but no differences were apparent after four or six months storage. Levels of three polyacetylenes (falcarinol, falcarindiol and falcarindiol-3-acetate) and the isocoumarin phytoalexin (6-methoxymellein) were analysed. No changes in individual and...

Quantification of root fungi using signature fatty acids Organic Eprints
Ravnskov, Sabine; Jensen, Birgit; Nielsen, Steen Lykke; Larsen, John.
Both deleterious (pathogenic) and beneficial (mycorrhizal) fungi inhabit plant roots with strong impact on plant growth and health. Various methods have been used to quantify these fungi, such as indirect measurements of plant parameters, disease index, staining techniques and serological/genetic/biochemical markers. The objective of this work is to evaluate the possibility of using signature fatty acids to quantify root-inhabiting fungi in planta and in soil. Different fatty acid-based methods can be used to quantify fungi. Phospholid fatty acids (PLFA) can be used for biomass estimation and neutral lipid fatty acids (NLFA) for estimation of fungal energy reserves and the NLFA/PLFA ratio can give information on the physiological status of the fungus. It...

Proteomic analysis of the Mycocentrospora acerina-carrot interaction during storage Organic Eprints
LOUARN, Sébastien; Jensen, Birgit; NAWROCKI, Arkadiusz; NØRREGAARD JENSEN, Ole.
During post harvest storage, a large proportion of carrots (more than 50%) may have to be discarded due to the development of liquorice rot caused by Mycocentrospora acerina. This fungus is soil borne and brought into the store in to soil adhering to the root. Liquorice rot development is mainly related to physiological or structural resistance of carrot, therefore the control of this storage disease is based on cultural practices and storage conditions. It is believed that carrots at the beginning of storage can resist disease developments due to chemical defence mechanisms involving some proteins, peptides and secondary metabolites. The hypothesis is that proteome changes during storage of carrots are related to the susceptibility to M. acerina. During...
