Induction of in vitro flowering in Capsicum frutescens under the influence of silver nitrate and cobalt chloride and pollen transformation Electron. J. Biotechnol.
Sharma,Ashwani; Kumar,Vinod; Giridhar,Parvatam; Ravishankar,Gokare Aswathanarayana.
The influence of silver nitrate (AgNO3) and cobalt chloride (CoCl2) on shoot multiplication and in vitro flowering in Capsicum frutescens Mill. was investigated. Exogenous administration of AgNO3 and CoCl2 at a concentration of 30 µM resulted in the maximum tissue response in terms of shoot length and number of shoots after 45 days culturing on MS medium. Both silver nitrate (40 µM) and cobalt chloride (30 µM) influenced in vitro flowering after 25 and 45 days respectively. This is the first report on in vitro flowering in C. frutescens. The study also demonstrated successful transformation of pollen obtained from the in vitro flowers. Since capsicum is highly recalcitrant to in vitro plant regeneration, the results of the study may be highly useful in...

Agrobacterium rhizogenes mediated genetic transformation resulting in hairy root formation is enhanced by ultrasonication and acetosyringone treatment Electron. J. Biotechnol.
Kumar,Vinod; Sharma,Ashwani; Narasimha Prasad,Bellur Chayapathy; Bhaskar Gururaj,Harishchandra; Aswathanarayana Ravishankar,Gokare.
The phytopathogenic bacteria Agrobacterium rhizogenes genetically transforms plants by transferring a portion of the resident Ri plasmid, the T-DNA to the plant. Plant species differ in their temporal competence for transformation. But various physical and chemical methods are found to enhance transformation frequency. Agrobacterium rhizogenes mediated transformation efficiency was assessed under the influence of sonication, calcium treatment, acetosyringone and macerating enzymes in suitable combinations in Nicotiana tabacum as a model system. Manual wounding resulted in 21% transformation frequency. Where as sonication resulted in 2.2 fold increase, followed by sonication with CaCl2 treatment resulted in 2.5 fold increase and sonication with...

Rudrappa,Thimmaraju; Neelwarne,Bhagyalakshmi; Kumar,Vinod; Lakshmanan,Venkatachalam; Venkataramareddy,Sreedhar Reddampalli; Aswathanarayana,Ravishankar Gokare.
The genetically transformed roots of red beet have been shown, for the first time, to produce very high levels of peroxidase (POD; EC 1.11.1.7) accounting for 1.21 x 10(6) Units L-1. Of the ten clones established using different strains of Agrobacterium rhizogenes, one was that from the strain LMG-150, three each from A 2/83, A 20/83 and A4. All the clones showed true integration of T-DNA when tested by PCR and Southern hybridization methods. Each clone differed significantly from the others in growth, hormone dependency and POD production where LMG-150 produced highest biomass (140 g FW L-1) as well as POD (ranging from 8000-9000 U g-1 FW and 1.18 x 10(6) U L-1 with a specific activity of 600 U mg-1 protein) on hormone-free medium, both in shake-flask as...

AgNO3: a potential regulator of ethylene activity and plant growth modulator Electron. J. Biotechnol.
Kumar,Vinod; Parvatam,Giridhar; Ravishankar,Gokare Aswathanarayana.
The aim of this review is to critically analyze the role of silver nitrate (AgNO3) in modulating plant growth and development. In recent years, basic studies on ethylene regulation opened new vistas for applied research in the area of micro-propagation, somatic embryogenesis, in vitro flowering, growth promotion, fruit ripening, and sex expression. Silver nitrate has proved to be a very potent inhibitor of ethylene action and is widely used in plant tissue culture. Few properties of silver nitrate such as easy availability, solubility in water, specificity and stability make it very useful for various applications in exploiting plant growth regulation and morphogenesis in vivo and in vitro. Silver ion mediated responses seem to be involved in polyamines,...

Isolation and characterization of hydrocarbon producing green alga Botryococcus braunii from Indian freshwater bodies Electron. J. Biotechnol.
Dayananda,Chandrappa; Sarada,Ravi; Kumar,Vinod; Ravishankar,Gokare Aswathanarayana.
Samples of green colonial unicellular microalga Botryococcus braunii were collected from Bear Shola Falls at Kodaikanal (latitude 10.31 N and longitude 77.32 E), Tamil Nadu, India. Specimens were isolated, cultured and examined for its hydrocarbon content, morphological features and DNA structural resemblance with the known strain to confirm its identity. Inter simple sequence repeats (ISSR) finger printing revealed strong genetic similarity among the authentic strain (B. braunii N-836) and the Indian isolated strain (B. braunii CFTRI- Bb1) from Kodaikanal. The type of hydrocarbons produced by the Kodaikanal isolates were analyzed and identified as saturated hydrocarbons in the range of C21 to C33 by GCMS. Tetracosane and octacosane were found as the major...

Recent developments in microbial oils production: a possible alternative to vegetable oils for biodiesel without competition with human food? BABT
Christophe,Gwendoline; Kumar,Vinod; Nouaille,Régis; Gaudet,Geneviève; Fontanille,Pierre; Pandey,Ashok; Soccol,Carlos Ricardo; Larroche,Christian.
Since centuries vegetable oils are consumed as human food but it also finds applications in biodiesel production which is attracting more attention. But due to being in competition with food it could not be sustainable and leads the need to search for alternative. Nowdays microbes-derived oils (single cell oils) seem to be alternatives for biodiesel production due to their similar composition to that of vegetable oils. However, the cold flow properties of the biodiesel produced from microbial oils are unacceptable and have to be modified by an efficient transesterification. Glycerol which is by product of transesterification can be valorised into some more useful products so that it can also be utilised along with biodiesel to simplify the downstream...

Leishmania donovani infection activates Toll-like receptor 2, 4 expressions and Transforming growth factor-beta mediated apoptosis in renal tissues BJID
Kumar,Vinod; Tiwari,Neeraj; Gedda,Mallikarjuna Rao; Haque,Rizwanul; Singh,Rakesh K..
Abstract The present study was aimed to identify the underlying mechanisms of improper renal function in Leishmania donovani infection that causes VL. Mice (BALB/c) were infected with L. donovani and different parameters for proteinuria were assessed. The levels of superoxide anion (O2 -), hydrogen peroxide (H2O2), lipid peroxidation (MDA), inflammatory cytokines, and toll-like receptor (TLR) 2 and 4 expression were found significantly elevated at 60th day in these animals and declined at 90th day post infection. However, TGF-β and caspase 3 activities were higher at 90th day in comparison to 60th day post infection. These findings suggested that exacerbated inflammatory conditions correlate with abnormal renal functions in L. donovani infection, which is...

Molecular characterisation of Aspergillus flavus isolates from peanut fields in India using AFLP BJM
Singh,Diwakar; Radhakrishnan,T.; Kumar,Vinod; Bagwan,N.B.; Basu,M.S.; Dobaria,J.R.; Mishra,Gyan P.; Chanda,S.V..
Aflatoxin contamination of peanut, due to infection by Aspergillus flavus, is a major problem of rain-fed agriculture in India. In the present study, molecular characterisation of 187 Aspergillus flavus isolates, which were sampled from the peanut fields of Gujarat state in India, was performed using AFLP markers. On a pooled cluster analysis, the markers could successfully discriminate among the ‘A’, ‘B’ and ‘G’ group A. flavus isolates. PCoA analysis also showed equivalent results to the cluster analysis. Most of the isolates from one district could be clustered together, which indicated genetic similarity among the isolates. Further, a lot of genetic variability was observed within a district and within a group. The results of AMOVA test revealed that...
